We got a slide, added a drop of water to it, and then put our Daphnia magna onto it.
We then put our slide under the microscope, tried to locate the Daphnia magna s
heart, and counted its heart beat per 10 seconds. We couldn t get the D. magna to
stop moving so what I did was add pieces of a cotton ball to help the Daphnia
magna stop moving as much. Then, after finally counting the Daphnia magna s
heart beat for 10 sec, we turned off the microscope s light for three minutes. We did
this two more times (for a total of 3 times) and recorded our data. We multiplied
the 10 sec heart beat of the D. magna by 6 to get a total of one minute. After we
finished with the water part (control group), we added alcohol to our slide and
inspected the difference in the Daphnia magna s heart beat through the microscope.
We counted the new heart beat for 10 sec. We then turned the light off for 3
minutes to let the D. magna rest. We repeated this two more times (for a total of 3
times). Then we got multiplied the 10 sec H.R by 6 to get a total of one minute and

Restriction digest involves the use of restriction enzymes (also known as restriction
endonucleases) to locate specific base pair sequences in DNA. These enzymes cut, or
cleave, DNA only at their designated sequence, which is referred to as a recognition
sequence. While there are four different types of restriction enzymes (1), the only
type that was worked with in the following experiment were type II restriction
enzymes (2). These enzymes have recognition sites that are mostly palindromic
and usually consist of around four to eight base pairs. They also require only
magnesium (Mg2+) as a cofactor to operate. Cofactors are molecules that bind to
enzymes in order to activate them (3). Additionally, they cut DNA only at, or very
near to, their specified restriction site, unlike other types, which cleave at various
distances from their recognition site (1). The restriction enzymes that will be used
in following experiment are Hind III, PVU II, and Bgl I (2). Hind III recognizes
and cuts DNA at the sequence AAGCTT. It is isolated from Haemophilus
influenzae (4), which is a bacteria that is the cause of several diseases, including
pneumonia, and meningitis. (5) When Hind III is used to cleave DNA, the end
result will have sticky ends, which means that there will be a few unpaired

realizes things were not as they seemed, the artist suffers severely; he almost feels as
if he s been physically harmed. The sink containing blood and crushed veneer
exemplifies the artist s unsuccessful attempts to wash away the hurt and brokenness
he is experiencing due the revelation that everything is not as it seemed.
Vernon now speaks of wanting to be free from this toxic relationship, but also admits
obtaining this freedom is not a painless undertaking. Ropes bind substances together,
and in this situation, they symbolize the people are tied to those that they love. The
artist does not want to feel bound to this person anymore, and he desires to be free
to love another. However, this freedom doesn t seem to be attainable as the artist
explains this order s tall, a phrase commonly used to describe an arduous task.
