We will cover some basic facts, protein expression, and CXCL13’s expected behavior in Western blot. All information in this video is based on public information and no proprietary experimental evidence was used.
Topic 1: Quick facts about CXCL13. CXCL13, also known as BLC or BCA1, is a chemotactic cytokine for B lymphocytes that binds to BLR1 or CXCR5. It stimulates calcium influx into, and chemotaxis of, cells transfected with Burkitt's lymphoma receptor 1. The mature form of CXCL13 is 109 amino acids long and runs about 12.6 kilodaltons on Western blot.
Topic 2: Where is CXCL13 expressed? High levels of CXCL13 RNA expression have been found in the tonsils and spleen, with the highest levels detected in the lymph node. They are also expressed at lower levels in the appendix, small intestine, stomach, thymus, prostate, urinary bladder, and liver. You can find more information about tissue expression on protein atlas dot org as well as which cell lines express CXCL13. This information will come in handy when designing positive and negative controls for western blot and immunohistochemistry.
Topic 3: CXCL13 in Western blotting. Antibody specificity can make or break an experiment. After looking through Western blot images from publications and antibody companies, we can see that typically one band can be expected in a CXCL13 Western blot with the mature form of CXCL13 between 12 and 15 kilodaltons. Topic 4: Some interesting facts about CXCL13. Overall, the CXCL13/CXCR5 signaling axis can recruit immune cells and enhance the capacity to defend the body. It was initially detected in stromal cells of B cell follicles and associated with the recruitment of B cells and T cells subsets in 1996. Therefore, the scientific community is interested in the use of CXCL13 as a prognostic and predictive factor in some ICI responsive cancer tumor types.
