This document discusses matrix effects (MEs) that can occur during LC-MS/MS bioanalytical methods and presents a systematic approach to overcome MEs through different sample extraction techniques. It finds that solid phase extraction produces the cleanest samples with the lowest MEs, while protein precipitation using methanol produces the dirtiest samples with the highest MEs. Different phospholipids are identified as contributing to MEs, with longer-retained phospholipids playing a more significant role. Among extraction methods, solid phase extraction is most effective at removing phospholipids and minimizing MEs, while protein precipitation is least effective.
