El árbol de granada posee efectos medicinales conocidos desde hace miles de años y referidos en Antiguo Testamento, Torá y Talmud.
Dioscórides describe las propiedades terapéuticas de la granada en Materia médica (+ 600 remedios vegetales) (77-78 DC). En Medicina Ayurvédica se considera la granada como una farmacia en si misma. Posee más de 100 compuestos fotoquímicos: 1- Polifenoles; 2- Flavonoides: a- antocianidinas (delfinidina, cianidina, pelargonidina), b- flavonoles (luteína, Quercetin y kaempferol); 3- Taninos condensados (proantocianidinas); 4- Taninos hidrolizables (TH) (elagitaninos, punicalaginos, galotaninos); 5- Esteroides (estradiol, estriol, estrona, testosterona, ácido ursólico) y 6- Catequinas. Los taninos hidrolizables son los polifenoles predominantes responsables de >90% de la actividad antioxidante. Los Punicalaginos son los responsables del 50% de la capacidad antioxidante de la granada. Las semillas poseen ácidos grasos (>65% ácico punícico). Además existen fotoquímicos en cáscara, pieles, hojas, corteza. Los polifenoles están asociados con menor riesgo a desarrollar enfermedades inflamatorias, cardiovasculares, neurodegenerativas y tumores. Los efectos antitumorales de la granada son: 1- Antiproliferativos y pro-apoptosis; 2- Inhibición factor nuclear κB (NF-κB); 3- Anti-angiogénesis; Anti invasión tumoral. Efectos antiproliferativos: detención del crecimiento. La apoptosis es la muerte celular inducida (suicidio). Diferentes partes de la granada (fruta, pieles, cáscara, semillas) al natural y fermentadas presentan efectos antiproliferativos y pro-apoptosis en estudios in vivo e in Vitro. Se han estudiados los efectos en los siguientes tumores: próstata, mama, colon, pulmón y piel. En el cáncer de próstata se han estudiados los efectos antiproliferativos y pro-apoptopsis en células hormono sensibles y hormono resistentes sin afectar a células prostáticas normales. Los animales que bebían zumo de granada mostraban reducción del 85% en el PSA frente a los que bebían agua. Las urolitinas (metabolitos ác. Elágico) inhiben cáncer de próstata. La inducción de apoptosis se realiza a través de inhibición del IGF. En cáncer de mama el aceite de semillas presenta efecto antitumoral en células de cáncer de mama. El extracto de toda la granada ofrece mejor resultado que por separado. El aceite de semillas, zumo natural o fermentado, inhiben síntesis de estrógenos y actividad de aromatasa. El zumo fermentado inhibe en 90% proliferación celular y en 47% la formación de tumores inducidos por el carcinógeno DMBA. En cáncer de colon el aceite de semillas suprime carcinogénesis colónica.
Todos los componentes de la granada inducen apoptosis celular. Los elagitaninos y sus metabolitos (urolitinas A y B) inhiben la proliferación e inducen apoptosis en células HT-29. Se ha observado además que la granada mejora la función eréctil y la calidad del semen.
