Plant tissue culture is a technique of growing plant cells, tissues, organs, seeds, or other plant parts in a sterile environment on a nutrient medium. Tissue culture had its origins at the beginning of the 20th century with the work of Gottlieb Haberlandt (plants). WHY PLANT TISSUE CULTURES ARE DONE ?? The production of clones of plants that produce particularly good flowers, fruits, or have other desirable traits. To quickly produce mature plants. The production of multiples of plants in the absence of seeds or necessary pollinators to produce seeds. The regeneration of whole plants from plant cells that have been genetically modified. The production of plants in sterile containers reduces disease transmission Allows production of plants from seeds that otherwise have very low chances of germinating and growing, i.e.: orchids and Nepenthes. To clean particular plants of viral and other infections and to quickly multiply these plants as 'cleaned stock' for horticulture and agriculture. ***For PTC, the laboratory must have the following facilities: Washing facility for glassware and ovens for drying glassware. Medium preparation room with autoclave, electronic balance and pH meter. Transfer area sterile room with laminar air-flow bench and a positive pressure ventilation unit called High Efficiency Particulate Air (HEPA) filter to maintain aseptic condition. Culture facility: Growing the explant inoculated into culture tubes at 22-28° C with illumination of light 2400 lux, with a photoperiod of 8-16 hours and a relative humidity of about 60%. *****Based on the explants some other plant tissue culture types are: 1. Organ culture 2. Meristem culture 3. Protoplast culture 4. Cell culture.

It is generally recognized that stained fecal films are the single most productive means of stool examination for intestinal protozoa. The permanent stained smear facilitates detection and identification of cysts and trophozoites and affords a permanent record of the protozoa encountered. Small protozoa, missed by wet mount examinations (of either unconcentrated or concentrated samples) are often seen on the stained smear. The Wheatley Trichrome technique for fecal specimens is a modification of Gomori's original staining procedure for tissue. It is a rapid, simple procedure, which produces uniformly well-stained smears of the intestinal protozoa, human cells, yeast, and artifact material.

Hereby you can get all about bacterial staining. MICROBIAL STAINING introduction Microbial Staining - giving color to microbes. Because microbes are colorless and highly transparent structures. Staining process in which microbes are getting color. STAINES / DYES Staines / dyes - organic compounds which carries either positive charges or negative charges or both . Based on the charges Basic stain / dyes :- stain with + ve charge . Acidic stain / dyes - stain with -ve charge . 2. Based on function of stain : Neutral stain / dyes - stain with both charges . Simple staining only one dye is used differentiation among bacteria is impossible Eg . Simple Staining. Differential staining- more than one dye is used- Differentiation among bacteria is possible- Eg. Gram's staining, Acid - fast staining. Special staining - more than one dye used - Special structures are seen. Eg. Capsule staining , Spore staining .

The tissue section is colourless because the fixed protein has the same refractive index as that of glass. We use dyes that have specific affinity with the different tissue proteins and colour them differently. Colour is seen by the eye as a result of the effect of certain electromagnetic waves on the rods and cones of the retina. These waves, which have a varying length, will determine the colour that is seen. White light being composed of all the colours of the visible spectrum varies in wavelength from 4,000 Â to 8,000 Â. If light of a specific wavelength is absorbed from white light the resultant light will then be coloured, the colour being dependent upon the particular wavelength that has been removed.
