We will cover some basic facts, protein expression, and BMP2’s expected behavior in Western blot. All information in this video is based on public information and no proprietary experimental evidence was used.
Topic 1: Quick facts about BMP2. Bone morphogenetic protein 2 or BMP2 is an important growth factor involved in the development of the heart, brain, and bones. BMP2 is 396 amino acids long and runs about 45 kilodaltons on a western blot.
Topic 2: Where is BMP2 expressed? BMP2 RNA is expressed in multiple tissues, such as the bone marrow, kidney and urinary bladder, endocrine tissues, respiratory system, brain, gastrointestinal tract, and other tissues. You can find more information on protein atlas dot org to check out which cell lines express BMP2 and which ones don’t. This information will come in handy when designing positive and negative controls for western blot and immunohistochemistry.
Topic 3: BMP2 in Western Blotting. Antibody specificity can make or break an experiment. After looking through Western blot images from publications and antibody companies, we can see that typically one band can be expected in a BMP2 western blot at about 45 kilodaltons.
