  2. We will cover some basic facts, protein expression, and Tau’s expected behavior in Western blot. All information in this video is based on public information and no proprietary experimental evidence was used.
  3. Topic 1: Quick facts about tau. Microtubule-associated protein tau is a highly soluble protein. Although Uniprot reports 9 isoforms produced by alternative splicing, 6 isoforms with a range of 352 to 441 amino acids are more commonly discussed. Isoform PNS-tau, a larger isoform of tau also known as Big tau, is 758 amino acids long. In general, tau proteins run about 50 kilodaltons to 80 kilodaltons on Western blot. Hyperphosphorylated tau is observed in Alzheimer’s disease, accompanying the tangles that progressively form in the brain. Tau is one of the most researched proteins of interest in Alzheimer’s disease and other dementias.
  4. Topic 2: Where is tau expressed? High levels of tau have been found in neurons. Isoform PNS-tau is primarily expressed in the peripheral nervous system while other isoforms are expressed in the central nervous system. According to Protein Atlas, tau is also expressed in breast tissue and the kidney, more specifically the renal tubules. Lower levels of expression are found in the salivary gland, testes, pancreas, adipose tissue, etc. For more information, visit protein atlas dot org and check out which cell lines express tau.   5. Topic 3: Tau in Western blotting. Antibody specificity can make or break an experiment. The last thing we want is to have our papers retracted due to a false positive figure. After looking through Western blot images from publications and antibody companies, we can see that several bands can be expected in a tau western blot, spanning from 50 kilodaltons to 80 kilodaltons.   6. Topic 4: Some interesting facts about tau. Marc Kirschner identified the first microtubule-stabilizing protein tau in 1975 at Princeton. Tau is observed to be present in several forms of dementia and neurodegenerative disorders. In healthy individuals, Tau is a highly soluble protein. However, in Alzheimer’s disease, hyperphosphorylated Tau is associated with insoluble neurofibrillary tangles that build up in the brain. Because of this, Tau’s involvement in the mechanism of action of dementia is of high interest to pathologists and some pharmaceutical companies.
