The dissolution test is an important means of assuring the continuing performance of non-solution orally administered drug products. The development of a dissolution test procedure is briefly discussed in USP general information chapter In Vitro and In Vivo Evaluation of Dosage Forms 1088, whereas general information chapter Validation of Compendial Procedures 1225 gives limited validation information for dissolution testing. Neither of these two chapters provides a level of detail and focus sufficient for dissolution testing. In 2001, a Stimuli article provided an initial rationale and discussion of content for a new general information chapter. The new chapter, The Dissolution Procedure: Development and Validation 1092, was intended to supplement the information in 1088 and 1225 and provided step-by-step detail for development and validation as well as offering information on new technology and equipment. In 2006, the chapter became official with the Second Supplement to USP 29–NF 24 (2–4).
The General Chapters—Dosage Forms Expert Committee 2010–2015 placed the review and possible revision of The Dissolution Procedure: Development and Validation 1092 on its work plan for the 2010–2015 revision cycle (2011) .

A simple and selective LC method is described for the determination of Quinapril and Tolcapone tablet dosage forms. Chromatographic separation was achieved on a c18 column using mobile phase consisting of a Mixed Phosphate buffer (KH2PO4 +K2HPO4): Acetonitrile 40:60, with detection of 239 nm. Linearity was observed in the range 50 - 150 µg /ml for Quinapril (r2 =0.995) and 62.5- 187.5µg /ml for Tolcapone (r2 =0.999) for the amount of drugs estimated by the proposed methods was in good agreement with the label claim.
The proposed methods were validated. The accuracy of the methods was assessed by recovery studies at three different levels. Recovery experiments indicated the absence of interference from commonly encountered pharmaceutical additives. The method was found to be precise as indicated by the repeatability analysis, showing %RSD less than 2. All statistical data proves validity of the methods and can be used for routine analysis of pharmaceutical dosage form.

The present analytical work is a unique method development and validation for the determination of dissolution of Eltrombopag using reverse phase high-performance liquid chromatography (HPLC) with isocratic elution technique. HPLC method for quantification of drug in dissolution samples of Eltrombopag tablet is developed and validated. About 0.5% polysorbate 80 in phosphate buffer of pH −6.8 is used as dissolution medium and paddle (USP-II) as apparatus at 50 rpm. The sample was withdrawn after 45 min. The developed HPLC method was used for quantitative estimation of drug release in dissolution samples of Eltrombopag tablet. Here, the stationary phase used was Xbridge C18 (50 mm × 4.6 mm × 5 μm), mobile phase was 25% ammonium formate and 75% acetonitrile. pH of the buffer solution was maintained at 3.0, flow rate 1.0 ml/min. Eluted material underwent for monitoring at the detector wavelength of 230 nm. Retention time for Eltrombopag was found to be 2.16 min; and linearity range was 3.516 µg/mL–131.862 µg/mL. The new method was evaluated according to the ICH guideline and as far as validation results are concern correlation coefficient value that was 1.0000 for the compound, percentage recovery 99.4%, and repeatability results relative standard deviation 0.6 for Eltrombopag. The developed HPLC method was found to be a simple and rapid one for regular analysis in professional laboratory.

HPLC is a chromatographic technique employed in active compound chemistry and biochemistry to separate a mixture and substances with the goal of identifying, measuring, and purifying the different components of the mixture. Its a much better variety of column and traditional chromatography. The objective of the research work is to develop and validate a simple and accurate reverse phase chromatographic method to estimate amount of drug in dosage form. The developed method successfully can be applied to estimate the amount of Dapagliflozin in tablet dosage form. After oral administration of dapagliflozin, the maximum plasma concentration Concentration max under two hours. High performance liquid chromatographic system was alleviated according to the chromatographic settings. After attaining the steady base line, to verify the system suitability, a single 40 µg ml of standard solution proportional to 100 test concentration of dapagliflozin was injected into the HPLC system. The gradient mobile phase flow rate programming assisted in optimising the lengthy run duration and resolution of sample analysis, making the approach more cost effective and quick. Validation of the developed and optimized HPLC method was carried out according to ICH guidelines with respect to parameters such as linearity, specificity, precision and accuracy. RESULTS: Overall life span (LS) was 2252.1±1742.5 days and cumulative 5-year survival (5YS) reached 73.2%, 10 years – 64.8%, 20 years – 42.5%. 513 LCP lived more than 5 years (LS=3124.6±1525.6 days), 148 LCP – more than 10 years (LS=5054.4±1504.1 days).199 LCP died because of LC (LS=562.7±374.5 days). 5YS of LCP after bi/lobectomies was significantly superior in comparison with LCP after pneumonectomies (78.1% vs.63.7%, P=0.00001 by log-rank test). AT significantly improved 5YS (66.3% vs. 34.8%) (P=0.00000 by log-rank test) only for LCP with N1-2. Cox modeling displayed that 5YS of LCP significantly depended on: phase transition (PT) early-invasive LC in terms of synergetics, PT N0—N12, cell ratio factors (ratio between cancer cells- CC and blood cells subpopulations), G1-3, histology, glucose, AT, blood cell circuit, prothrombin index, heparin tolerance, recalcification time (P=0.000-0.038). Neural networks, genetic algorithm selection and bootstrap simulation revealed relationships between 5YS and PT early-invasive LC (rank=1), PT N0—N12 (rank=2), thrombocytes/CC (3), erythrocytes/CC (4), eosinophils/CC (5), healthy cells/CC (6), lymphocytes/CC (7), segmented neutrophils/CC (8), stick neutrophils/CC (9), monocytes/CC (10); leucocytes/CC (11). Correct prediction of 5YS was 100% by neural networks computing (area under ROC curve=1.0; error=0.0).
CONCLUSIONS: 5YS of LCP after radical procedures significantly depended on: 1) PT early-invasive cancer; 2) PT N0--N12; 3) cell ratio factors; 4) blood cell circuit; 5) biochemical factors; 6) hemostasis system; 7) AT; 8) LC characteristics; 9) LC cell dynamics; 10) surgery type: lobectomy/pneumonectomy; 11) anthropometric data. Optimal diagnosis and treatment strategies for LC are: 1) screening and early detection of LC; 2) availability of experienced thoracic surgeons because of complexity of radical procedures; 3) aggressive en block surgery and adequate lymph node dissection for completeness; 4) precise prediction; 5) adjuvant chemoimmunoradiotherapy for LCP with unfavorable prognosis.

11. Aim & Objective
Aim
The main aim of the present study is to develop an accurate, precise, sensitive, selective, reproducible
and rapid analytical technique for simultaneous estimation of Levodropropizine, Chlorpheniramine in
bulk and Syrup dosage form.
Objective and Plan:
Following are the objectives of the present work:
To develop a new stability indicating HPLC method for simultaneous estimation of
Levodropropizine and Chlorpheniramine and to develop the validated method according to ICH
guidelines.
To apply the validated method for the simultaneous estimation of Levodropropizine and
Chlorpheniramine in pharmaceutical formulation

13. Materials and Method
• Materials:
Levodropropizine(LZD) and Chlorpheniramine(CPM) pure drugs (API), Combination
Levodropropizine and Chlorpheniramine syrup (RESWAS), Distilled water, Acetonitrile,
Phosphate buffer, , Methanol, Potassium dehydrogenate ortho phosphate buffer, Ortho-
phosphoric acid. All the above chemicals and solvents are from Rankem
• Instruments:
• Electronics Balance-Denver
• pH meter -BVK enterprises, India
• Ultrasonicator-BVK enterprises
• WATERS HPLC 2695 SYSTEM equipped with quaternary pumps, Photo Diode Array
detector and Auto sampler integrated with Empower 2 Software.
• UV-VIS spectrophotometer PG Instruments T60 with special bandwidth of 2 mm and
10mm and matched quartz cells integrated with UV win 6 Software was used for
measuring absorbances of Levodropropazine and Chloropheniramine solutions.

14. Standerd and Sample Preparation
• Diluent: Based up on the solubility of the drugs, diluent was selected, Acetonitrile and Water taken in the ratio of
50:50
• Preparation of Standard stock solutions: Accurately weighed 30mg of Levodropropizine, 2mg of
Chlorpheniramine and transferred to individual 50 ml volumetric flasks separately. 3/4 th of diluents was added to
both of these flasks and sonicated for 10 minutes. Flasks were made up with diluents and labeled as Standard stock
solution 1and 2. (600µg/ml of Levodropropizine and 40µg/ml of Chlorpheniramine )
• Preparation of Standard working solutions (100% solution): 1ml from each stock solution was pipetted out and
taken into a 10ml volumetric flask and made up with diluent. (60µg/ml Levodropropizine of and 4µg/ml of
Chlorpheniramine )
• Preparation of Sample stock solutions: Syrup equivalent to 30mg Levodropropizine and 2mg of Chlorpheniramine
was transferred into a 50 ml volumetric flask, 20ml of diluents was added and sonicated for 25min, further the volume
was made up with diluent and filtered by HPLC filters (600µg/ml of Levodropropizine and 40µg/ml of
Chlorpheniramine )
• Preparation of Sample working solutions (100% solution): 1ml of filtered sample stock solution was transferred to
10ml volumetric flask and made up with diluent. (60µg/ml of Levodropropizine and 4µg/ml of Chlorpheniramine )
• Preparation of buffer:
0.1% OPABuffer: 1ml of Ortho Phosphoric acid was diluted to 1000ml with water.

33. FORCED DEGRADATION STUDIES
Acid degradation: 2 ml of stock solution of Levodropropzine and Chlorpheniramine were
taken, and 1ml of 1N Hydrochloric acid was added and kept at 600c for 24 hrs. Then the
solution was neutralized with 1ml of 1N Sodium hydroxide. The resultant solution was
diluted with diluent and 20 µl solutions were injected into the system and chromatogram
was recorded.
Base degradation: 2 ml of stock solution of Levodropropizine and Chlorpheniramine were
taken, and 1 ml of 1N sodium hydroxide was added and kept at 60⁰c for 24hrs. Then the
solution was neutralized with 1 ml of 1N Hydrochloric acid. The resultant solution was
diluted with diluent and 20 µl were injected into the system chromatogram was recorded .
Peroxide degradation: 2ml of stock solution of Levodropropizine and Chlorpheniramine
were taken, and 3 ml of 30 % Hydrogen peroxide was added and kept at 60⁰c for 24 hrs.
The resultant solution was diluted with diluent. and 20 µl were injected into the system and
chromatogram was recorded.
Thermal degradation: 2 ml of stock solution of levodropropizine and Chlorpheniramine
were taken and placed in oven at 600c for 4hrs to study dry heat degradation For HPLC
study, the resultant solution was diluted with diluent and 20µl were injected into the system
and the chromatogram was recorded.
Photo degradation: 2 ml of stock solution of Levodropropizine and Chlorpheniramin
ewere taken and placed in sunlight for 24hrs For HPLC study, the resultant solution was
diluted with diluent solution and 20µl were injected into the system and the chromatogram
was recorded .

37. CONCLUSION:
The present work concluded that stability indicating assay method by RP-HPLC was
simple, accurate, precise, and specific and has no interference with the placebo and
degradation products. Hence these can be used for routine analysis of Levodropropizine and
Chlorpheniramine malete.
The developed HPLC method for the estimation of selected drugs is simple, rapid,
accurate, precise, robust and economical. The mobile phase and solvents are simple to
prepare and economical, reliable, sensitive and less time consuming.
