Organizational Development Department And Is Someone That...
When selecting my topic for this research paper I reached out to a co worker of mine, Michelle
Choate. Michelle works in our Organizational Development department and is someone that I have
taken communications classes from at St. Luke s. For many years I consistently had employment
evaluations that suggested that I had significant opportunity to improve my communication skills.
Through the courses taught by Michelle, I was able to discover some characteristics about myself and
implement her teachings into my life. I did not realize it prior to writing this paper but she has been a
valuable asset to increasing my emotional intelligence. When selecting my topic I wanted to know,
from her perspective, what topics in communication where ... Show more content on Helpwriting.net

Characteristics of self regulation include; trustworthiness, integrity and openness to change. Internal
motivation is your passion to complete a project or a goal that is beyond financial compensation.
Empathy is the ability to understand the emotions of others, and treating them in a way that is
compatible with their emotional reactions. And finally, social skills include one s ability to manage
and build social and professional networks, be persuasive, build teams and rapport.
Who has emotional intelligence?
Emotional intelligence (EQ) is similar to intelligence quotient (IQ) in the respect that we all have it to
some degree. Both emotional intelligence and intelligence quotient have standardized tests to evaluate
your EQ and IQ respectively. Even though one s IQ can change over time, you are likely remain in the
same place in the group in which you are compared to. Emotional intelligence however is something
you can increase (or decrease) over time. An EQ test can help you understand your strengths and
weaknesses and how to improve upon your areas of opportunity. Emotionally intelligent people have a
certain way of thinking, feeling, and behaving. They re naturally confident. They bring out the best in
others. When you re with someone who s highly emotionally intelligent, you feel like he or she is
completely interested in you. He or she isn t distracted when talking to you; you get his or her full
attention (Stein). The cornerstone to

Particles Lab
Pressure vs. Number of Particles Lab Report
Introduction: The experiment deals with many different parts and one of them is gas, a substance that
moves freel through the air and their particles are stread farther away from each other when compared
to the other states of matter. The difference between the states of matter are how close the particles are
to each other. The experiment was completed to find how pressure affects the number of particles.
Pressure, which is the force exerted on an object by physical contact, was used to apply force into the
pressure sensor. A pressure sensor measures the amount of force can object has and this will tell us
how the pressure is affecting the number of particles. Also the volume, of the pressure ... Show more

When the pressure was added to the pressure sensor, it increased the particles speed and added more

temperature increases, the particles sped up and this created greater force during the collisions. Also
the graph was supposed to start at zero however the y intercept was off by a little because of an error
that occurred in the lab. But overall, the graph expressed what was expected and shows a linear

Nicolas Poussin Essay
Nicolas Poussin
1594 1665
Pictori philosopho (Blunt, 3), Painter philosopher was a name given to Nicolas Poussin by Serous d
Agincourt in 1782. Agincourt later found out that the name Pictori philosopho had already been given
to the German artist Anton Raphael Mengas.
Nicolas Poussin was born in 1594 in the town of Les Andelys on the Seine. He came from a nobel
family that was ruined by religious wars according to Giovanni Pietro Bellori. No actual proof of this
has been established his father Jean Poussin was said to have had some descent of the hierarchy. His
mother was the daughter of an alderman. His father served in the military under the command of

At the age of thirty Poussin left Paris for Rome, Italy.
Poussin arrived at his destination in March of 1624. During his journey he made a short stop in
Venice. In Rome Poussin met painter Marcello Sacchetti on of the active patrons of the arts in Rome.
During his first couple of years in Rome Poussin stayed in various homes. In 1629 he took shelter with
Jacques Dughet, a French cook, and his family. Jacques and his family took care of Poussin when he
suffered from what is called mal di Francia. (Blunt, 55) After several years were spent recovering from
his illness Jacques gave his daughter Anne Marie to Poussin in marriage. The ceremony toke place on
September 1, 1630 in the city of Lucina. Jacques Dughet had two sons named Gaspard and Jean, who
were artist and worked under the supervision of their brother in law. Gaspard was so inspired by
Poussin that in the later years of his life he took the name of his great teacher Gaspard Poussin.
During this time Poussin kept company with many painters who he was to keep a close friend ship for
many years. He lived close to many painters and sculptors of the time. Among these was Jean
Lemaire, who later worked with Poussin on the execution of the Long Gallery in the Louvre from
1640 to 1642. While in Rome Poussin dressed as a Frenchman, until hostile Romans attacked him due
to the anti French sentiment at the time. He saved his

Dialogue And Authentic Dialogue
Authentic Dialogue When it comes to communicating with people, I do not have a hard time.
Although, this past fall semester I was put to the test. There was a student in our Dialogue and
Personality class that was, difficult. This caused the dynamic in class to be very off putting and hard to
communicate in. He was angry, negative and wasn t afraid to bring the class down with his negative
attitude. This is one example out of many how dialogue can be hard when you are dealing with
conflict. That being said, I would love to talk more about how conflict in dialogue can effect
conversation, how I have experienced authentic dialogue, as well as breaking down if authentic
dialogue was supported within my experiences. To begin, I would like to talk a little bit more about
conflict.
Conflict can be seen in many different situations in dialogue. It is important to be aware of signs so
that you can handle the situation properly. When there is conflict in dialogue it can hinder any
progress to be made within the conversation. Some things that may hinder dialogue are close minded
people, judgment, lack of empathy and people being attacked and blamed. All of these things our class
experienced with our former classmate. I remember the first class hoping he would drop because the
class environment was that uncomfortable. I think that shows that even how well you try to facilitate
or deal with conflict, sometimes its extremely difficult to find a resolution. Granted, our professor did

Essay about Compare and Contrast Religion in Two Works
Religion is sold to the masses daily. In 1925 it was sold to the masses in the form of printed material
by Bruce Barton. In 1960 it was sold to the masses by way of a film entitled Elmer Gantry. Bruce
Barton sold a different idea of Jesus than most people were familiar with. In the film, Elmer Gantry
sold religion with bravado and arrogance which was not the typical way of selling religion either. In
each work, religion is being sold but Elmer is a more exciting and relatable character than Bruce
Barton s Jesus. Elmer Gantry sold religion in a similar way to Barton s Jesus but did so in a more
relatable manner. In both works, each character sought out to sell religion. Although the major
characters in each work were selling ... Show more content on Helpwriting.net ...
Although, Jesus saved people because it was a duty imposed upon him by his father. Elmer Gantry
however, had a different reason for preaching the word of the lord. Elmer found it convenient to
preach about the lord when he knew he could get something in return. He had no ulterior motive than
to use the words in the Bible to his advantage to gain something for him. In the film, Elmer sees a
good thing in Sister Sharon Falconer s evangelical troupe and cons his way into her tent tabernacle,
her graces, and her heart (Weiler, 1960). Elmer used the Bible to get closer to and ultimately to win
over Sister Sharon. Elmer Gantry was an opportunistic character. He used religion when it could
benefit him. Not only was Gantry opportunistic about religion, but was opportunistic when it came to
sex and alcohol as well (Bluestone, 1961). Many people can relate to a character such as Elmer
Gantry. When the opportunity arises for something to go in his favor, he grabs the opportunity. Many
people may feel the same way today. If an opportunity arises to get something for yourself, you will.
Elmer is an ordinary person with ordinary impulses. He sees Sister Sharon as something he desires
and works his way into her inner circle to be closer to her. An ordinary man would do something
similar to be closer to a woman he finds attractive. Both characters had very different reasons for
selling religion, but both were extremely successful.

The Romantic American Male in Cooper’s Last of the...
Masculinity of the Romantic American Male in Cooper s Last of the Mohicans and Irving s Legend of
Sleepy Hollow
James Fenimore Cooper s Last of the Mohicans and Washington Irving s Legend of Sleepy Hollow are
valuable examples of literary heavyweights of the Romantic era, but in addition, can also be used to
chart sociological changes within the male gender during pre Romantic and Romantic years.
But because neither Cooper nor Irving s works should be distanced from their cultural backdrops
when considering the socially reflective nature of their work, exploring basic historical conditions
surrounding the changing concepts of masculinity can serve as a useful move.
Masculinity is primarily a social ... Show more content on Helpwriting.net ...
According to Rotundo, the ideal man was one who ruled as head of his household, believed in the
crucial concept of duty, and upheld stern Puritan religious of morality and social behavior (as the
pleasant, mild mannered, devoted man of the community ). This good man image seems to
coincidentally parallel the English patriarchal gentleman of the seventeenth century.
Social and economic changes around 1820 to 1860 stimulated the Commercial Period, which
produced two distinct spheres of influence: the business world of men and the private domestic world
of women (Grace 10). Instead of the home being a means of production, industry s influence was
being felt for the first time, and with its arrival, men became connected to the public realm more
clearly, and conversely, women to the private realm (10).
But some of the changes also stemmed from politics of the day. At the time of the Revolutionary War,
political upheaval pitched American sons against American fathers, and England s social influence
over her angered citizens began to wane (Manhood 18). Michael Kimmel, author of Manhood in
America: A Cultural History, comments on fledgling America s disenchantment with British culture:
The American colonies had few noblemen, like Sir William Randolph, but they had plenty of
substitutes, from upper class political elites to Dutch landed gentry [...]. There was a little tension

“What Role Do Minor Parties Perform in the Australian...
The role of the minor parties within the Australian political system can be as their title suggests, quite
minor. On the other hand, a minor party can influence the political proceedings of this country in more
ways than one. Minor parties can highlight socio economic problems that quite often fly under the
radar of the larger political parties, or simply can broaden electoral debate. Often spawning from a
social, or in the case of the Greens, an environmental movement, minor parties tend to only last
through one election, due to a lack of support and relevance in the mainstream social fabric. The
minor parties can become crucial players come election time, due to Australia s law of Compulsory
preferential voting. Jaensch (1983, p.21) ... Show more content on Helpwriting.net ...
For example, if the Federal Liberal party needed preferences in marginal seats in Tasmania, a deal
could be negotiated with the Greens, which could see protection of the old growth forest areas of the
state. In reality this can backfire, as it did for the Labor party during the 2004 Federal election. Latham
and the Labor party stated they would protect the Old grow forest areas from logging practices, in an
attempt to win votes. This lead to a revolt by the unions, and those involved in the logging industry,
claiming that Labor had abandoned them.
The second rule that Sartori created was, A party qualifies for relevance whenever its existence, or
appearance, affects the tactics of party competition and particularly when it alters the direction of the
competition by determining a switch from centripetal to centrifugal competition either leftward,
rightward, or in both directions of the governing oriented parties (1983, p.19). This rule further
highlights the importance of minor party preferences, and the role they have in the Australian system.
As an extension of Sartori s two criteria for minor party relevance, a third rule is often highlighted due
to Australia s application of preferential voting on the electoral system. This third rule as quoted by
Jaensch (1983, p.21) states that A minor party can be discounted as irrelevant whenever its preferences

Analysis Of The Painting Moving Day
The painting Moving day from 1967 seems to be about a black family who is moving to another
country. When a person who is different moves to another place, with other people, it s harder to
become a part of the people who is already living there. That s because they don t know anybody at
all. In this painting the black kids have a cat, and the white kids have a dog. I think that is a detail with
a lot of meaning. The cat and the dog are different, and they sometimes don t like each other because
of that. Dogs and cats can be friends too, but sometimes the dog can be (avstøtende), and not accept
the cats for who they are. That is exactly the same as black and white people do to each other.

Music of the South
As you are driving down the road of the countryside of the American South, you see the dry fields of
grass spread for miles. You turn on the radio and all you hear is country music or the blues. This is the
culture and music of the South. The South has two types of music, country music of the whites and the
blues of the blacks. Due to the isolation and conservatism of the South, the South had their own forms
of rural music. First, we look at the blues and how grief can become a form of popular music. Great
blues singers include Ma Rainey, Bessie Smith, B.B. King, and Muddy Waters. Secondly, we look at
the origins of country music. Examples of great country singers include Dolly Parton, Johnny Cash,
Loretta Lynn, and Randy Travis. ... Show more content on Helpwriting.net ...

Students With Autism Spectrum Disorder
Assignment 2
Part A.
Classroom engagement is always a key issue to rookie teachers. There are various types of learner
exist within a classroom and they each need to be treated differently. As teachers, we need to raise our
awareness of the attributes, competencies and challenges for each of those different learner types in
order to engage them in the class. This portfolio will introduce Autism Spectrum Disorder, Hearing
Impairment, Gifted and Talented Students and Challenging Behaviour in the classroom as example of
various learner types and discuss learning competencies and challenges, and then provide possible
engagement strategies for each of those learner types. Part B of this portfolio will discuss four
teaching strategies that are likely to engage students with autism spectrum disorder.
Autism Spectrum Disorder
Autism Spectrum Disorder (ASD) is a developmental condition that will impair individuals
communication ability and social interaction with others. Individuals with autism spectrum disorder
will more lilely to have restricted and repetitive behaviour or interest (Dawson Osterling, 1997).
Dawson Osterling then further added that students with autism spectrum disorder are often more
passive and more likely to be disengaged than their peers. Even when engaged, those students often
engage to object rather than people or surrounding enviroment (eg. class). This means students with
autism spectrum disorder will have significantly less interaction with

Metro Transit And The Better Bus Stop Engagement Essay
Berry Farrington of Metro Transit and the Better Bus Stop Engagement
Context
The bus stop is the front porch of the Metro Transit system, said Berry Farrington, senior planner at
Metro Transit and the Better Bus Stops (BBS) project manager. It is certainly where many
Minnesotans start their daily travels. Given the role of the bus stop, Metro Transit believes it is
necessary to make these places safe, secure, and comfortable for their riders. The BBS engagement
process began in March 2016 in an effort to not only add and improve bus shelters across Metro
Transit s service area but to engage with its riders in order to develop better parameters and guidelines
for future bus stop investments. The process will end in March 2017.
The funding for the BBS engagement comes from a $3.26 million dollar Ladders of Opportunity
Grant from the Federal Transit Administration for bus stop improvements that Metro Transit won in
late 2014. Besides improving bus shelters and stops, Metro Transit also saw this grant as an
opportunity to work towards the Metropolitan Council regional government s Thrive MSP 2040 plan
objective of raising awareness of the complex interdependencies of income, race, place and
opportunity and to challenge both itself and others to think regionally and act equitably for a better
region for all.
The BBS process is also aimed at addressing Metro Transit s negative press from failing to
appropriately balance its bus stop investments across its ridership. A

Technology’s Detrimental Effects on Communication
Recent advancements in technology, especially social networking, have severely hindered the
communication skills of today s society by promoting edited, impersonal and staged communication.
Today s society places a high value on websites such as Facebook, MySpace and Twitter; so much so,
that it is startling when someone does not own a profile on a social network. The dependency on
technological communication continues to reduce the ability of today s generation to read body
language, facial expressions, and to clearly vocalize what is important to them. Technology has
changed drastically in the past 50 years, and continues to grow and morph every day. Today s youth

Although technology was not an option to Baca, he realized that words do not stem from the physical
world. Language does not come from books, formulas, or from a glowing white screen. With
technology thrown into the mix, communication can become anonymous, edited, and thought out
before it is written down/typed up. Written, typed, and texted communication skills are vastly different
from the communication skills needed to hold a conversation in the flesh. It requires a level of grace
and mindfulness to communicate in the real world, especially when it comes to arguments and matters
of great importance. (Such as proposing to your significant other, or a job interview.) When you type a
sentence on a screen, you have the option to reread it, edit it, add to it, and even delete it. Typed
communication allows for the user to edit and censor their thoughts before they are sent out in a
message/post/e mail/etc. While this skill is useful for writing a paper and for creating a resume, it does
not translate well to genuine conversation. Since many people rely on spell check and the delete
button, it does not reinforce the skills that would allow them to catch those mistakes of their own
accord. A lack of mental filtering skills can lead an individual to misspeak and say something
embarrassing, insulting and even cruel in face to face

Yoga And The Yoga Community
Yoga, being one of the most effective self help approaches to life and living, invites me to help ensure
that it is being integrated with maximum wisdom related to breathing. This page is about supporting
and educating the Yoga community in its quest for superior knowledge and effectiveness about proper
breathing. It is our responsibility to alert everyone about this as there are probably millions of students
and teachers with an unclear or distorted idea of healthy breathing.
Nowadays, you can take weekend courses and call yourself a teacher. Many teachers are then first to
defend their breathing development proficiency but alas, last to prove it. One needs to experience
good breathing to teach it.
From a Breathing Times subscriber:
Breath is the unifying principal of the three systems mechanics, metabolism and mentality. It is the
psychopharmacological link between conscious and unconscious states. Each of the three mechanical
aspects of breath has specific neurotransmission function. Any technique(s) that emphasize(s) one
aspect of the mechanism exclusively will shift neurometabolism accordingly. Yoga practices are
designed for this purpose, stimulating a specific aspect or relationship of aspects. I realize that
contemporary yogis don t always explain it this way,and I think that also leads to misuse and abuse.
Right knowledge is yoga or union. That union is from bringing together all aspects, and applying
iscernment.

Displacement Reactions And The Single Displacement Reaction
Introduction
There are two types of displacement reactions : One type is called the single displacement reaction,
the other is called the double displacement reaction. During a single displacement reaction, an element
substitutes another element in a compound, creating a new element and a new compound as products.
In most single displacement reactions, either a metal replaces another metal, or a non metal replaces
another non metal. The more reactive element takes the place of the less reactive element in the
compound, otherwise the reaction will not occur. The general chemical equations of single
displacement can be: A (metal) + BC → AC + B,
A (non metal) + BC → BA + C. Double displacement reaction is a chemical reaction, where the
positive ions of two different chemical compounds exchange and produce two new compounds. The
products may contain a precipitate. The general chemical equation of double displacement is: AB+ CD
→ AD + CB
The purpose of the lab is to observe the reactants and the products of three chemical reactions and
determine which type of reaction occurs in each chemical reaction.
The solubility tables can be used to help better determine the reaction types. The solubility tables refer
to the solubility of ionic compounds in water. By using the solubility tables, we can know whether a
compound is soluble or insoluble.

The Creation of Federal Laws in Australia
The Creation of Federal Laws in Australia The Commonwealth of Australia was created with the 1901
unification of six recently independent British colonies. All were solidified under the authorship of a
Constitution that same year which predicated the form of government and the legal orientation of the
new nation. (AG, p. 1) In doing so, it also vested the power to create federal laws in Australian
Parliament and, in one sense, gave it exclusive authority to do so. And in this sense, one might render
the interpretation that one advantage Australia has is that it has a written constitution which gives
nearly all the power to make laws for Australia to the Commonwealth Parliament. However, a more
complete understanding of the structure of Australian law suggests that this is a great understatement
and one which does not make full accounting for the roles played either by the states that are
confederated to make up Australia or by the judicial branch of the federal government. A consideration
of the roles played by Parliament, the states and the court systems reveals that in fact, while the power
to legislate laws is entrusted in Australian parliament, this entitlement is only possible through an
interdependence with these other dimensions of the government. First, it is appropriate to examine the
conditions that will have allowed for one to make the blanket statement quoted here above. Namely,
the Australian Government (AG)(2011) documents that Section 51 of the

new cartoons. Betty Boop, Felix the Cat, Bugs Bunny, and of course Mickey Mouse were all popular
cartoons in the 1920s. Mickey Mouse was created by Walt Disney and was arguably the most famous
cartoon character in the 20s. Disney created Mickey Mouse and produced the first full length animated
movie (Schickel). During this time one of the greatest cartoon icons was created by Walt Disney after
his distributor stole his first character and it led to the creation of the most recognizable cartoon
character, Mickey Mouse.
Before Walt Disney s creation of Mickey Mouse he went through some struggles. He did not always
own his own business. He worked with a small group of men, who abandon him (Watts, 25). They
abandon him due to the fact the business was falling apart and running out of money. After being
abandoned, he sought out for work as a cartoon artist for two newspaper companies but was turned
down both times. After job hunting for many months, Disney befriended Iwerks and began working on

Paper Towns Psychology
The book Paper Towns, by John Green, follows the story of a seemingly invisible, love sick boy
named Quentin, on his journey to find the girl he has admired since childhood. Margo is one of the
most popular girls at her school, but is very good at keeping secrets and hiding how she feels inside.
She goes through a major moment of psychological development when her and Quentin discover a
man who has shot himself. This is proven when Quentin recounts a childhood story of her, when he
talks to her for the first time in nine years, and when he truly attempts to empathise with her. At the
beginning of the book, Quentin narrates a story of when he and Margo were nine years old. They had
came across a man who had committed suicide. Margo tried to become a detective, and get all the
information she could about the man they had seen. That night she relayed her new found clues from
the investigation. She was obsessed with the puzzle. After this flashback, Quentin thinks to himself,
Margo always loved mysteries. And in everything that came afterward, I could never stop thinking
that maybe she loved mysteries so much she became one (Green, 8). This quote supports the statement
that Margo is very secretive, because Quentin, who has known her since childhood, knows ... Show

Quentin was constantly looking at her and thinking of her, but acted like a fly on the wall. One night,
Margo needs help with something, so she goes into Quentin s room. It seems like an ordinary day to
Quentin, until, ...just before midnight, when Margo Roth Spiegelman slid open my screenless
bedroom window for the first time since telling me to close it nine years before (Green, 24). The
events that occurred when they were children were so scarring for Margo, she was not able to even
interact with Quentin for almost a decade. This disconnection proves that the psychological changes
Margo went through haunted her enough to change her

Fair Housing Act Case Study
Since the passage of the 1968 Fair Housing Act , there have been legal tools and policies that combat
segregation in housing, overall, promoting more diverse cities. While the Fair Housing Act have been
successful in diversifying formerly all white cities. A new problem is emerging for the people in the
integrated neighborhoods: the return of the great whites. Soon there will be no home for these people.
The mission district of San Francisco , soon will lose all its original dwellers to the high demands of

The first wave of gentrification ended when the dot com bubble burst and the number of Ellis act
evictions plunged. In 2005, however, after new technology companies like Google, Facebook, and
cisco began attracting thousands of high paid employees to the bay, the number of Ellis evictions
tripled.1 In 2013, Ellis evictions grew 175 percent from the year before.1 The companies moved to the
south bay but the city attracted many of these young techies. The techies would live in the city and
end up commuting to the Bay area. During that time more than half of the Ellis evictions were carried
out less than a year after a building had changed hands. Since 2011, 69 percent of the evictions have
occurred n the mission no more than a few blocks from the private tech company s shuttle bus stops.1
The Companies provide shuttle bus service to the city in order to carry their employers in to the bay.
Not only do the buses cause overcrowding of the streets but they also traffic and a trend of living more
than 30 miles from work. The tech workers flooding into the Mission creates the profit motive for
landlords to push people out. Whether the individual tech workers are conscious of it or not, they are
complicit in the process of gentrification. The Google bus protests struck a nerve because they
highlighted how the Tech sector is facilitating the forced displacement of families.

Silas Brady s Narrative
With a snap of the reins and a click of his tongue, Silas Brady coaxed his horse into a faster pace,
causing the tiny yellow house on wheels to lurch and rumble behind him. His instincts detected
potential business as his professional eye admired the quaint scene at the bottom of the hill: a
clapboard house and barn, a small corral, and a large plot of land ready for planting. From his
position, the bright green canopy of an oak peeked above the roofline of the house. Aside from a
single grazing cow, the homestead was quiet and still. A tall, fair haired man of middling years
approached from behind the house to wait at the bottom of the porch steps though the lack of a
customary greeting was glaringly obvious. As a precaution, Silas stopped his wagon a short distance
while he set the brake and adopted a friendly mien knowing that an aggressive manner was
counterproductive for business. Good day to you! The name s Silas Brady, photographer by trade, he
announced. Since I m passing your farm on my way to town, perhaps you d like a picture of your own,
mister . . . . The screech of a screened door interrupted Silas, and a woman stepped out of the shadows.
Nodding in acknowledgment, he continued his pitch. Maybe one of you and the missus? I can t ...

while they gathered their six year old son and daughter this he knew in his conversation with Karl

faces before meeting Karl s bleak look. Motioning toward the designated chairs, Silas added gently,

Mir34a Essay
MiR34a inhibits Aβ fibril uptake and autophagy flux
Regarding the compromised capability of Aβ clearance in AD, we suspected that miR34a may be an
indispensable player. To validate this, Aβ42 fibrils were prepared from Aβ42 monomer ex vivo and
used to treat glioma A172 and U251 cells. Aβ uptake was blocked by both transient transfection of a
miR34a mimic and stable miR34a overexpression (Supplementary Fig. 1B, Fig. 1A), but enhanced by
anti miR34a (Fig. 1B), indicating that miR34a suppressed Aβ phagocytosis in these cells. The fact that
enhanced and decreased levels of miR34a correlated with corresponding changes in intracellular Aβ
supports the notion that miR34a may slow down Aβ clearance by affecting its Aβ uptake and
degradation. ... Show more content on Helpwriting.net ...
Since mature autophagosome fuse with lysosomes to become autolysosome in the very late stage of
autophagy, we asked whether miR34a could retard autophagosome lysosome fusion by examining the
co localization of EGFP LC3 and LysoTracker Red, a specific dye for live cell lysosome labeling [47].
As expected, EGFP LC3 puncta were dramatically reinforced in miR34a mimic transfected cells but
did not co localize with LysoTracker Red. This was acting similar to cells treated with bafilomycin A1
(Baf), which blocked autophagosome lysosome fusion. In contrast, E64D treatment induced a
remarkable increased EGFP LC3 puncta, which were well co localized with LysoTracker Red (Fig.
1D). Since lysosome associated membrane glycoprotein 2 (LAMP2) is also critical for autophagosome
lysosome fusion [48], we examined the effect of miR34a on LAMP2 expression using WB analysis.
MiR34a caused an unexpected increase of LAMP2 (Supplementary Fig. 1C), suggesting that the
mechanism by which miR34a blocked autophagosome lysosome fusion was not due to LAMP2. As
LAMP2 is mostly important for chaperone mediated autophagy, its increase might be a compensation
for the decrease in macroautophagy. In summary, the above results reinforce that miR34a
compromised Aβ degradation by, at least, blocking autolysosome formation.

Analysis Of The Oresteia
In the Oresteia, there is a deep relationship between characters which is necessary to understand the
role of suffering in Greek tragedy. The most profound form of this relationship is between a mortal
and an immortal. Suffering of the mortal spurns a yearning on the part of the immortal to give
assistance or guidance to the mortal, like a mother helps her child in pain. Suffering on the part of the
mortal is a vital piece to stimulate emotional response from the audience. The same, to a lesser extent,
can is also said of many other inter mortal relationships. The passions felt by the reader because of
such a relationship brings an emotional cleansing in response to the suffering of the mortal. The role
of suffering in Greek tragedy is to enable the audience to empathize with the characters suffering
through their relationships.
Relationship is defined as any two characters having any relations with each other for an end. The
relationship between Orestes and Electra is that of love and use; Electra has a love for her brother, but
also wants to use him to kill her mother. That use and love portrays to the audience a connection that
allows them to more fully understand and relate to the relationship. This happens because of the past
experiences the audience had; all men have felt both love and the want for revenge.
The harnessing of emotion in Greek theater is the most powerful tool a playwright had. To be able to
manipulate the audience and their emotions is the highest

The Endings Of Sophocles Antigone
Although they are versions of the same play, the endings of Sophocles Antigone and Anne Carson s
Antigonick convey contrasting ideas of the power of fate, and its role in tragedy. In Antigone, fate is
described as an inevitable, insurmountable aspect of life that cannot be avoided. On the other hand,
Antigonick shows fate to be a malleable product which can be controlled by the decisions individuals
take in their lives. Through the role of the chorus, the addition of Nick, and the catharses of the
characters, Carson shows that fate is something which we can master. In both texts, the responsibility
of the chorus is to comment upon the play s proceedings, and to make key observations about the
characters actions. As Antigone is being paraded to her burial in Antigone, the chorus states, The
power of fate is a wonder,/ dark, terrible wonder / neither wealth nor armies/ towered walls nor ships/
black hulls lashed by the salt/ can save us from that force. (1045 1050) It emphasizes that fate is very
powerful, and that it is not within humans capacity to alter. Thus, Antigone and Creon are never
capable of influencing their own lives; they are not responsible for the tragedy that ensues, because
fate would have seen to it anyway. In contrast, the chorus in Antigonick says that it likes to find
precedents, pointing out that We re not at a loss how to think about this/ We re not without guidance.
The chorus states that Creon can avoid making the same mistakes

Essay on Power in of Mice and Men
Power
By Maria Liddy
The theme of power is prevalent throughout the novel Of Mice and Men. Steinbeck uses various
methods and techniques to establish the dynamics of power on the ranch. In the first extract, George
and Lennie are in the brush and we get a strong sense of George s parental control over Lennie, but it
also shows how Lennie s physical stature gives him a degree of power over George. In extract two we
meet Curley for the first time, and his authority over the ranch workers is clearly asserted through the
various ways in which Steinbeck describes him. And finally, in extract three, we see the first fight of
the novel. The fight is very diverse in how it portrays power. At different stages in the fight some

This is shown through Lennie s very simple language.
..But it didn t do no good
Steinbeck also uses many question marks to show how Lennie is constantly asking questions and
therefore constantly seeking guidance from George, and again, that s what gives George the edge over
Lennie.
When discussion moves to the bus tickets and work cards Lennie realises that he doesn t have his. He
looked down at the ground in despair which shows that he is ashamed and perhaps a little frightened
of George s reaction, but Steinbeck then goes on to say that George took responsibility for both of the
work cards, knowing that Lennie could not be trusted. This, again, demonstrates the balance of power
and shows that both George and Lennie are aware of how much power the other possesses.
After looking for his work card in his pocket, Lennie take out a mouse to which George replies
sharply.
What d you take outta that pocket?
George s tone is accusatory which shows that he is astute. He then continues to bombard Lennie with
questions, which gives Lennie the chance to demonstrate another type of power his cunning. He
attempts to deceive George.
Ain t a thing in my pocket
Although childlike, this sentence shows that Lennie s mind is able to think of plans. How would he
keep the mouse? By attempting to deceive George. Eventually, George s patience wears thin and he
exclaims Give it here! shouting at Lennie like a parent.
Throughout the extract

The Benefits Of A Single Stage To Orbit
A Single Stage to Orbit (SSTO) spaceplane would have many advantages over current methods of
getting people and equipment into space. Chief among these is major cost reduction. SSTOs would be
fully reusable, being that they are meant to send people to space and bring them back. A fully reusable
single stage space plane would have an advantage over a fully reusable rocket booster, similar to what
was used on the shuttle, and what is being used as the first stage on SpaceX s Falcon 9. In fact, the
Falcon 9 has already started to pay off to SpaceX. According to an article from SpaceNews.com, the
President of SpaceX, Gwynne Shotwell, reported that a rocket launched in March which had a reused
first stage was substantially less than half ... Show more content on Helpwriting.net ...
There is a challenge for TSTOs to overcome: air friction being directed on the vehicle from its booster,
while separating. The paper Aeroheating Characteristics for a Two Stage To Orbit Concept During
Separation at Mach 6 , investigates the issue. This study looked into the aeroheating on two crafts
when separating so one can make it into space, while the other, the first stage, returns to land. The
study found that in the small scale test conducted that there were not much negative effects of heating
from friction on one craft to the other. This would appear to be a positive development for the TSTO
community as it is possible to get a craft into space by attaching it to another.
As is seen in Investigation of Two Stage to Orbit Airbreathing Launch Vehicle Configurations , there
are many possible configurations for a TSTO. The kind this paper explored most was the booster craft
had the orbiter attacked on top of it, similar to how NASA transported the Shuttles on top of a
modified 747. Aeroheating Characteristics for a Two Stage To Orbit Concept During Separation at
Mach 6 shows the characteristics of aeroheating during the separation of the top bottom configuration.
The paper showed that it is possible for the top bottom configuration, and that the aerodynamic forces
from the two vehicles won t negatively affect each other.
TSTO launches have happened before, albeit

  • 2. Organizational Development Department And Is Someone That... When selecting my topic for this research paper I reached out to a co worker of mine, Michelle Choate. Michelle works in our Organizational Development department and is someone that I have taken communications classes from at St. Luke s. For many years I consistently had employment evaluations that suggested that I had significant opportunity to improve my communication skills. Through the courses taught by Michelle, I was able to discover some characteristics about myself and implement her teachings into my life. I did not realize it prior to writing this paper but she has been a valuable asset to increasing my emotional intelligence. When selecting my topic I wanted to know, from her perspective, what topics in communication where ... Show more content on Helpwriting.net ... Characteristics of self regulation include; trustworthiness, integrity and openness to change. Internal motivation is your passion to complete a project or a goal that is beyond financial compensation. Empathy is the ability to understand the emotions of others, and treating them in a way that is compatible with their emotional reactions. And finally, social skills include one s ability to manage and build social and professional networks, be persuasive, build teams and rapport. Who has emotional intelligence? Emotional intelligence (EQ) is similar to intelligence quotient (IQ) in the respect that we all have it to some degree. Both emotional intelligence and intelligence quotient have standardized tests to evaluate your EQ and IQ respectively. Even though one s IQ can change over time, you are likely remain in the same place in the group in which you are compared to. Emotional intelligence however is something you can increase (or decrease) over time. An EQ test can help you understand your strengths and weaknesses and how to improve upon your areas of opportunity. Emotionally intelligent people have a certain way of thinking, feeling, and behaving. They re naturally confident. They bring out the best in others. When you re with someone who s highly emotionally intelligent, you feel like he or she is completely interested in you. He or she isn t distracted when talking to you; you get his or her full attention (Stein). The cornerstone to ... Get more on HelpWriting.net ...
  • 3. Particles Lab Pressure vs. Number of Particles Lab Report Introduction: The experiment deals with many different parts and one of them is gas, a substance that moves freel through the air and their particles are stread farther away from each other when compared to the other states of matter. The difference between the states of matter are how close the particles are to each other. The experiment was completed to find how pressure affects the number of particles. Pressure, which is the force exerted on an object by physical contact, was used to apply force into the pressure sensor. A pressure sensor measures the amount of force can object has and this will tell us how the pressure is affecting the number of particles. Also the volume, of the pressure ... Also the object that withheld the pressure had a constant volume, meaning the shape did not change Then when the student increased the pressure the greater number of particles. When the air temperature increases, the particles sped up and this created greater force during the collisions. Also the graph was supposed to start at zero however the y intercept was off by a little because of an error that occurred in the lab. Agincourt later found out that the name Pictori philosopho had already been given to the German artist Anton Raphael Mengas. Nicolas Poussin was born in 1594 in the town of Les Andelys on the Seine. He came from a nobel family that was ruined by religious wars according to Giovanni Pietro Bellori. At the age of thirty Poussin left Paris for Rome, Italy. Poussin arrived at his destination in March of 1624. During his journey he made a short stop in Venice. In Rome Poussin met painter Marcello Sacchetti on of the active patrons of the arts in Rome. During his first couple of years in Rome Poussin stayed in various homes. In 1629 he took shelter with Jacques Dughet, a French cook, and his family. Jacques and his family took care of Poussin when he suffered from what is called mal di Francia. (Blunt, 55) After several years were spent recovering from his illness Jacques gave his daughter Anne Marie to Poussin in marriage. The ceremony toke place on September 1, 1630 in the city of Lucina. Jacques Dughet had two sons named Gaspard and Jean, who were artist and worked under the supervision of their brother in law. Gaspard was so inspired by Poussin that in the later years of his life he took the name of his great teacher Gaspard Poussin. During this time Poussin kept company with many painters who he was to keep a close friend ship for many years. He lived close to many painters and sculptors of the time. Among these was Jean Lemaire, who later worked with Poussin on the execution of the Long Gallery in the Louvre from 1640 to 1642. • 5. Dialogue And Authentic Dialogue Authentic Dialogue When it comes to communicating with people, I do not have a hard time. Although, this past fall semester I was put to the test. There was a student in our Dialogue and Personality class that was, difficult. This caused the dynamic in class to be very off putting and hard to communicate in. He was angry, negative and wasn t afraid to bring the class down with his negative attitude. This is one example out of many how dialogue can be hard when you are dealing with conflict. That being said, I would love to talk more about how conflict in dialogue can effect conversation, how I have experienced authentic dialogue, as well as breaking down if authentic dialogue was supported within my experiences. To begin, I would like to talk a little bit more about conflict. Conflict can be seen in many different situations in dialogue. It is important to be aware of signs so that you can handle the situation properly. When there is conflict in dialogue it can hinder any progress to be made within the conversation. Some things that may hinder dialogue are close minded people, judgment, lack of empathy and people being attacked and blamed. All of these things our class experienced with our former classmate. I remember the first class hoping he would drop because the class environment was that uncomfortable. I think that shows that even how well you try to facilitate or deal with conflict, sometimes its extremely difficult to find a resolution. Granted, our professor did a ... • 6. Essay about Compare and Contrast Religion in Two Works Religion is sold to the masses daily. In 1925 it was sold to the masses in the form of printed material by Bruce Barton. In 1960 it was sold to the masses by way of a film entitled Elmer Gantry. Bruce Barton sold a different idea of Jesus than most people were familiar with. In the film, Elmer Gantry sold religion with bravado and arrogance which was not the typical way of selling religion either. In each work, religion is being sold but Elmer is a more exciting and relatable character than Bruce Barton s Jesus. Elmer Gantry sold religion in a similar way to Barton s Jesus but did so in a more relatable manner. In both works, each character sought out to sell religion. Although the major characters in each work were selling ... Show more content on Helpwriting.net ... Although, Jesus saved people because it was a duty imposed upon him by his father. Elmer Gantry however, had a different reason for preaching the word of the lord. Elmer found it convenient to preach about the lord when he knew he could get something in return. He had no ulterior motive than to use the words in the Bible to his advantage to gain something for him. In the film, Elmer sees a good thing in Sister Sharon Falconer s evangelical troupe and cons his way into her tent tabernacle, her graces, and her heart (Weiler, 1960). Elmer used the Bible to get closer to and ultimately to win over Sister Sharon. Elmer Gantry was an opportunistic character. He used religion when it could benefit him. Not only was Gantry opportunistic about religion, but was opportunistic when it came to sex and alcohol as well (Bluestone, 1961). Many people can relate to a character such as Elmer Gantry. When the opportunity arises for something to go in his favor, he grabs the opportunity. Many people may feel the same way today. If an opportunity arises to get something for yourself, you will. Elmer is an ordinary person with ordinary impulses. He sees Sister Sharon as something he desires and works his way into her inner circle to be closer to her. An ordinary man would do something similar to be closer to a woman he finds attractive. Both characters had very different reasons for selling religion, but both were extremely successful. ... Masculinity of the Romantic American Male in Cooper s Last of the Mohicans and Irving s Legend of Sleepy Hollow James Fenimore Cooper s Last of the Mohicans and Washington Irving s Legend of Sleepy Hollow are valuable examples of literary heavyweights of the Romantic era, but in addition, can also be used to chart sociological changes within the male gender during pre Romantic and Romantic years. But because neither Cooper nor Irving s works should be distanced from their cultural backdrops when considering the socially reflective nature of their work, exploring basic historical conditions surrounding the changing concepts of masculinity can serve as a useful move. Masculinity is primarily a social ... Show more content on Helpwriting.net ... According to Rotundo, the ideal man was one who ruled as head of his household, believed in the crucial concept of duty, and upheld stern Puritan religious of morality and social behavior (as the pleasant, mild mannered, devoted man of the community ). This good man image seems to coincidentally parallel the English patriarchal gentleman of the seventeenth century. Social and economic changes around 1820 to 1860 stimulated the Commercial Period, which produced two distinct spheres of influence: the business world of men and the private domestic world of women (Grace 10). Instead of the home being a means of production, industry s influence was being felt for the first time, and with its arrival, men became connected to the public realm more clearly, and conversely, women to the private realm (10). But some of the changes also stemmed from politics of the day. At the time of the Revolutionary War, political upheaval pitched American sons against American fathers, and England s social influence over her angered citizens began to wane (Manhood 18). • 9. “What Role Do Minor Parties Perform in the Australian... The role of the minor parties within the Australian political system can be as their title suggests, quite minor. On the other hand, a minor party can influence the political proceedings of this country in more ways than one. Minor parties can highlight socio economic problems that quite often fly under the radar of the larger political parties, or simply can broaden electoral debate. Often spawning from a social, or in the case of the Greens, an environmental movement, minor parties tend to only last through one election, due to a lack of support and relevance in the mainstream social fabric. The minor parties can become crucial players come election time, due to Australia s law of Compulsory preferential voting. Jaensch (1983, p.21) ... Show more content on Helpwriting.net ... For example, if the Federal Liberal party needed preferences in marginal seats in Tasmania, a deal could be negotiated with the Greens, which could see protection of the old growth forest areas of the state. In reality this can backfire, as it did for the Labor party during the 2004 Federal election. Latham and the Labor party stated they would protect the Old grow forest areas from logging practices, in an attempt to win votes. This lead to a revolt by the unions, and those involved in the logging industry, claiming that Labor had abandoned them. The second rule that Sartori created was, A party qualifies for relevance whenever its existence, or appearance, affects the tactics of party competition and particularly when it alters the direction of the competition by determining a switch from centripetal to centrifugal competition either leftward, rightward, or in both directions of the governing oriented parties (1983, p.19). This rule further highlights the importance of minor party preferences, and the role they have in the Australian system. As an extension of Sartori s two criteria for minor party relevance, a third rule is often highlighted due to Australia s application of preferential voting on the electoral system. This third rule as quoted by Jaensch (1983, p.21) states that A minor party can be discounted as irrelevant whenever its preferences ... Get more on HelpWriting.net ...
  • 10. Analysis Of The Painting Moving Day The painting Moving day from 1967 seems to be about a black family who is moving to another country. When a person who is different moves to another place, with other people, it s harder to become a part of the people who is already living there. That s because they don t know anybody at all. In this painting the black kids have a cat, and the white kids have a dog. I think that is a detail with a lot of meaning. The cat and the dog are different, and they sometimes don t like each other because of that. Dogs and cats can be friends too, but sometimes the dog can be (avstøtende), and not accept the cats for who they are. That is exactly the same as black and white people do to each other. You turn on the radio and all you hear is country music or the blues. This is the culture and music of the South. The South has two types of music, country music of the whites and the blues of the blacks. Due to the isolation and conservatism of the South, the South had their own forms of rural music. First, we look at the blues and how grief can become a form of popular music. Great blues singers include Ma Rainey, Bessie Smith, B.B. King, and Muddy Waters. • 12. Students With Autism Spectrum Disorder Assignment 2 Part A. Classroom engagement is always a key issue to rookie teachers. There are various types of learner exist within a classroom and they each need to be treated differently. As teachers, we need to raise our awareness of the attributes, competencies and challenges for each of those different learner types in order to engage them in the class. This portfolio will introduce Autism Spectrum Disorder, Hearing Impairment, Gifted and Talented Students and Challenging Behaviour in the classroom as example of various learner types and discuss learning competencies and challenges, and then provide possible engagement strategies for each of those learner types. Part B of this portfolio will discuss four teaching strategies that are likely to engage students with autism spectrum disorder. Autism Spectrum Disorder Autism Spectrum Disorder (ASD) is a developmental condition that will impair individuals communication ability and social interaction with others. Individuals with autism spectrum disorder will more lilely to have restricted and repetitive behaviour or interest (Dawson Osterling, 1997). Dawson Osterling then further added that students with autism spectrum disorder are often more passive and more likely to be disengaged than their peers. Even when engaged, those students often engage to object rather than people or surrounding enviroment (eg. class). • 14. Metro Transit And The Better Bus Stop Engagement Essay Berry Farrington of Metro Transit and the Better Bus Stop Engagement Context The bus stop is the front porch of the Metro Transit system, said Berry Farrington, senior planner at Metro Transit and the Better Bus Stops (BBS) project manager. It is certainly where many Minnesotans start their daily travels. Given the role of the bus stop, Metro Transit believes it is necessary to make these places safe, secure, and comfortable for their riders. The BBS engagement process began in March 2016 in an effort to not only add and improve bus shelters across Metro Transit s service area but to engage with its riders in order to develop better parameters and guidelines for future bus stop investments. The process will end in March 2017. The funding for the BBS engagement comes from a $3.26 million dollar Ladders of Opportunity Grant from the Federal Transit Administration for bus stop improvements that Metro Transit won in late 2014. Besides improving bus shelters and stops, Metro Transit also saw this grant as an opportunity to work towards the Metropolitan Council regional government s Thrive MSP 2040 plan objective of raising awareness of the complex interdependencies of income, race, place and opportunity and to challenge both itself and others to think regionally and act equitably for a better region for all. The BBS process is also aimed at addressing Metro Transit s negative press from failing to appropriately balance its bus stop investments across its ridership. A ... Get more on HelpWriting.net ...

Technology’s Detrimental Effects on Communication Recent advancements in technology, especially social networking, have severely hindered the communication skills of today s society by promoting edited, impersonal and staged communication. Today s society places a high value on websites such as Facebook, MySpace and Twitter; so much so, that it is startling when someone does not own a profile on a social network. The dependency on technological communication continues to reduce the ability of today s generation to read body language, facial expressions, and to clearly vocalize what is important to them. Language does not come from books, formulas, or from a glowing white screen. With technology thrown into the mix, communication can become anonymous, edited, and thought out before it is written down/typed up. Written, typed, and texted communication skills are vastly different from the communication skills needed to hold a conversation in the flesh. It requires a level of grace and mindfulness to communicate in the real world, especially when it comes to arguments and matters of great importance. (Such as proposing to your significant other, or a job interview.) When you type a sentence on a screen, you have the option to reread it, edit it, add to it, and even delete it. Typed communication allows for the user to edit and censor their thoughts before they are sent out in a message/post/e mail/etc. While this skill is useful for writing a paper and for creating a resume, it does not translate well to genuine conversation. Since many people rely on spell check and the delete button, it does not reinforce the skills that would allow them to catch those mistakes of their own accord. • 16. Yoga And The Yoga Community Yoga, being one of the most effective self help approaches to life and living, invites me to help ensure that it is being integrated with maximum wisdom related to breathing. This page is about supporting and educating the Yoga community in its quest for superior knowledge and effectiveness about proper breathing. It is our responsibility to alert everyone about this as there are probably millions of students and teachers with an unclear or distorted idea of healthy breathing. Nowadays, you can take weekend courses and call yourself a teacher. Many teachers are then first to defend their breathing development proficiency but alas, last to prove it. One needs to experience good breathing to teach it. From a Breathing Times subscriber: Breath is the unifying principal of the three systems mechanics, metabolism and mentality. It is the psychopharmacological link between conscious and unconscious states. Each of the three mechanical aspects of breath has specific neurotransmission function. • 18. Displacement Reactions And The Single Displacement Reaction Introduction There are two types of displacement reactions : One type is called the single displacement reaction, the other is called the double displacement reaction. During a single displacement reaction, an element substitutes another element in a compound, creating a new element and a new compound as products. In most single displacement reactions, either a metal replaces another metal, or a non metal replaces another non metal. The more reactive element takes the place of the less reactive element in the compound, otherwise the reaction will not occur. The general chemical equations of single displacement can be: A (metal) + BC → AC + B, A (non metal) + BC → BA + C. Double displacement reaction is a chemical reaction, where the positive ions of two different chemical compounds exchange and produce two new compounds. The products may contain a precipitate. The general chemical equation of double displacement is: AB+ CD → AD + CB The purpose of the lab is to observe the reactants and the products of three chemical reactions and determine which type of reaction occurs in each chemical reaction. The solubility tables can be used to help better determine the reaction types. The solubility tables refer to the solubility of ionic compounds in water. By using the solubility tables, we can know whether a compound is soluble or insoluble. • 19. The Creation of Federal Laws in Australia The Creation of Federal Laws in Australia The Commonwealth of Australia was created with the 1901 unification of six recently independent British colonies. All were solidified under the authorship of a Constitution that same year which predicated the form of government and the legal orientation of the new nation. (AG, p. 1) In doing so, it also vested the power to create federal laws in Australian Parliament and, in one sense, gave it exclusive authority to do so. And in this sense, one might render the interpretation that one advantage Australia has is that it has a written constitution which gives nearly all the power to make laws for Australia to the Commonwealth Parliament. However, a more complete understanding of the structure of Australian law suggests that this is a great understatement and one which does not make full accounting for the roles played either by the states that are confederated to make up Australia or by the judicial branch of the federal government. A consideration of the roles played by Parliament, the states and the court systems reveals that in fact, while the power to legislate laws is entrusted in Australian parliament, this entitlement is only possible through an interdependence with these other dimensions of the government. First, it is appropriate to examine the conditions that will have allowed for one to make the blanket statement quoted here above. Namely, the Australian Government (AG)(2011) documents that Section 51 of the ... Get more on HelpWriting.net ...

Paper Towns Psychology The book Paper Towns, by John Green, follows the story of a seemingly invisible, love sick boy named Quentin, on his journey to find the girl he has admired since childhood. Margo is one of the most popular girls at her school, but is very good at keeping secrets and hiding how she feels inside. She goes through a major moment of psychological development when her and Quentin discover a man who has shot himself. This is proven when Quentin recounts a childhood story of her, when he talks to her for the first time in nine years, and when he truly attempts to empathise with her. At the beginning of the book, Quentin narrates a story of when he and Margo were nine years old. They had came across a man who had committed suicide. Margo tried to become a detective, and get all the information she could about the man they had seen. That night she relayed her new found clues from the investigation. She was obsessed with the puzzle. After this flashback, Quentin thinks to himself, Margo always loved mysteries. To validate this, Aβ42 fibrils were prepared from Aβ42 monomer ex vivo and used to treat glioma A172 and U251 cells. Aβ uptake was blocked by both transient transfection of a miR34a mimic and stable miR34a overexpression (Supplementary Fig. 1B, Fig. 1A), but enhanced by anti miR34a (Fig. 1B), indicating that miR34a suppressed Aβ phagocytosis in these cells. The fact that enhanced and decreased levels of miR34a correlated with corresponding changes in intracellular Aβ supports the notion that miR34a may slow down Aβ clearance by affecting its Aβ uptake and degradation. ... Show more content on Helpwriting.net ... Since mature autophagosome fuse with lysosomes to become autolysosome in the very late stage of autophagy, we asked whether miR34a could retard autophagosome lysosome fusion by examining the co localization of EGFP LC3 and LysoTracker Red, a specific dye for live cell lysosome labeling [47]. As expected, EGFP LC3 puncta were dramatically reinforced in miR34a mimic transfected cells but did not co localize with LysoTracker Red. This was acting similar to cells treated with bafilomycin A1 (Baf), which blocked autophagosome lysosome fusion. In contrast, E64D treatment induced a remarkable increased EGFP LC3 puncta, which were well co localized with LysoTracker Red (Fig. 1D). Since lysosome associated membrane glycoprotein 2 (LAMP2) is also critical for autophagosome lysosome fusion [48], we examined the effect of miR34a on LAMP2 expression using WB analysis. MiR34a caused an unexpected increase of LAMP2 (Supplementary Fig. 1C), suggesting that the mechanism by which miR34a blocked autophagosome lysosome fusion was not due to LAMP2. As LAMP2 is mostly important for chaperone mediated autophagy, its increase might be a compensation for the decrease in macroautophagy. In summary, the above results reinforce that miR34a compromised Aβ degradation by, at least, blocking autolysosome formation. ... Get more on HelpWriting.net ...
  • 28. Analysis Of The Oresteia In the Oresteia, there is a deep relationship between characters which is necessary to understand the role of suffering in Greek tragedy. The most profound form of this relationship is between a mortal and an immortal. Suffering of the mortal spurns a yearning on the part of the immortal to give assistance or guidance to the mortal, like a mother helps her child in pain. Suffering on the part of the mortal is a vital piece to stimulate emotional response from the audience. The same, to a lesser extent, can is also said of many other inter mortal relationships. The passions felt by the reader because of such a relationship brings an emotional cleansing in response to the suffering of the mortal. The role of suffering in Greek tragedy is to enable the audience to empathize with the characters suffering through their relationships. Relationship is defined as any two characters having any relations with each other for an end. The relationship between Orestes and Electra is that of love and use; Electra has a love for her brother, but also wants to use him to kill her mother. That use and love portrays to the audience a connection that allows them to more fully understand and relate to the relationship. This happens because of the past experiences the audience had; all men have felt both love and the want for revenge. The harnessing of emotion in Greek theater is the most powerful tool a playwright had. To be able to manipulate the audience and their emotions is the highest ... Get more on HelpWriting.net ...
  • 29. The Endings Of Sophocles Antigone Although they are versions of the same play, the endings of Sophocles Antigone and Anne Carson s Antigonick convey contrasting ideas of the power of fate, and its role in tragedy. In Antigone, fate is described as an inevitable, insurmountable aspect of life that cannot be avoided. On the other hand, Antigonick shows fate to be a malleable product which can be controlled by the decisions individuals take in their lives. Through the role of the chorus, the addition of Nick, and the catharses of the characters, Carson shows that fate is something which we can master. In both texts, the responsibility of the chorus is to comment upon the play s proceedings, and to make key observations about the characters actions. As Antigone is being paraded to her burial in Antigone, the chorus states, The power of fate is a wonder,/ dark, terrible wonder / neither wealth nor armies/ towered walls nor ships/ black hulls lashed by the salt/ can save us from that force. (1045 1050) It emphasizes that fate is very powerful, and that it is not within humans capacity to alter. Thus, Antigone and Creon are never capable of influencing their own lives; they are not responsible for the tragedy that ensues, because fate would have seen to it anyway. In contrast, the chorus in Antigonick says that it likes to find precedents, pointing out that We re not at a loss how to think about this/ We re not without guidance. He looked down at the ground in despair which shows that he is ashamed and perhaps a little frightened of George s reaction, but Steinbeck then goes on to say that George took responsibility for both of the work cards, knowing that Lennie could not be trusted. After looking for his work card in his pocket, Lennie take out a mouse to which George replies sharply. What d you take outta that pocket? George s tone is accusatory which shows that he is astute.
