6. Principle:
This test is used to differentiate keratin from the rest of the
proteins.
Principle:
The solubility test is based on the ability of proteins to
dissolve in water other than keratin. Keratin is a large fibrous
protein having numerous disulfide bonds. These disulfide
bonds make it insoluble in water while the rest of the proteins
are highly soluble due to hydrophilic amino acids present in
them.

Take 10 ml distilled water in
a test tube
Add the given powder into
the water
Shake the test tube

• If the given powder is
insoluble in water, it
indicates the insoluble
protein is keratin.

If the sample is insoluble in
distilled water then it confirms
the presence of keratin.
