Non compartmental
Pharmacokinetics
• Non-compartmental analysis also known as model
independent methods because it does not require the
assumption of specific compartment model.
• Also called SHAM (Shape, Height, Area, Moments)
• This method is based on the assumption that the drugs or
metabolites follow linear kinetics, so this technique can be
applied to any compartment model.
• This method is based on statistical moments theory where it
involves collection of experimental data following a single
dose of drug.
• Non-compartmental analysis (NCA) is a simple and quick
method for evaluating the exposure of a drug. It allows you
to evaluate things like linearity and in vivo exposure.
Compartment model Vs Non
Compartment models
Parameters measured
• Clearance: CL=Dose/AUC
• Mean residence time: MRT=AUMC/AUC
• MRT= is defined as the average amount of time
spent by the drug in the body before being
eliminated.
• Steady state volume of distribution: Vss=MRT*CL
= Dose* AUMC/AUC2
• Half-life: Terminal slope of the natural log of the data
• F% = AUCEV/AUCIV
Areas of the trapezoids
•  Area under the curve,
AUC = Area of one trapezoid
=(1/2)(Cn-1 + Cn)(tn – tn-1)
