We will cover some basic facts, protein expression, and Cathepsin B’s expected behavior in Western blot. All information in this video is based on public information and no proprietary experimental evidence was used.
Topic 1: Quick facts about CTSB. Cathepsin B belongs to the papain-like family of cysteine proteases. It is a bilobal protein whose catalytic site is located at the interface between the two lobes. CTSB exhibits both exopeptidase and endopeptidase activity. The precursor of CTSB is 339 amino acids long and runs about 35 kilodaltons on western blot while the mature form is 254 amino acids long and runs about 25 kilodaltons on Western blot.
Topic 2: Where is Cathepsin B expressed? High levels of CTSB have been found in the brain, lungs, GI tract, pancreas, male and female tissues, kidney, and bladder. They are also expressed at lower levels in the heart, bone marrow, and lymphoid tissues. You can find more information on protein atlas dot org to check out which cell lines express CTSB and which ones don’t. This information will come in handy when designing positive and negative controls for western blot and immunohistochemistry.
Topic 3: Cathepsin B in Western blotting. Antibody specificity can make or break an experiment. After looking through Western blot images from publications and antibody companies, we can see that typically two bands can be expected in a Cathepsin B Western blot, the pro-form CTSB at around 35 kilodaltons and the mature CTSB at around 25 kilodaltons. Topic 4: Some interesting facts about Cathepsin B. In 1986, subcellular fractionation of melanoma cells revealed the presence of single chain form of cathepsin B on the plasma membrane. Cathepsin B is involved in various oncogenic processes and pathologies in humans, making it of great importance in cancer therapy. The correlation of overexpression of cathepsin B with metastatic phenotypes in cancer has been shown in various studies. Abnormal regulation of CTSB causes the cell to acquire an oncogenic character. Suppressing the expression of CTSB slows down the infiltrative behaviour of tumor cells. Therefore, targeting the activity or expression of cathepsin B is of significant therapeutic importance for cancer treatment.
