MICROBIOLOGYโ€ฆโ€ฆ.
โ€ข Microbiology is the study of living organisms
that are invisible to the naked eye, such as โ€ฆโ€ฆ
โ€ข Bacteria
โ€ข Virus
โ€ข Fungi
1/24/2018 3SUNIL KUMAR .P
Microbiological Culture
โ€ข It is a method of multiplying microbial
organisms by letting them reproduce in
predetermined culture media under
controlled laboratory conditions.
โ€ข Microbial cultures are used to determine the
type of organisms and its abundance in the
sample being tested or both
1/24/2018 4SUNIL KUMAR .P
Sensitivity Testโ€ฆโ€ฆโ€ฆ
โ€ข A laboratory test perform to check the
effectiveness of a drug against microorganisms
to select the best drug regimen that is
effective..
1/24/2018 5SUNIL KUMAR .P
Purpose of culture & sensitivity test
โ€ข To guide the clinician selecting the best drug
for an individual patient.
โ€ข To control the use of inappropriate drug in
clinical practice.
โ€ข To accumulate epidemiological information on
the resistance of microorganism of public
Health importance within the community.
โ€ข To overcome the microbial drug resistance.

Culture
โ€ข Semi-Quantitative cultures are doneโ€ฆ. on
blood agar and Agar with a calibrated loop.
โ€ข Colony count of 105/ml is considered
significant.
โ€ข Counts between 104/ml and 105 ml are of
doubtful significance.
1/24/2018 SUNIL KUMAR .P 7
โ€ข Counts less than this are considered
significant if :โ€ฆ.
โ€ข The patient is on prior antibiotics
โ€ข There is obstruction in the urinary tract
โ€ข A fungal infection is present
โ€ข If pyelonephritis is present
1/24/2018 SUNIL KUMAR .P 8
Antibiotic sensitivity Testingโ€ฆ.
โ€ข If culture is suggestive of infection, an
antibiotic sensitivity test is done.
1/24/2018 SUNIL KUMAR .P 9
Types of Antibiotic sensitivity test
1/24/2018 SUNIL KUMAR .P 10
Kirby-Bauer
โ€ข This is in common use
โ€ข In this method a suitable dilution of broth
culture or a broth suspension of the test
bacterium is inoculated on the surface of solid
medium ( MHA- Mueller-Hinton or Nutrient
Agar) as a lawn by spreading with cotton
swab.
1/24/2018 SUNIL KUMAR .P 11
โ€ข After overnight incubation, the degree of
sensitivity is determined by measuring the
zones of inhibition of growth around the discs.
โ€ข Growth will be inhibited around discs
containing antibiotics to which the bacterium
is susceptible but not around those to which it
is resistant.

Stokes method
โ€ข This is suitable method for routine diagnostic
use in diagnostic laboratories.
โ€ข This technique was originally described by
stokes.
โ€ข This incorporates built-in controls against
many variables and therefore provides depend
results.
1/24/2018 SUNIL KUMAR .P 14
Dilution tests
โ€ข Here, serial dilution of the drug are prepared
and inoculated with the test bacterium.
โ€ข This is too laborious for routine use.
โ€ข Dilution tests are generally employed when
the therapeutic dose is to be regulated
accurately as in the treatment of bacterial
endocarditis.
