2. INTRODUCTION
 Spectroscopy is the study of interaction between
matter and electromagnetic radiation.
 Electromagnetic radiation refers to waves of
electromagnetic field, radiating through space carrying
electromagnetic radiant energy
 UV- Visible Spectroscopy uses UV radiation ranging
from 100nm to 380nm and Visible radiation ranging
from 400nm to 700 nm form the electromagnetic
spectrum.

3. PRINCIPLE
 UV-Visible Spectroscopy
refers to absorption
spectroscopy.
 Molecules containing
bonding and nonbonding
electrons can absorb energy
in the form of UV or visible
light and gets excited to
higher molecular orbitals.

5. 1) Light Source - Emits light of particular
wavelength.
UV light- Deuterium arc lamp
Visible light- Tungsten filament lamp
2) Sample Holder - called as Cuvettes, made up
of quartz
3) Monochromator - selects particular
wavelength of light. Prisms are used.
4) Detector – measures light transmitted from
the sample. Typically photomultiplier tube
and photodiode are used as detectors. 5

6. APPLICATIONS
 Qualitative as well as quantitative determination
of different analytes such as metal ions, organic
compounds and biological macro and micro
molecules.
 Detection of impurities.
 Detection of functional groups.
 Structural elucidation of organic compounds.
 Quantitative analysis of pharmaceutical
substances.
 As HPLC Detector.
