Dysfunction
Generic Tadalafil is a prescription medication which is FDA-approved to
treat the following in men: erectile dysfunction (ED), a condition in which
you can’t get or keep an erection, symptoms of benign prostatic hyperplasia
(BPH), a prostate condition that may cause problems with urination and
ED and symptoms of BPH together.
Tadalafil's effect starts working in 30 minutes and lasts for upto 36 hours.
Tadalafil can also be used daily by using the 5 mg dose, so you can be ready
anytime. It’s available in three strengths: 5mg, 10 mg and 20 mg. It is also
available as an Oral jelly in the strength of 20 mg.
Tadalafil tablets are also indicated to improve the ability to exercise in
adults with Pulmonary Arterial Hypertension (PAH).

Uses of Generic Tadalafil 5mg, 10mg and 20mg:
Generic Tadalafil (tablets and oral jelly) is a treatment for adult men
suffering from Erectile Dysfunction (ED). This is a medical condition when a
man cannot get, or keep a hard, erect penis suitable for sexual activity.
When a man is sexually stimulated, his body's normal response is to increase
blood flow to his penis to produce an erection. By controlling the enzyme,
Tadalafil helps to maintain an erection after the penis is stroked by increasing
blood flow to the penis. Without physical action to the penis, such as that
occurring during sexual intercourse, Tadalafil will not work to cause an
erection.
Generic Tadalafil has been observed to significantly improve the ability of
obtaining a hard erect penis suitable for sexual activity.

Dysfunction
Urinary symptoms associated with a common condition called
Benign Prostatic Hyperplasia (BPH). This is when the prostate
gland gets bigger with age. Symptoms include difficulty in starting
to pass water, a feeling of not completely emptying the bladder
and a more frequent need to pass water even at night.
Generic Tadalafil improves blood flow to, and relaxes the muscles
of, the prostate and bladder which may reduce symptoms of benign
prostatic hyperplasia.
Generic Tadalafil has been shown to improve these urinary
symptoms as early as 1 to 2 weeks after starting treatment.

Dysfunction
This medicine is also used to treat ED and the signs and symptoms
of BPH (ED/BPH) at the same time.
Tadalafil tablets are also used for the treatment of pulmonary
arterial hypertension (PAH) (WHO Group 1) to improve exercise
ability.
Studies establishing effectiveness included predominately patients
with NYHA Functional Class II – III symptoms and etiologies of
idiopathic or heritable PAH (61%) or PAH associated with
connective tissue diseases (23%).

Dysfunction
Recommended Adult Dosage
For most patients, the dose of Generic Tadalafil to treat
Erectile Dysfunction (ED) for use as needed is 10 mg as
needed prior to sexual activity. Increase to 20 mg or
decrease to 5 mg based upon efficacy/tolerability.
This medicine improves erectile function compared to
placebo up to 36 hours post dose. Not to be taken more
than once per day. i.e. 24 hours must pass before you take
your next dose.

Dysfunction
Generic Tadalafil to treat Erectile Dysfunction (ED)
for once daily use: 2.5mg taken once daily, without
regard to timing of sexual activity. May increase to 5 mg
based upon efficacy and tolerability.
Benign Prostatic Hyperplasia (BPH): 5 mg, taken at
approximately the same time every day.
ED and BPH: 5 mg, taken at approximately the same
time every day.

Dysfunction
Pulmonary Arterial Hypertension: The
recommended dose of Tadalafil tablets is 40 mg (two
20 mg tablets) taken once daily with or without
food.
Dividing the dose (40 mg) over the course of the day
is not recommended.

Generic Tadalafil Contraindications

Generic Tadalafil is contraindicated in patients with a
known hypersensitivity to Tadalafil or any component of
the tablet.
➔
Administration of Generic Tadalafil tablets to patients
using any form of organic nitrate is contraindicated.
Tadalafil was shown to potentiate the hypotensive effect
of nitrates.

Storage Instructions
Generic Tadalafil tablets have to be stored at
controlled room temperature i.e. from 20°C to
25°C (68°F to 77°F) with excursions permitted
between 15° to 30°C (59° to 86°F).
Keep this as well as all other medicines away
from children and pets.

Warnings
Patients should not use Generic Tadalafil Tablets if sex is NOT
advisable due to cardiovascular status.
Use of Generic Tadalafil with alpha blockers, antihypertensives or
substantial amounts of alcohol (5 units or greater) may lead to
hypotension.
Generic Tadalafil is not recommended in combination with alpha
blockers for the treatment of BPH because efficacy of the combination
has not been adequately studied and because of the risk of blood
pressure lowering. Caution is advised when Generic Tadalafil is used as
a treatment for ED in men taking alpha blockers.

Warnings
If taking potent inhibitors of CYP3A4, dose should be adjusted:
Generic Tadalafil for use as needed: Less than 10 mg every 72 hours.
For once daily use: dose not to exceed 2.5 mg.
Patients should seek emergency treatment if an erection lasts for
more than 4 hours. Use Generic Tadalafil with caution in patients
predisposed to priapism.
Patients should stop Generic Tadalafil and seek medical care if a
sudden loss of vision occurs in one or both eyes, which could be a
sign of Non Arteritic Ischemic Optic Neuropathy (NAION). Discuss
increased risk of NAION in patients with history of NAION.

Warnings
Patients should stop Generic Tadalafil tablets and seek
prompt medical attention in the event of sudden
decrease or loss of hearing.
Before starting treatment with Generic Tadalafil for
BPH, consideration should be given to other
urological conditions that may cause similar
symptoms.

Possible Side Effects
Most common Tadalafil tablets side effects with an
incidence rate of greater than or equal to 2% and more
frequent on drug than placebo observed during clinical
trials were Headache, Dyspepsia, Nasopharyngitis, Back
pain, Upper respiratory tract infection, Flushing,
Myalgia, Cough, Diarrhea, Nasal congestion, Pain in
extremity, Urinary tract infection, Gastroesophageal
reflux disease and Abdominal pain.

Dysfunction
Additional, less frequent side effects (Less than 2%) reported in controlled
clinical trials of Tadalafil tablets for once daily use or use as needed. A causal
relationship of these events to Tadalafil is NOT certain.
Body as a Whole — asthenia, face edema, fatigue, pain
Cardiovascular — angina pectoris, chest pain, hypotension, myocardial
infarction, postural hypotension, palpitations, syncope, tachycardia
Digestive — abnormal liver function tests, dry mouth, dysphagia, esophagitis,
gastritis, GGTP increased, loose stools, nausea, upper abdominal pain,
vomiting, gastroesophageal reflux disease, hemorrhoidal hemorrhage, rectal
hemorrhage
Musculoskeletal — arthralgia, neck pain Nervous — dizziness, hypesthesia,
insomnia, paresthesia, somnolence, vertigo

Dysfunction
Additional, less frequent side effects (Less than 2%) reported in controlled
clinical trials of Tadalafil tablets for once daily use or use as needed. A causal
relationship of these events to Tadalafil is NOT certain.
Renal and Urinary — renal impairment
Respiratory — dyspnea, epistaxis, pharyngitis
Skin and Appendages — pruritus, rash, sweating
Ophthalmologic — blurred vision, changes in color vision, conjunctivitis
(including conjunctival hyperemia), eye pain, lacrimation increase, swelling of
eyelids
Otologic — sudden decrease or loss of hearing, tinnitus
Urogenital — erection increased, spontaneous penile erection

Dysfunction
Generic Tadalafil Overdosage
Single doses up to 500 mg of Tadalafil tablets have
been given to healthy subjects, and multiple daily doses
up to 100 mg have been given to patients.
Adverse events were similar to those seen at lower
doses. In cases of overdose, standard supportive
measures should be adopted as required. Hemodialysis
contributes negligibly to Tadalafil elimination.

Dysfunction
Generic Tadalafil Tablets During Pregnancy
Tadalafil tablets has been classified by the US FDA as Pregnancy
Category B. Tadalafil is not indicated for use in women. There are no
adequate and well controlled studies of Tadalafil tablets use in pregnant
women.
Animal reproduction studies in rats and mice revealed no evidence of
fetal harm.
Animal reproduction studies showed no evidence of teratogenicity,
embryotoxicity, or fetotoxicity when Tadalafil was given to pregnant rats
or mice at exposures up to 11 times the maximum recommended human
dose (MRHD) of 20 mg/day during organogenesis.
