Motilium Tablets are used for the relief of the
symptoms of nausea and vomiting. This
medicine is used in adults to treat the following
symptoms: nausea (feeling sick)
and vomiting (being sick), a heavy stomach or
bloating, a sensation of discomfort in the
stomach area or when stomach contents flow
back into the gullet, as with belching and
heartburn.
Domperidone can increase milk supply by
increasing prolactin levels. It is prescribed when
other methods of stimulating breast milk
production have proved ineffective.

➢The usual dose in adults for the treatment of acute
nausea and vomiting is one 10 mg tablet three to four
times a day, best taken 15 to 30 minutes before meals
or food. The maximum treatment duration should not
exceed one week for the treatment of acute nausea
and vomiting.
➢For other indications, the initial duration of treatment
is up to four weeks with the dosage being one tablet
of 10 mg 3 to 4 times a day.
➢Do not take medicines that neutralise stomach acid
or medicines that reduce the production of stomach
acid within 2 hours of taking Motilium (Domperidone)

➢ It has not been indicated for increasing milk supply,
but Domperidone tablets have been used successfully
in Canada and other parts of the world, and has
significantly fewer side effects than Reglan.
➢Dosage To Increase Lactation: Take one 10 mg
tablet, three times a day,. e.g. 6 am, 2 pm, 10 pm
➢After initiating treatment with Domperidone, it could
take three or four days before you notice any effect,
though sometimes mothers notice an effect within 24
hours. It appears to take two to three weeks to get a
maximum effect, but some mothers have observed
effects only after 4 or more weeks.

temperature i.e. from 20°C to 25°C (68°F
to 77°F) with excursions permitted
between 15° to 30°C (59° to 86°F).
Keep this as well as all other medicines
away from children and pets.

Contraindications
Motilium is contraindicated in patients with a
hypersensitivity to Domperidone or any other
component of this medication.
It is also contraindicated in patients with a prolactin-
releasing pituitary tumour (prolactinoma).
In patients who have known existing prolongation of
cardiac conduction intervals, particularly QTc, patients
with significant electrolyte disturbances underlying
cardiac diseases such as congestive heart failure.
Co-administration with medicines that prolong the QTc
interval, Co-administration with potent CYP3A4
inhibitors is also contraindicated.

11. Warnings

The elimination half-life of Domperidone is prolonged in cases
of severe renal impairment. For repeated administration, the
dosing frequency of Domperidone should be reduced to once
or twice daily depending on the severity of the impairment. The
dose may also need to be reduced.
Domperidone has been associated with prolongation of the QT
interval on the electrocardiogram. During post-marketing
surveillance, there have been very rare cases of QT
prolongation and torsades de pointes in patients taking
domperidone
Domperidone is contraindicated with QT-prolonging drugs
including Apomorphine, unless the benefit of co-administration
with apomorphine outweighs the risks, and only if the
recommended precautions for co-administration mentioned in
the apomorphine summary of product characteristics are
strictly fulfilled.

➢Common Motilium side effects can include: Depression,
Anxiety, Libido Decreased/Loss of Libido, Headache,
Somnolence, Akathisia, Diarrhoea, Galactorrhoea,
Amenorrhoea, Breast Pain, Menstruation Irregular, Lactation
Disorder, Rash, Pruritus, Breast enlargement /
Gynaecomastia, Breast Tenderness and Asthenia.
➢Tell your doctor immediately in case you notice any of the
following as you may need urgent medical attention: fast or
irregular heart beats , swelling of hands, ankles or feet,
passing urine more frequent or pain when passing urine.
➢STOP using Motilium (Domperidone Tablets) and inform
your doctor immediately in case any of the following occur:
you have an allergic reaction to Motilium (allergy can be
recognised, for instance, by skin rash, itching, shortness of
breath and/or swelling of the face, lips, mouth, tongue or throat
which may cause difficulty in swallowing or breathing)

Symptoms of Motilium (Domperidone
Tablets) overdosage may include
agitation, altered consciousness,
convulsion, disorientation, somnolence
and extrapyramidal reactions.

There is no specific antidote to
Domperidone Maleate (Motilium tablets),
but in the case of a large overdose, gastric
lavage within one hour of ingestion as
well as the administration of activated
charcoal could be useful.
Anticholinergics, antiparkinson drugs could
be useful in controlling extrapyramidal
reactions.
The patient should be observed closely and
supportive measures must be used.

15. Domperidone During Pregnancy

Motilium tablets (Domperidone Maleate)
has been classified by the US FDA as
Pregnancy Category Not Classfied (AU
Category B2).
There no adequate and well controlled
studies in pregnant women.
Since animal studies are not always
predictive of human response and there
are limited post-marketing data on the use
of Domperidone in pregnant women, this
medicine should be used during pregnancy
only if clearly needed.
