A Reverse phase HPLC method was developed for estimation of the Lamivudine in bulk and tablet formulation by using ODS column (250mm×4.6mm, 5µm) and Acetate buffer: acetonitrile (50:50) as mobile phase, at a flow rate of 1.5ml/min. The detection was carried at the 272nm the retention time of the Lamivudine is 1.850. The developed method was validated for the various parameters as per the ICH guidelines like accuracy precision, linearity and range, Robustnes. Linearity was obtained in the concentration range of 10µg/ml to 50µg/ml with correlation coefficient of 0.999. The accuracy of the method was assessed by recovery studies at three different concentration levels. The percentage recovery of Lamivudine was found to be in the range of 98% -102%. The method was found to be precise as indicated by the repeatability, inter-day, intra-day analysis, showing %RSD less than 2. Key words: RP-HPLC, Lamivudine, Pharmaceutical dosage form.

Dutasteride and Tamsulosin is Anti-hyperplasia and Anti-hypertensive drug.The determinationwas carried out byusingsymmetryC-18columnwith Methanol:0.1M Monobasic potassiumdihydrogenphosphate buffer(75:25) Adjusted the pH to 2.5 with Ortho phosporic acid as the mobile phase and with the detection wavelength of274 nmrespectively.The flow rate is 0.7 ml/min.TheRetentiontime of Dutasteride,Tamsulosin Hcl was 2.218 minand 6.599 min respectively.Linearityforthe Dutasteride and Tamsulosin Hcl were found inthe rangeof 25-75µgmand 20-60µgm respectively.The limitof quantificationforbothdrugs wasfound to be30,24µg respectively.The recoveries of Tamsulosin and Dutasteride were found to be inthe range of 99.81-99.90 %and98.00-102.00%, respectively. The proposed method was validated suitably and canbeused for routine analysis. The degradation studies indicated Dutasteride and Tamsulosin Hcl to besusceptible to neutralhydrolysis, while Dutasteride and Tamsulosin Hcl showed degradation inacid, H2O2,photolytic and inpresenceof UV radiation.The degradation productsof Dutasteride andTamsulosin Hcl inacidic and photolytic conditions were well resolved from the pure drug with significant differences in the irretention time values. This method can be successfully employed forsimultaneous quantitative analysis of Dutasteride and Tamsulosin Hcl in formulations.

SIMULTANEOUS QUANTIFICATION OF TELMISARTAN AND METOPROLOL SUCCINATE IN TABLET...Jing Zang
An accurate and precise liquid chromatographic method was developed for the simultaneous estimation of telmisartan and metoprolol succinate in tablets. The chromatographic analysis was performed on Nucleosil C18 Column (250*4.6mm 5µ particle size) with mobile phase consisting of acetonitrile and potassium di-hydrogen orthophosphate buffer (pH- 2.8) in the ratio 60:40v/v, at a flow rate of 0.8ml/min and eluents monitored at 220nm. The retention time for telmisartan was found to be 3.392 and for metoprolol succinate it was found to be 5.221 minutes. The proposed method is simple, accurate and precise and could be successfully employed in routine quality control for the simultaneous estimation of telmisartan and metoprolol succinate in tablets.

Major process related unknown impurity associated with the synthesis of Hydralazine hydrochloride bulk drug was detected by high performance liquid chromatography (HPLC) and was subjected to high resolution accurate liquid chromatography mass spectroscopy (HR/AM-LCMS) for identification. The proposed impurity was isolated from Hydralazine hydrochloride active pharmaceutical ingredient (API) by preparative chromatographic method and was injected on HPLC for comparison of retention time with that of the unknown process related impurity in Hydralazine hydrochloride. The molecular ion peak of preparatively isolated impurity and that of unknown process related impurity in Hydralazine hydrochloride were compared for confirmation. The postulated structure was unambiguously confirmed with the help of HR/AM- LC MS/MS, NMR and FTIR data proposed to be 1-(2-phthalazin-1-ylhydrazino)phthalazine (Hazh Dimer). This impurity of Hydralazine hydrochloride is not been previously reported. A rapid Acquity H-class gradient method with runtime of 15.0min was developed for Quantitation on Unisphere Cyno column and validated for parameters such as accuracy, precision, linearity and range, robustness. The LOD and LOQ of method were 0081% and 0.0246% respectively.

Analytical Method Development and Validation for the Estimation of Zolmitript...ijtsrd
In this work the authors have proposed a simple, specific, economic and accurate reverse phase liquid chromatographic method for the estimation of Zolmitriptan as an active pharmaceutical ingredient and in pharmaceutical formulation. The main objective of the current research paper is to To develop simple, precise and accurate RP HPLC method for Zolmitriptan also to validate the developed method as per ICH guideline Q2R1 and to explore the applicability of the method in finished product formulation for estimation of Zolmitriptan during its lifecycle. The objective was achieved by optimized condition with Phonemenex C18 column 150mm×4.6mm , 5µm. And mobile phase Phosphate buffer pH 3.5 85 Methanol 15. The separation was done with a flow rate of 0.9ml min, detection with 224nm. The retention was found to be 3.57 minute. LOD and LOQ were found to be 2.45 and 7.42 respectively. So in order to obtain the correct results various validations methods are performed to get the results. A novel and simple reversed-phase liquid chromatographic method has been established for the determination of saxagliptin and metformin HCl Saxagliptin and metformin HCl is used to control Type 2 diabetes. The proposed work was performed on Young Lin (S.K) isocratic System UV Detector. Saxagliptin and metformin HCl is used to control Type 2 diabetes. The proposed work was performed on Young Lin (S.K) isocratic System UV Detector C18 column (150 mm × 4.6 mm). A mixture of potassium phosphate, mobile phase in this method with flow rate of 0.7 mL/min (UV detection at 203 nm) and the method was validated as per the ICH guidelines. Forced degradation studies were performed by exposing the drug saxagliptin and metformin HCl to acidic, alkaline, oxidation, and thermal stress degradations. The proposed reversed-phase-high-performance liquid chromatography method was found to be robust and specific, and this method is suitable for the assay of pharmaceutical dosage forms as well as kinetic studies.

The Ohio State University Comprehensive Cancer Center, will provide insight into new directions in targeted therapeutic approaches for older adults with mantle cell lymphoma.
STATEMENT OF NEED
Mantle cell lymphoma (MCL) is a rare, aggressive B-cell non-Hodgkin lymphoma (NHL) accounting for 5% to 7% of all lymphomas. Its prognosis ranges from indolent disease that does not require treatment for years to very aggressive disease, which is associated with poor survival (Silkenstedt et al, 2021). Typically, MCL is diagnosed at advanced stage and in older patients who cannot tolerate intensive therapy (NCCN, 2022). Although recent advances have slightly increased remission rates, recurrence and relapse remain very common, leading to a median overall survival between 3 and 6 years (LLS, 2021). Though there are several effective options, progress is still needed towards establishing an accepted frontline approach for MCL (Castellino et al, 2022). Treatment selection and management of MCL are complicated by the heterogeneity of prognosis, advanced age and comorbidities of patients, and lack of an established standard approach for treatment, making it vital that clinicians be familiar with the latest research and advances in this area. In this activity chaired by Michael Wang, MD, Professor in the Department of Lymphoma & Myeloma at MD Anderson Cancer Center, expert faculty will discuss prognostic factors informing treatment, the promising results of recent trials in new therapeutic approaches, and the implications of treatment resistance in therapeutic selection for MCL.

As it possible to develop a HPLC method for simultaneous determination
of losartan, hydrochlorothiazide and their degradation product by using a
single sample preparation ?
Losartan potassium is the active ingredient in tablets COZAAR and is
combined in the product HYZAAR.
WHY COMBINATION ?
Studies have shown an increase in Losartan efficacy when a low dose of
hydrochlorothiazide (HCTZ) is administered concomitantly due to stimulation
of the renin–angiotensin system.
GOAL OF WORK :
To develop and validate a single high-performance liquid chromatography
method selective for the eight main components of tablets HYZAAR®. 4

MOBILE PHASE : (Both the Losartan tablets
and the Losartan/HCTZ tablets)
Mobile Phase A : phosphate buffer solution of KH2PO4 and Na2HPO4 (pH 7.0,
0.02 M): Acetonitrile in the ratios of 85:15(v/v) and 93:7(v/v).
Mobile Phase B: 100% Acetonitrile
CONDITIONS:
For Losartan tablets:
the profile is a linear gradient from Mobile Phase A–Mobile Phase B (80:20,
v/v) to Mobile Phase A–Mobile Phase B (40:60, v/v) over 10 min. The mobile
phase composition is ramped back to Mobile Phase A Mobile Phase B (80:20,
v/v) for equilibration prior to the next injection for a total run time of 15 min.
Mobile phase flow rate: 1ml/min.
Ambient column temperature
Injection volume: 10 µl.
UV detection at: 250 nm.

For Losartan/HCTZ tablets:
The two-step gradient profile begins with a 12 min linear gradient from 100%
Mobile Phase A to Mobile Phase A–Mobile Phase B (92:8, v/v). This is
followed by a linear ramp over the next 16 min to Mobile Phase A–Mobile
Phase B (38:62, v/v). Finally, the composition is returned to 100% Mobile
Phase A over 2 min followed by a 5 min equilibration for a total run time of
35 min.
Mobile phase flow rate: 1ml/min.
Column temperature: 35ºC.
Injection volume: 20 µl.
UV detection at: 280 nm.

STANDADRD AND SAMPLE SOLUTION PREPARATION:
Losartan tablets:
1) 100% working std. solution for losartan tablets:
Losartan potassium reference std. dissolve in mobile phase A to get
0.25 mg/ml conc.
diluated by mobile phase A
1% degraded level std. 0.1% sensitivity solution.
2) System suitability solution:
containing 0.2-0.5% of losartan degrades, compound E and F was
produced by acid stressing under accelerated heating condition of 105ºC for
1-2 hrs.
3) Sample losartan tablet solution:
tablet dissolved in mobile phase A
Diluted by mobile phase A Sonication and shaking
losartan final conc. Of 0.25 mg/ml.

Losartan/ HCTZ Tablets:
1)100% working std. for losartan/HCTZ Tablet:
Dissolving reference std. in diluent(0.02 M, NaH2PO4, pH 2.5) and
acetonitrile (70:30 v/v) to get a conc. Of 0.4 mg/ml losartan, 0.1 mg/ml HCTZ
and 0.001 mg/ml CTZ and DSA.
DILUTION
1% Degrade level std. 0.1% sensitivity std.
2) System suitability solution:
Combing 100% working std. solution +stressed losartan solution as described
previously (contain HCTZ, CTZ, Losartan degrades, E and F)
3) A single sample solution:
by dissolving tablet in (60:40 v/v) acetonitrile and diluent with mechanical
shaking for 30 min. further dilute by a diluent to yield losartan and HCTZ Conc.
At 0.4 and 0.1 mg/ml resp.

METHOD DEVELOPMENT AND OPTIMIZATION:
Losartan tablets:
1) The method was developed for the simultaneous determination of the active
ingredient at the 100% level and degradation products at 0.1% levels in
Losartan tablets.
2) To obtain the adequate retention time for polar compound losartan and
maintaining reasonable elution time for less polar compound F and F
symmetry C8 (150×3.9 mm, 5 µ𝑚 particle size) give a best resolution and
shortest retention time for active and degrade compound.
3) The interference of acetonitrile at the detection wavelength 230 nm so use
of 250 nm detection wavelength.
4) Sensitivity to impurities in phosphate salt so use of fresh phosphate salt.
5) By use high conc. Of acetonitrile in mobile phase and methanol to elute
active drug and degrade isocratically but the isocratic method of run less
than 1 hr. for all the 3 compound are not realised so use of 15 min. gradient
method developed and validated successfully to quantitation for losartan
and its degrade.

Losartan/HCTZ Tablets:
1) The mobile phase (phosphate buffer) pH was changed from 2.5 to 7.0 to
optimize the Losartan retention time, while still allowing separation of CTZ,
DSA, and HCTZ from the solvent front and each other.
2) Normal fluctuations in ambient laboratory temperatures affected the
resolution of the HCTZ–CH2–HCTZ impurity and Losartan so column temp.
Control at 35ºC.
3) Losartan is present at four times the concentration of HCTZ in the tablet
formulations; therefore, a higher wavelength of 280 nm was chosen to
achieve similar degree of absorbance values for the two active ingredient
peaks.

4) PRECISION:
For losartan tablet:
RSD 0.2% and 0.3% at 1 and 100% level respectively.
For losartan/HCTZ tablets:
RSD 0.1% and 0.0.2% at 100 and 1% level resp. for losartan and HCTZ.
5) RUGGEDNESS:
Ruggedness chemist reported retention times for all major components
detected within 0.5 min. and 0.2 min. from those observed by the developing
chemist for losartan and losartan/HCTZ tablets resp.
6)ROBUSTNESS:
For losartan tablet: The method was found to be very robust.

CONCLUSION:
1)Successful method develop of HPLC for separation and quantitation of all
related compounds in COZAAR and HYZAAR TABLET.
3) This method provide advantages of time saving, decrease no. of samples
preparation, instrument run time, solvent and drug waste etc.
4) Losartan tablet method was found to be very robust, whereas losartan/HCTZ
tablet method found temp. And pH as critical conditions.
