We will cover some basic facts, protein expression, and ESM1’s expected behavior in Western blot. All information in this video is based on public information and no proprietary experimental evidence was used.
Topic 1: Quick facts about Endocan. Endothelial cell-specific molecule 1 or ESM1 is a soluble proteoglycan, which is expressed by the vascular endothelium. It is a key player in the regulation of major processes such as cell adhesion, inflammatory disorders, and tumor progression. The total chain length is 184 amino acids, out of which the signal peptide of 19 amino acids is cleaved to form the active protein of 165 amino acids. The active protein runs about 25 kilodaltons on Western blot.
Topic 2: Where is endocan expressed? High levels of ESM1 RNA have been found in the thyroid gland, lung, parathyroid gland, and adrenal gland with the highest levels detected in the kidney. They are also expressed at lower levels in the spleen, bone marrow, amygdala, cerebellum, and thymus. You can find more information at protein atlas dot org including which cell lines express endocan and which ones don’t. This information will come in handy when designing positive and negative controls for western blot and immunohistochemistry.
Topic 3: Endocan in Western blotting. Antibody specificity can make or break an experiment. After looking through Western blot images from publications and antibody companies, we can see that one band can be expected in an ESM1 Western blot at around 20 to 25 kilodaltons. Topic 4: Some interesting facts about Endocan. ESM1 is an important protein involved with angiogenesis, inflammation, and tumor progression. Research has shown that it is overexpressed in human tumors, with elevated serum levels being observed in late-stage lung cancer patients. Since the overexpression of endocan aids in tumor progression, ESM1 could be a biomarker for both inflammatory disorders and tumor progression, potentially serving as a therapeutic target in cancer.
