To observe the Raman spectrum, it is necessary to separate the collected Raman scattered
light into individual wavelengths. In dispersive Raman instruments, this is accomplished by
focusing the Raman signal on a grating, which spatially separates the different wavelengths.
This spatially dispersed beam is directed to a CCD.
Fluorescence occurs when the virtual energy level overlaps an upper electronic level, so
as the energy of the laser gets higher (shorter wavelength), the likelihood of fluorescence
increases. The phenomenon is excitation wavelength dependent, so a sample that fluoresces
at one wavelength may not at another. Thus, when selecting an instrument, it is important to
look for rapid and effortless exchanges between two difficult excitation lasers.
The grating has a strong influence on spectral resolution and instrument throughput.
Gratings have many lines or grooves “blazed” into the surface, which disperse the incoming
light. The higher the number of grooves on the grating, the wider the dispersion angle of
the exiting rays.
Raman signal from molecules adsorbed on certain
metal surfaces can be 5-6 orders of magnitude stronger then the Raman signal from the same molecules in bulk volume.
The exact reason for such dramatic improvement is still under discussion. However, since intensity of Raman signal is
proportional to the square of electric dipole moment P = αE, there are two possible reasons - the enhancement of
polarizability α, and the enhancement of electrical field E.
The first enhancement of polarizability a may occur because of a charge-transfer effect or chemical bond formation
between metal surface and molecules under observation. This is a so-called chemical enhancement.
The second one takes into account interaction of the laser beam with irregularities on the metal surface such as metal
micro-particles or roughness profile. It is believed that laser light excites conduction electrons at the metal surface leading
to a surface plasma resonance and strong enhancement of electric field E. It is also called electromagnetic enhancement.
Frequency
of the first laser is usually constant, while the frequency of the second one can be tuned in a way that the frequency
difference between the two lasers equals exactly the frequency of some Raman-active mode of interest. This particular
mode will be the only extremely strong mode in the Raman signal.
Two laser beams with frequencies υ1 and υ2 (υ1 > υ2) interact coherently, and because of the wave mixing, produce strong
scattered light of frequency 2υ1 - υ2 (Fig. 3). If the frequency difference between two lasers υ1 - υ2 is equal to the frequency
um of a Raman-active rotational, vibrational or any other mode then a strong light of frequency υ1 + υm is emitted.
In other words, to obtain strong Raman signal the second laser frequency should be tuned in a way that υ2 = υ1 - υm . Then
the frequency of strong scattered light will be 2υ1 - υ2 = 2υ1- (υ1 - υm) = υ1 + υm, which is higher then the excitation frequency
u1 and therefore considered to be Anti-Stokes frequency.
Coherent Anti-Stokes Raman Spectroscopy derives its name from the fact that it uses two Coherent laser beams and the
resulting signal has Anti-Stokes frequency.
Very strong laser pulse with electric field strength > 109 V·cm-1 transforms up to 50% of all laser pulse energy into coherent beam at Stokes frequency υ0 - υm (Fig. 2).
The Stokes beam is unidirectional with the incident laser beam. Only the mode um which is the strongest in the regular
Raman spectrum is greatly amplified. All other, weaker Ramanactive modes are not present. The Stokes frequency is so
strong it acts as a secondary excitation source and generates the second Stokes line with frequency υ0 - 2υm. The second
Stokes line generates the third one with the frequency υ0 - 3υm etc. Stimulated Raman technique enjoys 4-5 orders of
magnitude enhancement of Raman signal as compared to the spontaneous Raman scattering.
The advantages
of the Fourier transform technique are necessary to provide the sensitivity to extract functional
spectral information from this lower intensity signal.
By virtue of the Fourier transform technique, FT-Raman offers:
• High resolution with minimal throughput loss
• Measurement of all wavelengths at once
• Increased signal-to-noise by signal averaging
• Superior wavelength accuracy due to the internal calibration inherent
to an interferometer

that the radiation from each path is reflected back by mirrors onto the divider
called the beam splitter. The recombined two beams may either interact
constructively or destructively depending upon the phase difference of the
two optical paths. This constructive or destructive interference will vary as
the path length in one of the arms is varied. The resulting pattern forms the
interferogram that represents the relationship between the energy and the
path difference in the two arms of the interferometer. The interference
pattern clearly will be a function of the wavelength of light because the
relative path differences will be expressed as different integral values of
wavelength. Interferograms represent the interference of the incident wavelengths,
and hence they are converted into a spectrum by using a Fourier
transform algorithm. The Michelson interferometer, such as illustrated in
Figure 2, was the first instrument to split a source radiation into two separate
beams, change the path length of one of the paths, and recombine the
radiation on the beam splitter to cause interference.
Raman spectra yield more information about certain types of organic compounds than do their infrared counterparts.
The Raman technique is often superior to infrared for spectroscopy investigating inorganic systems because aqueous solutions can be employed. In addition, the vibrational energies of metal-ligand bonds are generally in the range of 100 to 700 cm-1, a region of the infrared that is experimentally difficult to study. These vibrations are frequently Raman active, however, and peaks with values in this range are readily observed. Raman studies are potentially useful sources of information concerning the composition, structure, and stability of coordination compounds.
