The development and manufacture of biopharmaceuticals is a dynamic and rapidly growing industry. By the nature of their production, biopharmaceuticals are highly complex heterogeneous mixtures that require many analytical techniques for characterization and routine testing. Simplifying Chromatographic Methods Transfer: Novel Tools for Replicating You...Waters Corporation
Gain a good understanding on the parameters that impact the successful transfer of an LC method from one instrument to another as well as some of the novel tools (i.e., Arc Multi-flow path technology and gradient SmartStart) that have been created to enable the ACQUITY Arc System to replicate established HPLC methods from previous generations of LC equipment.
Transfer of established gradient reversed-phase methods across both HPLC and UHPLC chromatographic instrumentation requires careful consideration of each instrument’s operating parameters and design. The dwell volume, or the system volume between when the solvents are first mixed and the head of the column, can impact the separation. In addition to the dwell volume, the manner in which the solvents are mixed and the formation of the gradient can also vary from one type of LC system to another. Finally, the manner in which the column and solvent are heated can also effect the separation, specifically whether or not the solvent is passively or actively heated prior to the column, or if the air is static or circulated within the column oven.
To understand the effect of these factors may have on methods transfer, both method conditions and instrument specifications must be factored and evaluated when transferring an LC method from one instrument to another.

Interested in what your customers (or their customers) are talking about, and how that’s changing over time? The most popular 'topic trending' analyses depend on the investigation of how keyword (or term) usage change over time. Using keyword trends to index topic trends is highly suitable for short-form text, such as search terms, hashtags, or tweets. However, exploring trends in topic prevalence across longer, more free-form texts, such as call center telephone transcripts, is better served by grouping together topically-related words that co-occur. In this talk, we'll show an example of using Latent Dirichlet Allocation (LDA; Blei, Ng, & Jordan, 2003) in R to group words in call center transcripts into multiword topics over which we explore trends. We’ll also demonstrate the how the use of Structural Topic Modeling (Roberts, Stewart, Tingley, & Airoldi, 2013) can aid in further investigation of how document-level covariates (in this case, additional call- or caller-based characteristics) can affect topic prevalence and topic trends.
This presentation describes development of a routine tandem quadrupole LC/MS/MS method for milk and egg allergens based on proteomic studies to identify the allergenic peptide markers. Initial studies are done using a proteomic workflow and quadrupole time of flight mass spectrometry.
Comprehensive Investigation of the Utilization of SFC/ESI Positive Mode MS fo...Waters Corporation
Bioanalysis and drug metabolism studies are critical parts of the drug development process. The aim of these studies is to identify and quantify drugs and their associated metabolites in biofluids such as plasma and urine. Typically reversed-phase chromatography coupled with mass spectrometry is often the analytical technique of choice utilized in the analyses due to the specificity and sensitivity of the technique. However, due to the complexity of the biofluid samples accurate and precise measurements can become challenging due to poor chromatographic peak shape, insufficient chromatographic resolution from matrix components and incompatible sample compositions.
Recent advancements in the field of super critical fluid chromatography (SFC) have lead to the development of sub 2 micron chromatographic separations coupled mass spectrometry operating under positive ESI mode. In the work presented here the applicability of SFC for both chiral and achiral bioanalysis is shown. The orthogonal separation selectivity compared with reversed-phase separations and tolerance for high organic sample compositions will be discussed. This work further presents a critical evaluation of the influence of mobile phase and make up flow modifiers on the sensitivity and selectivity of probe pharmaceuticals analyzed under SFC/ESI positive mode MS conditions.

Environmental analysis can be extremely challenging due to the low detection levels for toxic contaminants specified by legislation, particularly in drinking water, and the complexity of matrices encountered. Consequently highly selective and sensitive detection methods are required. This presentation provides an introduction to tandem quadrupole mass spectrometry and describes the use of high sensitivity tandem quadrupole mass spectrometry for the analysis of various environmental contaminants including pesticides, endocrine disruptors and polyfluorinated compounds such as PFOS.
Allergens are a major food safety concern and incidences of food allergy in industrialised populations has increased in recent times. One of the most common food allergies is that of peanuts. Food regulations for allergens exist in many countries and are being modified regularly as more is understood about allergens and the reactions they cause. This presentation describes the use of time-of-flight mass spectrometry to locate, identify and quantify an allergenic protein in both raw and roasted peanuts. Typical food processing (e.g. food processing) can alter the markers peptides present and amount that they are present in the samples which adds complexity to the analysis.

This presentation describes the operation and application of the Waters APGC (Atmospheric Pressure Gas Chromatography) ion source which provides a highly sensitive GC-MS, MS/MS capability for tandem quadrupole and time of flight MS systems. It is very easy to swap between APGC, Electrospray (for UPLC) and other ion sources without instrument venting in minutes.
APGC provides significant performance advantages over traditional GC/MS ionisation methods, giving high sensitivity and less fragmented spectra.
Method Development, Validation and Forced Degradation Studies of Dapagliflozi...ijtsrd
A simple, sensitive, robust, precise, and efficient RP HPLC approach for the simultaneous determination of Dapagliflozin and Pioglitazone Hydrochloride in Synthetic Mixture. As per ICH Q2 R1 guidelines, the final chromatographic conditions were Optimized with a mobile phase ratio of 25 75 v v in ACN Potassium Dihydrogen Phosphate Buffer pH 4 was adjusted by adding OPA at a flow rate of 1 mL min, column temperature of 30 °C, injection volume of 20 µL, Kromstar Vertex C18 analytical column, and UV detection at 228 nm wavelength. Dapagliflozin and Pioglitazone Hydrochloride reported retention times of 3 min and 6.5 min, respectively. Validation of a method was found to be linear in the range of 2 10 µg ml for Dapagliflozin and 3–15 µg mL for Pioglitazone Hydrochloride. The Recovery for Dapagliflozin was discovered to be 98.52 99.90 , while for Pioglitazone Hydrochloride, it was found to be 99.67 99.94 . The Precision results for both drugs were within the limits while expressed Intraday and Interday. For Dapagliflozin, the LOD and LOQ were reported to be 0.041 µg mL and 0.13 µg mL, respectively, and for Pioglitazone Hydrochloride, 0.105 µg mL and 0.32 µg mL. As per ICH Q1A R2 guidelines, the synthetic mixture was subjected to acid, base, oxidation, thermal, and photolysis stress conditions. Since volcanic activity was first discovered on Io from Voyager images in 1979, changes
on Io’s surface have been monitored from both spacecraft and ground-based telescopes.
Here, we present the highest spatial resolution images of Io ever obtained from a groundbased telescope. These images, acquired by the SHARK-VIS instrument on the Large
Binocular Telescope, show evidence of a major resurfacing event on Io’s trailing hemisphere. When compared to the most recent spacecraft images, the SHARK-VIS images
show that a plume deposit from a powerful eruption at Pillan Patera has covered part
of the long-lived Pele plume deposit. Although this type of resurfacing event may be common on Io, few have been detected due to the rarity of spacecraft visits and the previously low spatial resolution available from Earth-based telescopes. The SHARK-VIS instrument ushers in a new era of high resolution imaging of Io’s surface using adaptive
optics at visible wavelengths.

In this book, we use conservation of energy techniques on a fluid element to derive the Modified Bernoulli equation of flow with viscous or friction effects. We derive the general equation of flow/ velocity and then from this we derive the Pouiselle flow equation, the transition flow equation and the turbulent flow equation. In the situations where there are no viscous effects , the equation reduces to the Bernoulli equation. From experimental results, we are able to include other terms in the Bernoulli equation. We also look at cases where pressure gradients exist. We use the Modified Bernoulli equation to derive equations of flow rate for pipes of different cross sectional areas connected together. We also extend our techniques of energy conservation to a sphere falling in a viscous medium under the effect of gravity. We demonstrate Stokes equation of terminal velocity and turbulent flow equation. We look at a way of calculating the time taken for a body to fall in a viscous medium. We also look at the general equation of terminal velocity.
