Abstract Peroxidase is a turnip enzyme; it is used in the oxidation of hydrogen peroxide to lower
activation energy, speeding up the reaction. The activity of peroxidase is highly dependent on its
environment and most importantly the pH level. Peroxidase has been the focus of many recent studies
and is believed to possibly reduce swelling among other things. We conducted an experiment testing
the effect different levels of pH had on the reaction rate of peroxidase. In the experiment we created
different solutions all containing hydrogen peroxide, peroxidase, and guaiacol. However each cuvette

In the blank cuvettes, the 2 mL of pH levels 2,5,7, and 10 were added first, followed by the 1mL of
peroxidase. However our experimental cuvettes contained both hydrogen peroxide and guaiacol.
Before the cuvettes were placed in the spectrophotometer we added 0.1 mL of H202, followed by 0.02
mL of guaiacol these two substances were added last immediately before they were placed in the
spectrophotometer. Para film was placed over the cuvette opening, and the assay solution was shaken
and placed into the spectrophotometer where its saturation level was then tested. We recorded the
saturation level of the solution every 15 seconds for 3 minutes. This process was repeated two times
for each pH level for a total of two trials.
Results
Figure one depicts the reaction rate of peroxidase enzyme over time. The y axis shows the absorbance
of the assay solutions, and the x axis depicts time it took for the reaction to occur in seconds.
Figure 2 is a representation of the average saturation of each cuvette at a specific point time as a
function. The y axis shows the specific saturation points from figure 1, and the x axis provides the
different levels of pH. The pH scale provided on the x axis ranges from 0 to 14, 0 being the most
acidic and 14 being the most basic. The point chosen from figure 1 was the saturation levels of each
