The active ingredient contained in Champix
is Varenicline Tartrate. Each 0.5 mg
Champix tablet contains 0.85 mg of
Varenicline Tartrate equivalent to 0.5 mg of
Varenicline free base and each 1mg
Champix tablet contains 1.71 mg of
Varenicline Tartrate equivalent to 1 mg of
Varenicline free base
6. Uses of Champix Tablets

Champix is a nicotinic receptor partial
agonist indicated for use as an aid to
smoking cessation treatment. To increase
your chance of success, use this medicine
with a stop-smoking program that includes
education, support, and counseling.
7. Champix Dosage

➢The recommended dose is 1 mg Champix
(Varenicline Tartrate Tablets) twice daily following a
1-week titration as follows: Days 1-3: 0.5 mg once
daily, Days 4-7: 0.5 mg twice daily and Day 8 – End
of treatment: 1 mg twice daily.
➢The patient must set a date to stop smoking.
Dosing should usually start 1 week to 2 weeks
prior to this date. Patients who are not willing or able
to set the target quit date within 1 to 2 weeks, can
be offered to start treatment and then choose their
own quit date within 5 weeks.
Champix must be taken orally after eating and
with a full glass of water.

➢Patients must be treated with Champix for 12
weeks. For patients who have successfully
stopped smoking at the end of 12 weeks, an
additional course of 12 weeks treatment at 1
mg twice daily can be considered for the
maintenance of abstinence.

Champix (Varenicline Tartrate Tablets)
have to be stored at controlled room
temperature i.e. 25°C (77°F) with
excursions permitted between 15° to
30°C (59° to 86°F).
Keep this as well as all other medicines
away from children and pets.

Dosage adjustment may be required for
some some medicinal products (examples
include theophylline, warfarin and insulin)
after stopping smoking.
Changes in behaviour or thinking, anxiety,
psychosis, mood swings, aggressive
behaviour, depression, suicidal ideation and
behaviour and suicide attempts have been
reported in some patients attempting to quit
smoking with Champix in the post-marketing
experience.

Depressed mood, rarely including suicidal ideation and
suicide attempt, may be a symptom of nicotine
withdrawal. Clinicians should be aware of the possible
emergence of serious neuropsychiatric symptoms in
patients attempting to quit smoking with or without
treatment. In case serious neuropsychiatric symptoms
occur whilst on Varenicline treatment, patients should
discontinue Varenicline tablets immediately.
Care must be taken with patients with a history of
psychiatric illness and patients should be advised
accordingly. Patients taking Champix Tablets must
be informed to notify their doctor of new or worsening
cardiovascular symptoms and to seek immediate
medical attention in case they experience signs and
symptoms of myocardial infarction or stroke.

During clinical trials and post-marketing experience
there have been reports of seizures in patients with
or without a history of seizures, treated with
Champix.
Champix must be used cautiously in patients with a
history of seizures or other conditions that potentially
lower the seizure threshold.
At the end of treatment, discontinuation of
Varenicline Tartrate Tablets was associated with
an increase in irritability, urge to smoke, depression,
and/or insomnia in up to 3% of patients, therefore
dose tapering may be considered.

There have been post-marketing reports of
hypersensitivity reactions including
angioedema and reports of rare but severe
cutaneous reactions, including Stevens-
Johnson Syndrome and Erythema Multiforme
in patients using varenicline.
Patients that experience such symptoms
must discontinue treatment with Champix
(Varenicline Tartrate Tablets) and contact
their doctor immediately.

Contraindications
Champix is contraindicated in patients with a
hypersensitivity to Varenicline Tartrate or any
other component of this medication.
15. Varenicline Tartrate Side Effects

➢Most common Varenicline Tartrate Side
effects with an incidence arte of more
than 5% and twice the rate seen in
placebo-treated patients were nausea,
abnormal (e.g., vivid, unusual, or strange)
dreams, constipation, flatulence, and
vomiting.

In case of Champix (Varenicline Tartrate
Tablets) overdose, standard supportive
measures must be instituted as required.

Varenicline Tartrate (Champix tablets) has
been classified by the US FDA as
Pregnancy Category C.
Varenicline was not teratogenic in rats and
rabbits at oral doses up to 15 and 30 mg/kg/day,
respectively which is 36 and 50-times the
maximum recommended human daily exposure.
There are no adequate and well-controlled
studies conducted in pregnant women.
Champix can be used during pregnancy only if
the potential benefit justifies the potential risk to
the fetus.
