Dexona (Generic Dexamethasone Tablets) is used to treat
conditions such as arthritis, blood/hormone disorders,
allergic reactions, skin diseases, eye problems, breathing
problems, bowel disorders, cancer, and immune system
disorders. It is also used as a test for an adrenal gland
disorder (Cushing's syndrome).
Dexamethasone belongs to a class of drugs known as
corticosteroids. This medicine decreases your immune
system's response to various diseases to reduce symptoms
such as swelling and allergic-type reactions.

The active ingredient contained in Dexona
tablets is Dexamethasone.
Each Dexona tablet contains 0.5 mg of
Dexamethasone.

Uses of Dexona 0.5mg Tablets:
Allergic States: Control of severe or incapacitating allergic conditions intractable
to adequate trials of conventional treatment in asthma, atopic dermatitis,
contact dermatitis, drug hypersensitivity reactions, perennial or seasonal allergic
rhinitis, and serum sickness.
Dermatologic Diseases: Bullous dermatitis herpetiformis, exfoliative
erythroderma, mycosis fungoides, pemphigus, and severe erythema multiforme
(Stevens-Johnson syndrome).
Endocrine Disorders: Primary or secondary adrenocortical insufficiency
(hydrocortisone or cortisone is the drug of choice; may be used in conjunction
with synthetic mineralocorticoid analogs where applicable; in infancy
mineralocorticoid supplementation is of particular importance), congenital
adrenal hyperplasia, hypercalcemia associated with cancer, and nonsuppurative
thyroiditis.

Uses of Dexona 0.5mg Tablets:
Gastrointestinal Diseases: To tide the patient over a critical period of the disease
in regional enteritis and ulcerative colitis.
Hematologic Disorders: Acquired (autoimmune) hemolytic anemia, congenital
(erythroid) hypoplastic anemia (Diamond-Blackfan anemia), idiopathic
thrombocytopenic purpura in adults, pure red cell aplasia, and selected cases of
secondary thrombocytopenia.
Miscellaneous: Diagnostic testing of adrenocortical hyperfunction, trichinosis
with neurologic or myocardial involvement, tuberculous meningitis with
subarachnoid block or impending block when used with appropriate
antituberculous chemotherapy.
Neoplastic Diseases: For the palliative management of leukemias and
lymphomas.

Uses of Dexona 0.5mg Tablets:
Nervous System: Acute exacerbations of multiple sclerosis, cerebral edema associated
with primary or metastatic brain tumor, craniotomy, or head injury.
Ophthalmic Diseases: Sympathetic ophthalmia, temporal arteritis, uveitis, and ocular
inflammatory conditions unresponsive to topical corticosteroids.
Renal Diseases: To induce a diuresis or remission of proteinuria in idiopathic nephrotic
syndrome or that due to lupus erythematosus.
Respiratory Diseases: Berylliosis, fulminating or disseminated pulmonary tuberculosis
when used concurrently with appropriate antituberculous chemotherapy, idiopathic
eosinophilic pneumonias, symptomatic sarcoidosis.
Rheumatic Disorders: As adjunctive therapy for short-term administration (to tide the
patient over an acute episode or exacerbation) in acute gouty arthritis, acute rheumatic
carditis, ankylosing spondylitis, psoriatic arthritis, rheumatoid arthritis, including juvenile
rheumatoid arthritis (selected cases may require low-dose maintenance therapy). For the
treatment of dermatomyositis, polymyositis, and systemic lupus erythematosus.

Recommended Dosage
The dose of Dexona (Generic Dexamethasone Tablets) is
given below:
Adults: Usually, daily oral dosages of 0.5 mg to 10 mg are sufficient. In
some patients higher dosages may be temporarily required to control
the disease. Once the disease is under control the dosage should be
reduced or tapered off to the lowest suitable level under continuous
monitoring and observation of the patient.
Paediatric population: Usually, daily oral dosages of 0.01 mg to
0.1mg/kg of body weight.
Dosage of glucocorticoids should be adjusted on the basis of the
individual patient's response.

Contraindications
➔
Dexona is contraindicated in patients with a known
hypersensitivity to Dexamethasone or any component of the
tablet.
➔
Systemic infection unless specific anti-infective therapy is
employed.
➔
Avoid live vaccines in patients receiving immunosuppressive
doses (serum antibody response diminished).

Storage Instructions
Dexona (Generic Dexamethasone Tablets) have to be
stored at controlled room temperature i.e. from 20°C to
25°C (68°F to 77°F) with excursions permitted between
15° to 30°C (59° to 86°F).
Keep this as well as all other medicines away from
children and pets.

Warnings
In post-marketing experience tumour lysis syndrome (TLS) has been
reported in patients with haematological malignancies following the
use of dexamethasone alone or in combination with other
chemotherapeutic agents. Patients at high risk of TLS such as
patients with high proliferative rate, high tumour burden, and high
sensitivity to cytotoxic agents, should be monitored closely and
appropriate precaution taken.
Preterm neonates: Available evidence suggests long-term
neurodevelopmental adverse events after early treatment (less than
96 hours) of premature infants with chronic lung disease at starting
doses of 0.25 mg/kg twice daily.

Warnings
Dexamethasone Withdrawal: Adrenal cortical atrophy develops during prolonged
therapy and may persist for years after stopping treatment. Withdrawal of
corticosteroids after prolonged therapy must therefore always be gradual to avoid
acute adrenal insufficiency, being tapered off over weeks or months according to
the dose and duration of treatment.
Suppression of the inflammatory response and immune function increases the
susceptibility to infections and their severity. The clinical presentation may often
be atypical, and serious infections such as septicaemia and tuberculosis may be
masked and may reach an advanced stage before being recognised.
Appropriate anti-microbial therapy should accompany glucocorticoid therapy
when necessary e.g. in tuberculosis and viral and fungal infections of the eye.
Chickenpox is of particular concern since this normally minor illness may be fatal
in immunosuppressed patients.

Warnings
Measles: Patients should be advised to take particular care to avoid exposure to measles
and to seek immediate medical advice if exposure occurs; prophylaxis with intramuscular
normal immunoglobulin may be needed.
Fungal Infections: Corticosteroids may exacerbate systemic fungal infections and therefore
should not be used in the presence of such infections unless they are needed to control life-
threatening drug reactions. There have been cases reported in which concomitant use of
amphotericin B and hydrocortisone was followed by cardiac enlargement and congestive
heart failure.
Special Pathogens: Latent disease may be activated or there may be an exacerbation of
intercurrent infections due to pathogens, including those caused by Amoeba, Candida,
Cryptococcus, Mycobacterium, Nocardia, Pneumocystis, Toxoplasma.
It is recommended that latent amebiasis or active amebiasis be ruled out before initiating
corticosteroid therapy in any patient who has spent time in the tropics or any patient with
unexplained diarrhea.

Warnings
Visual Disturbance: Visual disturbance may be reported with systemic and topical
corticosteroid use. If a patient presents with symptoms such as blurred vision or other
visual disturbances, the patient should be considered for referral to an ophthalmologist
for evaluation of possible causes which may include cataract, glaucoma or rare diseases
such as central serous chorioretinopathy (CSCR) which have been reported after use of
systemic and topical corticosteroids.
Particular care is required when considering the use of systemic corticosteroids in patients
with the following conditions and frequent patient monitoring is necessary:
Osteoporosis , Hypertension or congestive heart failure, Existing or previous history of
severe affective disorders, Diabetes mellitus, History of tuberculosis, Glaucoma, Previous
corticosteroid-induced myopathy, Liver failure, Renal insufficiency, Hypothyroidism,
Epilepsy, Peptic ulceration, Migraine, Certain parasitic infestations in particular
amoebiasis, Incomplete natural growth since glucocorticoids on prolonged administration
may accelerate epiphyseal closure.

Warnings
Caution should be exercised when using corticosteroids in patients who have recently
suffered myocardial infarction as myocardial rupture has been reported.
After administration of glucocorticoids serious anaphylactoid reactions such as glottis
oedema, urticaria and bronchospasm have occasionally occurred particularly in patients
with a history of allergy.
Dexamethasone Tablets contain lactose. Patients with rare hereditary problems of galactose
intolerance, the Lapp lactase deficiency or glucose-galactose malabsorption should not take
this medicine.
Paediatric Population: Corticosteroids cause dose-related growth retardation in infancy,
childhood and adolescence, which may be irreversible.
Use in the Elderly: The common adverse effects of systemic corticosteroids may be
associated with more serious consequences in old age, especially osteoporosis,
hypertension, hypokalaemia, diabetes, susceptibility to infection and thinning of the skin.
Close clinical supervision is required to avoid life-threatening reactions.

Possible Side Effects
The following side effects have been reported with Dexamethasone (Dexona
Tablets) or other corticosteroids:
Allergic Reactions: Anaphylactoid reaction, anaphylaxis, angioedema.
Cardiovascular: Bradycardia, cardiac arrest, cardiac arrhythmias, cardiac
enlargement, circulatory collapse, congestive heart failure, fat embolism,
hypertension, hypertrophic cardiomyopathy in premature infants, myocardial
rupture following recent myocardial infarction (see WARNINGS: Cardio-Renal),
edema, pulmonary edema, syncope, tachycardia, thromboembolism,
thrombophlebitis, vasculitis.
Dermatologic: Acne, allergic dermatitis, dry scaly skin, ecchymoses and petechiae,
erythema, impaired wound healing, increased sweating, rash, striae, suppression
of reactions to skin tests, thin fragile skin, thinning scalp hair, urticaria.

Possible Side Effects
Endocrine: Decreased carbohydrate and glucose tolerance, development of cushingoid
state, hyperglycemia, glycosuria, hirsutism, hypertrichosis, increased requirements for
insulin or oral hypoglycemic agents in diabetes, manifestations of latent diabetes mellitus,
menstrual irregularities, secondary adrenocortical and pituitary unresponsiveness
(particularly in times of stress, as in trauma, surgery, or illness), suppression of growth in
pediatric patients.
Fluid and Electrolyte Disturbances: Congestive heart failure in susceptible patients, fluid
retention, hypokalemic alkalosis, potassium loss, sodium retention, tumor lysis syndrome.
Gastrointestinal: Abdominal distention, elevation in serum liver enzyme levels (usually
reversible upon discontinuation), hepatomegaly, increased appetite, nausea, pancreatitis,
peptic ulcer with possible perforation and hemorrhage, perforation of the small and large
intestine (particularly in patients with inflammatory bowel disease), ulcerative
esophagitis.

Possible Side Effects
Metabolic: Negative nitrogen balance due to protein catabolism.
Musculoskeletal: Aseptic necrosis of femoral and humeral heads, loss of muscle mass,
muscle weakness, osteoporosis, pathologic fracture of long bones, steroid myopathy,
tendon rupture, vertebral compression fractures.
Neurological/Psychiatric: Convulsions, depression, emotional instability, euphoria,
headache, increased intracranial pressure with papilledema (pseudotumor cerebri)
usually following discontinuation of treatment, insomnia, mood swings, neuritis,
neuropathy, paresthesia, personality changes, psychic disorders, vertigo.
Ophthalmic: Exophthalmos, glaucoma, increased intraocular pressure, posterior
subcapsular cataracts, vision blurred.
Other: Abnormal fat deposits, decreased resistance to infection, hiccups, increased or
decreased motility and number of spermatozoa, malaise, moon face, weight gain.

Dexona Tablets Overdosage
It is difficult to define an excessive dose of a corticosteroid as
the therapeutic dose will vary according to indication and
patient requirements.
Exaggeration of corticosteroid related adverse effects may
occur. Treatment should be asymptomatic and supportive as
necessary.

Dexona Tablets During Pregnancy
Dexamethasone (Dexona Tablets) has been classified by the US FDA as
Pregnancy Category C.
Corticosteroids have been shown to be teratogenic in many species when
given in doses equivalent to the human dose. Animal studies in which
corticosteroids have been given to pregnant mice, rats, and rabbits have
yielded an increased incidence of cleft palate in the offspring. There are no
adequate and well-controlled studies in pregnant women.
Corticosteroids should be used during pregnancy only if the potential
benefit justifies the potential risk to the fetus. Infants born to mothers who
have received substantial doses of corticosteroids during pregnancy should
be carefully observed for signs of hypoadrenalism.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Dexona (Generic Dexamethasone Tablets) is used to treat conditions such as arthritis, blood/hormone disorders, allergic reactions, skin diseases, eye problems, breathing problems, bowel disorders, cancer, and immune system disorders. It is also used as a test for an adrenal gland disorder (Cushing's syndrome). Dexamethasone belongs to a class of drugs known as corticosteroids. This medicine decreases your immune system's response to various diseases to reduce symptoms such as swelling and allergic-type reactions.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Uses of Dexona 0.5mg Tablets: Allergic States: Control of severe or incapacitating allergic conditions intractable to adequate trials of conventional treatment in asthma, atopic dermatitis, contact dermatitis, drug hypersensitivity reactions, perennial or seasonal allergic rhinitis, and serum sickness. Dermatologic Diseases: Bullous dermatitis herpetiformis, exfoliative erythroderma, mycosis fungoides, pemphigus, and severe erythema multiforme (Stevens-Johnson syndrome). Endocrine Disorders: Primary or secondary adrenocortical insufficiency (hydrocortisone or cortisone is the drug of choice; may be used in conjunction with synthetic mineralocorticoid analogs where applicable; in infancy mineralocorticoid supplementation is of particular importance), congenital adrenal hyperplasia, hypercalcemia associated with cancer, and nonsuppurative thyroiditis.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Uses of Dexona 0.5mg Tablets: Gastrointestinal Diseases: To tide the patient over a critical period of the disease in regional enteritis and ulcerative colitis. Hematologic Disorders: Acquired (autoimmune) hemolytic anemia, congenital (erythroid) hypoplastic anemia (Diamond-Blackfan anemia), idiopathic thrombocytopenic purpura in adults, pure red cell aplasia, and selected cases of secondary thrombocytopenia. Miscellaneous: Diagnostic testing of adrenocortical hyperfunction, trichinosis with neurologic or myocardial involvement, tuberculous meningitis with subarachnoid block or impending block when used with appropriate antituberculous chemotherapy. Neoplastic Diseases: For the palliative management of leukemias and lymphomas.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Uses of Dexona 0.5mg Tablets: Nervous System: Acute exacerbations of multiple sclerosis, cerebral edema associated with primary or metastatic brain tumor, craniotomy, or head injury. Ophthalmic Diseases: Sympathetic ophthalmia, temporal arteritis, uveitis, and ocular inflammatory conditions unresponsive to topical corticosteroids. Renal Diseases: To induce a diuresis or remission of proteinuria in idiopathic nephrotic syndrome or that due to lupus erythematosus. Respiratory Diseases: Berylliosis, fulminating or disseminated pulmonary tuberculosis when used concurrently with appropriate antituberculous chemotherapy, idiopathic eosinophilic pneumonias, symptomatic sarcoidosis. Rheumatic Disorders: As adjunctive therapy for short-term administration (to tide the patient over an acute episode or exacerbation) in acute gouty arthritis, acute rheumatic carditis, ankylosing spondylitis, psoriatic arthritis, rheumatoid arthritis, including juvenile rheumatoid arthritis (selected cases may require low-dose maintenance therapy). For the treatment of dermatomyositis, polymyositis, and systemic lupus erythematosus.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Recommended Dosage The dose of Dexona (Generic Dexamethasone Tablets) is given below: Adults: Usually, daily oral dosages of 0.5 mg to 10 mg are sufficient. In some patients higher dosages may be temporarily required to control the disease. Once the disease is under control the dosage should be reduced or tapered off to the lowest suitable level under continuous monitoring and observation of the patient. Paediatric population: Usually, daily oral dosages of 0.01 mg to 0.1mg/kg of body weight. Dosage of glucocorticoids should be adjusted on the basis of the individual patient's response.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Storage Instructions Dexona (Generic Dexamethasone Tablets) have to be stored at controlled room temperature i.e. from 20°C to 25°C (68°F to 77°F) with excursions permitted between 15° to 30°C (59° to 86°F). Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Warnings In post-marketing experience tumour lysis syndrome (TLS) has been reported in patients with haematological malignancies following the use of dexamethasone alone or in combination with other chemotherapeutic agents. Patients at high risk of TLS such as patients with high proliferative rate, high tumour burden, and high sensitivity to cytotoxic agents, should be monitored closely and appropriate precaution taken. Preterm neonates: Available evidence suggests long-term neurodevelopmental adverse events after early treatment (less than 96 hours) of premature infants with chronic lung disease at starting doses of 0.25 mg/kg twice daily.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Warnings Dexamethasone Withdrawal: Adrenal cortical atrophy develops during prolonged therapy and may persist for years after stopping treatment. Withdrawal of corticosteroids after prolonged therapy must therefore always be gradual to avoid acute adrenal insufficiency, being tapered off over weeks or months according to the dose and duration of treatment. Suppression of the inflammatory response and immune function increases the susceptibility to infections and their severity. The clinical presentation may often be atypical, and serious infections such as septicaemia and tuberculosis may be masked and may reach an advanced stage before being recognised. Appropriate anti-microbial therapy should accompany glucocorticoid therapy when necessary e.g. in tuberculosis and viral and fungal infections of the eye. Chickenpox is of particular concern since this normally minor illness may be fatal in immunosuppressed patients.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Warnings Measles: Patients should be advised to take particular care to avoid exposure to measles and to seek immediate medical advice if exposure occurs; prophylaxis with intramuscular normal immunoglobulin may be needed. Fungal Infections: Corticosteroids may exacerbate systemic fungal infections and therefore should not be used in the presence of such infections unless they are needed to control life- threatening drug reactions. There have been cases reported in which concomitant use of amphotericin B and hydrocortisone was followed by cardiac enlargement and congestive heart failure. Special Pathogens: Latent disease may be activated or there may be an exacerbation of intercurrent infections due to pathogens, including those caused by Amoeba, Candida, Cryptococcus, Mycobacterium, Nocardia, Pneumocystis, Toxoplasma. It is recommended that latent amebiasis or active amebiasis be ruled out before initiating corticosteroid therapy in any patient who has spent time in the tropics or any patient with unexplained diarrhea.
  • 15. Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Warnings Visual Disturbance: Visual disturbance may be reported with systemic and topical corticosteroid use. If a patient presents with symptoms such as blurred vision or other visual disturbances, the patient should be considered for referral to an ophthalmologist for evaluation of possible causes which may include cataract, glaucoma or rare diseases such as central serous chorioretinopathy (CSCR) which have been reported after use of systemic and topical corticosteroids. Particular care is required when considering the use of systemic corticosteroids in patients with the following conditions and frequent patient monitoring is necessary: Osteoporosis , Hypertension or congestive heart failure, Existing or previous history of severe affective disorders, Diabetes mellitus, History of tuberculosis, Glaucoma, Previous corticosteroid-induced myopathy, Liver failure, Renal insufficiency, Hypothyroidism, Epilepsy, Peptic ulceration, Migraine, Certain parasitic infestations in particular amoebiasis, Incomplete natural growth since glucocorticoids on prolonged administration may accelerate epiphyseal closure.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Warnings Caution should be exercised when using corticosteroids in patients who have recently suffered myocardial infarction as myocardial rupture has been reported. After administration of glucocorticoids serious anaphylactoid reactions such as glottis oedema, urticaria and bronchospasm have occasionally occurred particularly in patients with a history of allergy. Dexamethasone Tablets contain lactose. Patients with rare hereditary problems of galactose intolerance, the Lapp lactase deficiency or glucose-galactose malabsorption should not take this medicine. Paediatric Population: Corticosteroids cause dose-related growth retardation in infancy, childhood and adolescence, which may be irreversible. Use in the Elderly: The common adverse effects of systemic corticosteroids may be associated with more serious consequences in old age, especially osteoporosis, hypertension, hypokalaemia, diabetes, susceptibility to infection and thinning of the skin. Close clinical supervision is required to avoid life-threatening reactions.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Possible Side Effects The following side effects have been reported with Dexamethasone (Dexona Tablets) or other corticosteroids: Allergic Reactions: Anaphylactoid reaction, anaphylaxis, angioedema. Cardiovascular: Bradycardia, cardiac arrest, cardiac arrhythmias, cardiac enlargement, circulatory collapse, congestive heart failure, fat embolism, hypertension, hypertrophic cardiomyopathy in premature infants, myocardial rupture following recent myocardial infarction (see WARNINGS: Cardio-Renal), edema, pulmonary edema, syncope, tachycardia, thromboembolism, thrombophlebitis, vasculitis. Dermatologic: Acne, allergic dermatitis, dry scaly skin, ecchymoses and petechiae, erythema, impaired wound healing, increased sweating, rash, striae, suppression of reactions to skin tests, thin fragile skin, thinning scalp hair, urticaria.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Possible Side Effects Endocrine: Decreased carbohydrate and glucose tolerance, development of cushingoid state, hyperglycemia, glycosuria, hirsutism, hypertrichosis, increased requirements for insulin or oral hypoglycemic agents in diabetes, manifestations of latent diabetes mellitus, menstrual irregularities, secondary adrenocortical and pituitary unresponsiveness (particularly in times of stress, as in trauma, surgery, or illness), suppression of growth in pediatric patients. Fluid and Electrolyte Disturbances: Congestive heart failure in susceptible patients, fluid retention, hypokalemic alkalosis, potassium loss, sodium retention, tumor lysis syndrome. Gastrointestinal: Abdominal distention, elevation in serum liver enzyme levels (usually reversible upon discontinuation), hepatomegaly, increased appetite, nausea, pancreatitis, peptic ulcer with possible perforation and hemorrhage, perforation of the small and large intestine (particularly in patients with inflammatory bowel disease), ulcerative esophagitis.
  • 19. Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Possible Side Effects Metabolic: Negative nitrogen balance due to protein catabolism. Musculoskeletal: Aseptic necrosis of femoral and humeral heads, loss of muscle mass, muscle weakness, osteoporosis, pathologic fracture of long bones, steroid myopathy, tendon rupture, vertebral compression fractures. Neurological/Psychiatric: Convulsions, depression, emotional instability, euphoria, headache, increased intracranial pressure with papilledema (pseudotumor cerebri) usually following discontinuation of treatment, insomnia, mood swings, neuritis, neuropathy, paresthesia, personality changes, psychic disorders, vertigo. Ophthalmic: Exophthalmos, glaucoma, increased intraocular pressure, posterior subcapsular cataracts, vision blurred. Other: Abnormal fat deposits, decreased resistance to infection, hiccups, increased or decreased motility and number of spermatozoa, malaise, moon face, weight gain.

Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Dexona Tablets Overdosage It is difficult to define an excessive dose of a corticosteroid as the therapeutic dose will vary according to indication and patient requirements. Exaggeration of corticosteroid related adverse effects may occur. Treatment should be asymptomatic and supportive as necessary.
  • 21. Dexona (Generic Dexamethasone Tablets) © The Swiss Pharmacy Dexona Tablets During Pregnancy Dexamethasone (Dexona Tablets) has been classified by the US FDA as Pregnancy Category C. Corticosteroids have been shown to be teratogenic in many species when given in doses equivalent to the human dose. Animal studies in which corticosteroids have been given to pregnant mice, rats, and rabbits have yielded an increased incidence of cleft palate in the offspring. There are no adequate and well-controlled studies in pregnant women. Corticosteroids should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Infants born to mothers who have received substantial doses of corticosteroids during pregnancy should be carefully observed for signs of hypoadrenalism.
