HIV
1. The HIV (human immunodeficiency virus) is a virus that attacks the
immune system of the human body. If it doesn't get treated it can lead
to AIDS (Acquired Immune Deficiency Syndrome)
2. With the proper medical attention, it can be controlled. People
infected with HIV that receive effective treatment can have a long and
healthy life, and protect their partners.
3. HIV is classified as a retrovirus, and it belongs to the lentivirus
subfamily
HIV REPLICATION
OBJECTIVE
“Demonstrate that aptamer CA15-2 significantly inhibits
HIV-1 replication in producer cells, but not in target cells”.
METHODS
PCR
Is based on the
enzymatic amplification
of genetic material.
In this study it was used
to generate transcription
templates, amplifying the
expression of plasmids
in the aptamers used in
the experiment.
Genetic recombination
Refers to the
rearrangement of DNA
sequences by the
breakage and rejoining of
one to others. In this
study it was used with the
purpose of selecting RNA
aptamers with affinity to
the assembled HIV-1 CA
lattice
METHODS
PAGE
(Polyacrylamide gel
electrophoresis):is a technique
widely used to separate
biological macromolecules,
usually proteins or nucleic acids,
according to their
electrophoretic mobility. In this
study we use 1 of the PAGE
technique methods.
- SDS-PAGE (sodium dodecyl
sulfate polyacrylamide gel
electrophoresis).
EMSA
The gel electrophoresis
mobility shift assay is used to
detect protein complexes with
nucleic acids.
In the study this technique
was used in different ways to
evaluate properties of the CA-
152.

Importantly, due to the cross-linking, these stabilized tubes can
be removed from high salt buffers without disrupting the lattice
and have been shown to maintain interactions with lattice-
binding host factors.

Recent work has suggested that the immature lattice is formed
via recruitment of Gag dimers to the site of assembly to form
hexamers, which then assemble into the immature lattice.

Indeed, we have previously demonstrated that, at the dosage of
aptamer expression plasmid utilized in these experiments,
aptamers, including non-binding controls, can be non-
specifically encapsidated into viral particles
Conclusions
● Experimenting with the structure of the HIV virus using different biological
techniques helps us to know its functioning and how can we apply this
knowledge to create ways to control it more.
● Understanding how the replication of the HIV is structured, we can learn
how to stop it and this way manage to make a step forward in the fight
against this disease that has been affecting this world for a long time.
