Next generation sequencing (NGS) provides higher throughput and lower costs compared to Sanger sequencing. Common NGS platforms include 454, Illumina, SOLiD, Ion Torrent, and PacBio. The Illumina platform uses reversible terminators and bridge amplification to sequence millions of DNA fragments in parallel. 454 pyrosequencing detects pyrophosphate release during nucleotide incorporation. SOLiD uses ligation of fluorescently labeled di-base probes. Ion Torrent detects hydrogen ions released during DNA polymerization. PacBio sequences single molecules in real-time using zero-mode waveguides. NGS costs have dropped dramatically and throughput has increased exponentially over the past decade.
