Bgla Enzyme Lab Report
Gene BglA was sub cloned in pET21a (+), recombinant vector transformed and propagated in E. coli
BL21 CodonPlus under the control of T7/lac promoter system. The positive bacterial transformants
were grown in LB ampicillin broth (100 µg mL−1) and induced with IPTG for the production of
recombinant BglA enzyme. Optimal heterologous expression and efficient folding of BglA protein
was observed with 0.5 mM IPTG, after 72 h incubation at 22°C. The use of reduced concentration of
IPTG for getting maximum gene expression, was proved an efficient method for the production of
active soluble enzyme. The protein was purified by applying two subsequent purification steps
employing ion exchange and hydrophobic interaction chromatography after heat treatment ... Show

BglA was remain active even in the existence of 600 mM glucose. However, BglA activity was
gradually repressed with the increase of glucose concentration, and Ki value for glucose was found to
be 1200 mM as shown in figure 4b. Organic solvents have proven an obvious constrain for the BglA
activity, enzyme was completely inactive by adding 40% (v/v) methanol, ethanol, n butanol and
isopropanol whereas, 40% (v/v) acetone, ethyl acetate and acetonitrile strongly repressed BglA
activity up to 18%, 38%, 50%, respectively.
3.6. Shelf life of recombinant BglA
Storage stability of BglA crude media supernatant, heat treated supernatant (at 70°C for an hour) and
purified enzyme was an important and crucial parameter for commercial utilization. BglA enzyme has
an exceptional storage stability, retaining 100% initial activity after the storage for one year at 4°C, in
all the three ways of storage (crude supernatant, heat treated and purified). Both crude heat treated and
purified BglA has retained 100% initial activity at room temperature (30 35°C) up to 240 days but
after 95 days the crude enzyme showed marginal decline

How Did Davy Crockett Influence American History
Davy Crockett, an American icon, actually made a huge impact in American history. His early life and
his background is interesting and may be unique to others. Crockett served the country in the US
government. He is also one of the many heroes who died defending the Alamo. Davy Crockett s early
life may have helped him later in his life. Crockett was born on August 17, 1786 in Tennessee. He
learned to shoot a rifle when he was just eight years old. He ran away for more than two years in the
wilderness after he got into a fight with a school bully. He was just thirteen according to
Biography.com. He married Polly Finley, and had three children. When she died, he married Elizabeth
Patton, a widow with two children. Crockett was successful in life. He became a scout in the militia to
fight the Creek Indians in 1813, and returned home in 1815. All of this experienced led to time as a
government official. ... Show more content on Helpwriting.net ...

James Agee and Walker Evans Essay
James Agee and Walker Evans
Fortune Magazine, in July and August of 1936, sent James Agee and Walker Evans to research a story
on sharecropping. In the preface of Let Us Now Praise Famous Men, Agee describes it as a curious
piece of work. They were to produce an article on cotton tenantry in the United States, in the form of a
photographic and verbal record of the daily living and environment of an average white family of
tenant farmers, (IX). James Agee and Walker Evans set out to write and photograph an article for a
magazine, and ended up experimenting with the form of the novel itself.
James Agee was born in 1909 in Knoxville, Tennessee, but was in the urban middle class, so he had no
experience with cotton farming. When ... Show more content on Helpwriting.net ...
Agee and Evans set out to publish an article in Fortune Magazine, but when they brought back their
work to the editors, they were rebuked. The article was over 400 pages of controversial writing and
even various book editors refused to publish it as a novel. Finally in 1941 it was published by Mifflin
in Massachusetts after the author was forced to take out a few outlawed words in the state. They failed
in their original mission to produce an article, but undoubtedly presented a photographic and verbal
record of the average white southern tenant farmer families. By living with three different families for
four weeks, Evans depicted their innermost sorrows through his photographs, and Agee portrayed and
emboldened their spirit by his written words. The novel itself was an experiment in form, with Evans
photos all placed in the front without captions, rather than dispersed throughout the novel. Even in the
text, Agee continually stops himself mid description and enters into a stream of consciousness
describing the difficulties of articulating accurately the tenant families true expressions. For these
reasons, it was considered a failure when it was published, but also for the same reasons it was
considered an amazing success twenty years later in the 1960s.
In the Preface, Agee conveys that this is a book only by necessity. More seriously, it is an effort in
human actuality, in which the reader is no less centrally involved than the

Branding And Marketing Of Mdl For The Twin Cities Market...
Branding and Marketing of MDL for the Twin Cities Market Although they may seem one in the
same, we want to explain and differentiate the terms Branding and Marketing. Although they are
closely related and should actively complement each other for a successful business strategy, they
should have separate and individualized goals and budgets. Branding a long term process by which we
will create and manufacture the presence of MDL through consistent visibility, creating simple, yet
powerful, to the point messages which boost the awareness of MDL, along the lines of Have You Been
To? Branding is built by presenting repetitive and cohesive messages, increasing the familiarity of the
name, its purpose and vision to build a reputation and ultimately an experience. Brand isn t traceable,
but is the tool that makes the marketing successful. Marketing is an immediate action, which puts
consumers in the midst of the daily, monthly, seasonal, and special event specifics and ongoings at
MDL. BRANDING increasing awareness of MDL
Thus far, our branding campaign has been limited primarily to the following:
1. Six billboards on Northbound 35
2. Face to face
3. MDL brochure displays we have instituted with ski, bike, archery, and disc golf shops throughout
the metro area, further motivated by trade incentives for employees of the above mentioned
businesses. Secondary elements in our branding campaign:

As planning for the new Barrett Complex had just begun, they went to the Dean and presented their
idea for a sustainable residence hall at Barrett. The Dean was very receptive to the idea and formed the
Sustainable Living Community to be part of the planning for the new complex (History of BSC 2013).
While Construction kept getting delayed, students in the interim formed the Sustainability House at
Barrett as a club to establish a series of short term and long term goals until they could have their own
residential community on campus. Together, the students developed the final mission statement for the
Sustainability House at Barrett:
Understanding sustainability as a holistic perspective from which to view the world allows us to
recognize the interconnectedness of all things. This perspective leads to a critical examination of our
role in society and the complex and evolving relationship between environmental, economic, and
social issues. We realize that our success and happiness depends not only on ourselves, but also on our
community and surroundings. As a student initiated and consensus based group, the Sustainability

I have always believed that joining cheer was one of the best decisions I have ever made. Cheer has
always been a big part of my life; from practices to competitions I have always been kept busy by it. I
can always go to practice and forget about everything else going on in my life. My coaches and

my team and our success mattered. All was good for about four seasons; my team and i had one most

GDP is an important measure that indicates an economy s performance and affects employment,
inflation and foreign trade among others. Extensive research has been done on the factors that impact
real GDP growth, but many different models exist and there is no consensus that is universally
accepted. One important influence is government expenditure but economic theory does not clearly
state the relationship between government spending and GDP growth. Free market supporters believe
that government expenditure will lower real GDP per capita since they expect that government

Effects Of Xenophobia In Hungary
The root of this continuing and constantly high level of xenophobia can be found in the country s
historical legacy. As Pál Nyíri (2003) points out when talking about the possible systemic sources of
xenophobia in the country, xenophobia can have economic and cultural factors, where economic the
historical factors refer to previous incidents ethnic or religious conflicts (p. 30). Neményi and Takács s
(2005) findings relate to this idea: in their study they found that historical experiences can have an
influencing effect on the extent of xenophobia (p. 51).
From a historic point of view these theories are supported in the case of Hungary, as the country has
experienced being taken over by other nations more than one time, which can explain the current fear
of being occupied and the authority being taken out of Hungarian hands again. The most important
and longest of these occupations were the Ottoman occupation that lasted 150 years and the Soviet
occupation that lasted nearly 50 years. These invasions are particularly important because the former
brought on the first big change of demographics in the country, and the latter due to the strong
influence it still has on the political system. ... Show more content on Helpwriting.net ...
9). With Hungary s history, even the quite recent one being rich of experiences of foreign subjugation,
this is especially important in this case of refugees, where the portrayal of Brussels is crucial in the

the first time Hungary faced serious demographic changes, and the ethnic map of the country

Amplitude Modulation Report
Introduction
Radio Stations within the UK are listened to frequently by large audiences throughout the country.
This is achieved through the use of a number of different network media platforms either in Analogue
or Digital form; including the Internet and traditional wired and wireless broadcasting such as AM/
FM networks. The objective of this report is to choose two examples of a broadcast radio platform one
being digital with the other being analogue and then describe in detail of the modulation methods
employed in each case. The 2 broadcast radio platforms that will be focused on in this report will be
AM/ FM and Digital Radio (DAB); providing background information on each technology and

Once the signal is received, these receivers will demodulate the signal meaning to extract the
modulation from the received signal.
AM
AM (Amplitude Modulation) is the original way of broadcasting radio. Lee de Forest and Reginald
Fessenden are given credit for the development of the technique which was carried out successfully
for the first time in the mid 1870s, its frequency ranges from 535 to 1705 KHz and the typical
bandwidth of an AM channel is 10 kHz. The name AM itself gives a clear indication to the type of
modulation technique used; Amplitude Modulation. Amplitude Modulation is a modulation technique
which works is by modifying the carrier wave s strength by increasing or decreasing its peaks in line
with the variations of the input sound; the strength of the wave is known as amplitude, through all of
this the frequency remains constant the same. This type of modulation technique is more susceptible
to noise and or interference due to the fact noise effects amplitude directly. The change in signal
strength allows the conveyance of information through the radio wave that is sent out via a transmitter
and is received by a radio device otherwise known as the receiver. When the signal is received,
demodulation takes place; this can be dealt with in two ways depending on the device. Either a Diode
AM detector will carry out the demodulation (which is

The Genetic Code Essay
In the year of 1920 a theory created by two different scientists was born, Russian Scientist/Chemist
A.I.Oparin and an English Geneticist J.B.S both developed this theory known as The Primordial Soup
Theory . This theory basically states that all life 3.8 3.5 billon years ago came from a tiny puddle filled
with amino acids and elements from Earth s atmosphere. An experiment to test this theory was called
Miller Urey experiment courtesy of scientist Stanley Miller and Harold Urey. In this experiment the
two used electric shocks/charges and were able to birth organic mixtures (compounds).Oparin and

Anyways this RNA world theory claims that RNA is the origin of all living organisms on planet earth.
Even though there is no experiment to back this hypothesis there is however research and claims in
Carl Woese s book by the name of The Genetic Code . RNA is a protein also known as ribonucleic
acid, both RNA and DNA house tons of our (human beings) genetic codes. Seeing that the theory says
that it might not be the exact origin of life on earth it stilled played a part in evolution. Also recent
cells and those that came before it were said to have evolved and bonded with enzyme compound to

of dioxyribose nucleic acid (DNA) and forms of proteins. These chemicals were said to have had

the structure and the processes of the old business entities. Josiah Wedgwood and his pottery business

First of all, financial institutions were rare in Britain at this time. Therefore, it was very hard to find an
investor in order to buy machines and equipment. Once again, networking practices were crucial to
contact aristocrats who were willing to lend money. Another financial problem is related to the lack of
standards and tools to run a business at this time. As Wedgwood explained during the vase madness
period, the financial health of the company was in peril even if they sold a considerable amount of
vases. In fact, he bought a lot of raw materials but in the same time his business partner didn t collect
quite quickly the bills. This situation influenced negatively the liquid assets of Wedgwood s business.
As Wedgwood and Bentley were afraid of making the same mistake in the future, they created a price
book of workmanship and separate fixed and variable costs. These tools are still used nowadays, the
price book could be compared to the profit and loss statement and fixed and variable costs are one of
the most famous theories in management

finding that our sexual identity, is ever changing most of our lives. I have given my own identity some

Some researchers state that genetics and environmental factors are important determining element of
homosexual behavior. They also state that birth place relates to the sexual orientation of marriage
partner. Being born in urban settings increased the probability of homosexual marriage and decreased
the probability of heterosexual marriage. Everyone environment effects your sexuality in some way or
another either negative or positively by supporting or repressing it. It plays a role in whether your
homosexual, heterosexual, or bi sexual. (Rathus, Nevid, Fichner Rathus, 2005) Our text states that
history places sexual attitudes and behavior in context. It informs us as to whether sexual behavior
reflects trends that have been with us through the millennia or the customs of a particular culture and

from one time and place to another. (Rathus, Nevid, Fichner Rathus, 2005)

Back at the fire nation docks run by commander Zhao. Zuko, Khai Li, and Iroh were currently
listening to resent battle plans told by Zhao himself. He was facing a map of the world on the wall of
the tent, showing all four nations and the various landmarks. Zuko had his back to Zhao still angered
by having to be in the same tent as Zhao, Khai Li sitting next to him on his left trying to give him
some sort of comfort for this situation, while Iroh was to the right side of the tent looking at the
various spear like weapons Zhao had collected.
And by years end, the earth kingdom capital will be under our rule. Zhao concluded, turning to face
them. the fire lord will finally claim victory in this war.

No, we haven t found a thing. Khai Li replied quickly almost stuttering, trying to avoid Zhao s gaze
while cursing to herself how stupid she sounded as Zhao turned back to Zuko.
Prince Zuko, the avatar is the only one that can stop the fire nation from winning this war, Zhao
informed, getting up from his chair where he had been sitting and walking closer to Zuko, getting in
his face. If you have an ounce of Loyalty left, you ll tell me what you ve found.
I haven t found anything, Zuko shot back at him keeping his cool. It s like you said, the avatar

Zuko got up from his seat, walking forward, after bowing and saying her farewells to Zhao, Khai Li
close behind Zuko when the guards inside of the tent pulled their weapons in Zuko s path and two new
guards grabbed Khai Li by her arms to keep her from attacking or moving, one of Zhao s subordinates

Misprinted Type is a type foundry based in Belo Horizonte, MG, Brazil. This type foundry was

about the creator, Eduardo Recife. From there you can then understand the creations of Misprinted

piece of artwork. Recife decided to create his own typefaces in 1997, at first for himself. The then
figured it would be good to let others use it, so he distributed it online. He then thought to name the

Summary OfThe Merger Of Two Competing Hospitals
Healthcare Management Final Paper
In the short story The Merger of Two Competing Hospitals , we see both Porter Regional Medical
Center and Banner Regional Medical Center reach an agreement to form a merger. Both Hospitals had
at least one major current flaw and by teaming up they no longer had to worry about the aspect of
competing with each other. PRMC faced the problem of annually losing large sums of money. While
over at BRMC the problem laid within the infrastructure and not the problem of money.
After the completion of both hospitals coming together to form a newly named hospital, the question
of creating an executive team in charge of management comes about. In my opinion, before moving
forward in any way to create the executive team, candidates must be considered for their potential
leadership. Just imagine how hard it would be to fail if the newly created hospital looked for
leadership comparable to Arthur T. DeMoulas, when hiring for the executive team. While Market
Basket is not a hospital, the whole idea of the managers acting as leaders would be great for a new
hospital1. This could make all employees act loyal and show great customer service, and lastly having
all the customers (patients) come and leave happy.
According to MIT Sloan Management Review, there are six steps that should be taken to properly
rebuild a management team. The first includes Reducing Role Ambiguity. This is essentially
individuals who leave the new company because of the uncertainty the future holds for them. To help
this, the board should do its best to ensure the company will be fine to the new executive team, as well
as quickly remove any individuals who doubt the new company will be a success. Having just a few
individuals who doubt the potential of the merger could quickly ruin a whole team and bring moral
down. The second step states Due Diligence Around Talent Is a Dangerous Corner to Cut . To solve
this, the board needs to get to know all the managers on both sides of the merger and actually find out
about their full talent potential. The third part states Recognize Old Habits Die Hard But Not All
Should . Each company before the merger had a certain way of doing things, and while some of those
methods

Essay on CRIMES Are Committed By...CRIMINALS, Guns Are Not...
Calls for more gun control came after the Sandy Hook Elementary School massacre in Newtown,
Connecticut where 20 school students and 6 adults were killed by a man with an assault rifle (Cary).
Soon after the school massacre, President Obama proposed a ban assault rifles and high capacity

Any debate on crime needs to focus on crime prevention and not the guns of law abiding citizens.
Controlling guns is not a crime prevention strategy, it only prevents the those who are physically
weaker than their attackers like women, elderly, and disabled from defending themselves against
bigger, faster and stronger aggressors. Since crime and gun control are serious issues,I used a white
poster board for a simple background and red and black permanent markers to convey that
seriousness. Since the words crimes and criminals go to together and are central to my argument, I
made them both bold, in all caps, and in red so they are the most eye catching. Red is also associated
with blood, so by making criminals red instead of guns I m also indicating what really causes people
to bleed; other people. The the first and last phrases are where I set up my argument and draw my
conclusions, so they are important and therefore bold as well. The first three checklist options are for
comparison only and are not as central to my argument so they are not in bold letters. I am trying to
reach and convince people who don t agree with me, are unsure about the issue, don t have strong
feeling about it, and especially those who don t understand the issue. By using a checklist, I m trying
to explain the core of the issue and my position on it as simply and as clearly as possible. Even if

The Race And Racial Struggle Essay
Numerous scholars and historians believe that Hitler s ideas concerning race and racial struggle have
originated from the philosophies of Charles Darwin, either directly or through various other sources.
In today s context, the word racism means to dislike an individual or a group of individuals based on
the color of their skin. However, during the mid twentieth century, the word racism meant to dislike a
group of individuals because they were intellectually and/or morally inferior. Thus, it becomes the
responsibility of the superior race to take control of the inferior race, or wipe them out completely in
the case of Adolf Hitler. Hitler attempted to create and protect the superior race from the inferior race
by implementing administrative policies designed to protect the former. This belief of superiority
comes from Darwin s theory of survival of the fittest which argued that there are significant
inequalities within each species and that some species get taken advantage of while others take
advantage of others. //This evolutionary theory could have been used as a justification for Hitler to do
something he would have done anyhow, to exterminate roughly six million Jews who, according to
him, belonged to the inferior race . It is fairly clear that Darwin did not mean for his work to be
interpreted the way it did; however, it can be argued that Hitler used this theory of progress through
the elimination of the weak as a method to rationalize his actions to his

Blindness By Jose Saramago s Blindness
Extensionalism that is Presented in Blindness by José Saramago A person is not just characterized by
those particular circumstances that they find themselves in, but by how they respond to those
moments. The phenomenal novel, Blindness, written by Jose Saramago, showcases the storyline of a
country that encounters a widespread disease that leads to several people catching a type of blindness
where they can only see white. The unfortunate people who have received the terribly contagious

possibility of catching a disease where one can only see white in order to remain with her husband
who is ill. By staying with her husband while he is in desperate need of help, she proves that she is
one of the kindest characters in the novel. However, as the book progresses, her trait of kindness
quickly fades away after experiencing an atrocious situation. A group of people, formed in the

Things are Not Always as They Seem with Good Country People
Why that looks like the nice dull young man that tried to sell me a Bible yesterday, Mrs. Hopewell

all that simple (145). At the end of the story, Mrs. Hopewell considers
Manley Pointer simple. Little did she know that this simple man had just caused Hulga severe mental

greatest fault. Situations are not always as they seem, and unless one first looks within, his or her

The Bible carried by Manley turns out to be nothing more than a hiding place for a pocket flask of
whiskey, a pack of cards, and a small blue box with printing on it (135). Joy Hulga appears to be
accepting of her situation, but she is still very ashamed of her appearance. In the story, both Hulga and
Manley wear masks over their true natures. Their final confrontation, however, reveals Manley to be a
cunning atheist while Hulga is exposed as a girl whose naivete sharply contradicts the nihilistic cliches
she professes. After the confrontation in the hay loft with Hulga, Manley is revealed as an abuser and
a thief, but only to Hulga. There are two types of irony throughout this story, situational irony and
verbal irony. Situational irony is found when Hulga is expecting to seduce an innocent, however, it
turns out to be quite different. Joy Hulga is easily the most taken advantage of character in the entire
story. As her ugly sounding name implies, she appears ugly and evil, but deep down she has a desire
for love and happiness. She says, I am one of those people who see through to nothing (115). Little
does she know, that she recognizes one of her greatest weaknesses in this statement. Hulga has said
one thing but her hidden desires cause her several problems later on. For example, the whole scene
that takes place in the hay loft, which is know as verbal irony. Manley was

Effectiveness of a Jury Essay
Evaluate the effectiveness of the jury system in the criminal trial
Juries exists in the criminal trial to listen to the case presented to them and, as a third, non bias party,
decide beyond reasonable doubt if the accused is guilty. For the use of a trial by juror to be effective,
no bias should exists in the jurors judgments, the jurors should understand clearly their role and key
legal terms, and the jury system should represent the communities standards and views whilst
upholding the rights of the accused and society and remain cost and time effective.
The jury system has been used in the criminal trial since the Constitution stated the trial on indictment

This amendment was introduced after the Bruce Burrell case (2005) where, after two weeks of
deliberations, all jurors were firmly in favor of one verdict except for one who was firmly against the
majority verdict. The case illustrated the vulnerabilities of the previous jury system in creating a time
and cost ineffective measure of trialing defendants where justice was not necessarily achieved.
Another recent reform is the passing of legislation in 2006 which provided three additional spare
jurors to be empanelled on lengthy criminal trials for example in the case R v. Wood. These recent
reforms in legislation have led to the jury system being more cost and time effective in the criminal
trial. Allowing a majority vote instead of strict unanimous verdict reduces the risk of a hung jury and
hence prevents jury bribing and jury disagreements. By avoiding a retrial, the criminal trial process
saves huge amounts of time and money for all individuals involved and hence maintains the integrity
of the jury system. Equally, allowing spare jurors in lengthy trials is hugely cost and time effective as
retrials are often avoided in the case where there is misconduct amongst the jurors.
Although the jury system can be argued to be an

Medical errors are among the five most common causes of death in the United States, among them

Implementation of the system to contain costs for voluntary health insurance does not mean that

Choosing A Smart Property Purchase
Choosing where to purchase properties is an important decision. You might be buying your first home
or a home to rent out as an investment property. The fact is, everyone has different needs.
Affordability, accessibility to public transport, and proximity to schools can all have a big impact on
the value of a potential investment property. If you re looking for your dream home, the criteria can be
even more personal.
Knowing how to make a smart property purchase decision can seem overwhelming, so it s important
to understand the basics.
What is your personal situation?
Identifying the factors that are important to you is a good place to start. Ask yourself what you need
the property to achieve for you. Then, see if the property ticks all those important boxes. Knowing
whether you need an investment that is cash flow positive, or if you can sustain an investment
property that might build some long term capital gain, is a good basis for your decision making.
If your plan is to renovate for capital gain, then that off the plan property might not be the right
investment for you. Also, before you think about buying in a regional location, look at the industries
that are integral to the region and consider how secure those industries are. There are no guarantees in
life and if your personal finances can t withstand a downturn, then that location may not be the best
choice for you. How much you can actually afford to borrow is another important factor. Talk to a

The Characteristics And Differences Between Radio And Radio
1. The skills one needs to be a radio listener, are more sophisticated then the skills one would use to
understand and interpret a television program. When listening to the radio, one has to be an active
listener. It requires you to pay more attention as the information is being processed solely through
auditory stimulation. This is unlike watching television which provides both auditory and visual
stimulation. Personally, when listening to Orson Welles War of the Worlds from October 30th 1938, I
found myself more easily distracted comparing to when watching a television program or news story.
It is much easier to pay attention to the later. Moreover, listening to the radio requires a person to use
their own imagination more vividly. The audience has to take in what is being described and create
their own personal picture of the event. Television seems to give us a more realistic understanding of
what it is news anchor is talking about as the consumers can be shown pictures or film footage of the
event being discussed. This add to the audience s view of the incident in its entirety. On the radio, the
news broadcasters have to be more descriptive when describing the currents events as their
descriptions are the sole foundation that the listener s imagination will be based on. It is only through
their description that the listener will get an understanding of whatever is being discussed. The radio
host must therefore put in more detail than a television host. As expected, a

Symphony Orchestra Analysis
A symphony is an extended classical composition for an orchestra, found in Western Art Music. It
often consists of four separate and unique, sections known as movements which are, in some way
however, related to one another. The symphony as a genre began developing during the late 17th
century and continued to do so up until the late 18th century. This important genre of Western Art
Music is closely related to the development of the orchestra. It is therefore critical, in understanding
the symphony, to investigate the development of the orchestra.
In the stretch of a hundred years, the orchestra underwent considerable change in developing into the
standard symphony orchestra of the modern age. When it is first found in the late 1600s during ...

It was truly a breeding ground for musical excellence and innovation. This orchestra was by far the
largest of its kind in the area, and boasted many of the greatest virtuosi of the time. Upon Mozart s
first visit to Mannheim in 1763, the orchestra made a huge impression on him. Besides a large
orchestra, symphonies included various specific musical features and techniques, that had become
trademarks of the style developed in Manheim. One Manheim composer in particular had a large
impact on the orchestra and the symphony. His name is Jan Vaclav Antonin Stamic (1717 1757)
(Germanized to Johann Wenzel Anton Stamitz). He started writing orchestral works to be independent
works in their own right. Orchestras no longer needed to accompany vocal genres to attain any
meaning. This paved the way for what was to become known as abstract music. Stamitz wrote 58
symphonies in his lifetime, and numerous conciertos. He expanded the orchestra, with woodwinds
starting to play an essential role in the orchestral timbre. The orchestra was no longer made up simply
of a haphazard group of instruments, but was starting to take a fixed formal shape. Along with his
contributions to the expansion of the orchestra he also adopted the formal structure of four movements
for the symphony and started using a distinct development section in the first movements, which
became known as sonata form. The opening movement is fast, followed by a slow second movement,
the third movement would be a minuet and trio, and the fourth would end the symphony on a final

Goodnight And Good Luck And The Crucible Comparison
The Crucible and Goodnight and Good Luck are currently relevant due to recent news articles and

Goodnight and Good Luck, people would get fired and denied acceptance in society; They were

To begin, in today s society people are shunned or denied jobs based solely on how they prefer to be

women and men are fleeing from America to seek protection from gender identity discrimination. Like

Capitalist Business Case Study
1. My church is set up very similar to a capitalist business in many different ways. One of these ways
is we take collection every Sunday. I see taking collection as similar to how the government taxes. We

in collection. The church also uses this money to pay for utility bills, and to pay the pastor and any

then the congregation votes. An example of this is when we have a guest speaker in the church, or are

business meeting every year with all the members of the church to elect new deacons and secretaries.
The pastor and elders are voted on differently. Pastors and Elders are only voted on once every four
years, just as the president of the United States is. All of these things are greatly influenced by the
government of the country we live in. We use it as a model to model what should be done in our

The Most Advanced Technology to Prevent Diabetes is the...
Present Technology The most advanced technology for preventing diabetes currently is the insulin
pump. This is a box about the size of a computer mouse and it supplies insulin to your pancreas. It has
many advantages but sadly, it has much more disadvantages. An advantage of having an insulin pump
is that the device delivers insulin to you. It keeps your blood glucose level in range. If your blood
glucose level gets too high you can press a button on your pump to produce insulin (American
Diabetes Association Insulin Pumps ). It is a great machine to have if you have diabetes, it will make
your life much easier. This has lots of advantages but there are also lots of disadvantages of the
present technology which is why we are improving this a lot.
One disadvantage of the present technology is you have to carry it around everywhere. This is
basically shows everyone that you have diabetes and people might feel uncomfortable. That is why
our invention is a microchip inside you that nobody can see. Another disadvantage of the insulin pump
is every hour or two you have to measure your blood glucose level. People have to monitor their blood
glucose rate for the rest of their lives. But with our microchip you never have to worry about checking
your levels again. Also when people take baths, showers or go swimming they have to disconnect the
pump. With our microchip this never has to be worried about. You have to prick your finger 4 5 times
a day to check your

Racism In Cry The Beloved Country
Luke 6:35 declares, But love your enemies, and do good, and lend, expecting nothing in return, and
your reward will be great, and you will be sons of the Most High, for he is kind to the ungrateful and
evil (English Standard Version).Christians should strive to treat others in this way always.
Unfortunately, society fails to follow this verse in the present times as well as the past. Not only does
racism and prejudice occur in the United States, but South Africa as well. Alan Paton illustrated the
unjust in the novel Cry the Beloved Country. James Jarvis is a white, wealthy, Christian farmer that
lives on a hill above Ndotsheni, a town where native South Africans reside and are impoverished.
Jarvis s outlook on the natives became negative. Many events occur, and Jarvis experiences a change
within himself and starts loving the natives how the Bible says Christians should. Mr. Jarvis sinned
against the natives for majority of his life by judging them. It takes a moment of realization to push
him to come to a different outlook on those around him. Mr. Jarvis s thoughts about the natives of
South Africa that are incorrect. Ironically, his son, Arthur Jarvis thoughts pushed him to analyze things
and change some of the ways he lived. Because Mr. Jarvis s did not contain much knowledge on the
everyday life of the natives, his outlook on them became negative. Mr. Jarvis s negative outlook on the
natives consists of his belief that the natives were, ignorant, and knew nothing

The Characters of Antigone and Creon in Antigone by Sophocles
The Characters of Antigone and Creon in Antigone by Sophocles
Antigone is story of divine retribution and human imperfectness. In this tragedy a powerful king,
Creon is brought down by the Gods because of his contempt against their divine laws and true justice
is shown to triumph at the end. Creon makes the mistake of putting his personal views over and above
the divine laws and fails in the eyes of the Gods. He makes the mistake of testing the Gods power and
the remaining story is basically the degeneration of Creon. After the crime of Antigone, Creon is
increasingly shown to be lone warrior in his cause and family and well wishers start deserting him. As
the tragedy progresses Creon becomes ... Show more content on Helpwriting.net ...
Both are to some extent right in their thinking but in the end both are shown to be mere humans in the
eyes of God and fail to rise beyond their puny existence.
At the beginning of the play, Thebes has fought back an armed attack and the brothers, Eteocles and
Polynices have died ;Creon crowns himself the king and issues edicts to proclaim Eteocles a hero to
be buried with full military honours and decrees that the body of the traitor, Polynices be refused
burial and left to rot. He issues warns that any person disobeying his edicts will be put to death.
However, his edicts clash with the divine laws which explicitly state that every dead body deserves a
burial, even that of a traitor. Creon thus starts the chain of his own destruction. He considers himself to
be paramount in his ideals and expects everyone to obey his edicts without question. And for this, he
is willing to risk everything he has : his prestige, his son, throne and his own life at the altar of his
pride. And in the end, he loses.
The Gods however give a number of chances to Creon to remedy his folly of judgement but he misses
every one of them. When Creon gets the news of Polynices burial he hastily judges that the sentry or
the sentries have been bribed and is on the verge of having the latter tortured or even killed. In his
illogical state of mind he accuses the Chorus of lunacy and illogical behaviour

Opiate Dependency
Opiate Dependency vs. Opiate Addiction
Today the recent growth of prescription opioid painkillers has made opiate use far more domesticated
and widespread than ever before. Even though heroin use has declined, the use of prescription opiates
has increased. The use of prescription opiates for people who are dependent on the drugs for pain
reduction has lead to an increase in abuse. When a family member or friend begins taking the drugs,
not because they need them, but because they want to feeling, it becomes an addiction. Even though
an addict is dependent on opiates, a person who is opiate dependent is different because of the
psychological, physical, and financial effects. An addict has very different psychological behaviors, ...

An opiate dependent person does not have the inability to think clearly and adapt easily into a life
without the opiates. When an addict is presented with the challenge of stopping the use of opiates, the
brain needs it more than ever before. Even long after the withdrawal symptoms are gone; an addict
must deal with post acute withdrawal syndrome (PAWS). The syndrome lasts from six to eighteen
months after the last use. It has importance to the recovering addict s ability to benefit from recovery,
treatment, function effectively on the job, interact with family and friends, and regain emotional
health. When a person becomes an addict or even dependent for a prolonged period of time, treatment
is necessary. Though there are a variety of different ways that each person can be treated. Usually a
person who is dependent is gradually taken off the opiates, by a process known as dose reduction.
Subsequently a dependent person may also have some type of therapy to help alleviate the pain at the
same time as the dose reduction. On the other hand for an addict, things are different because they
must stop using the opiates all together. Today there are many medications such as methadone,
suboxone, or subutex that doctors use to help ease the symptoms of withdrawal. Similarly the process
of dose reduction is used in this case as well. Most addicts must also enter a program like in patient or
intensive out patient rehab, where they must learn to cope with problems

The Rocking Horse-Horse Winner
In David Herbert Lawrence s short story, The Rocking Horse Winner, a boy sets out on a quest to look
for luck. His luck inevitably allows him to win money betting on horses. The money he has earned is
used in an effort to gain his mother s love. Although, he was left with was his mother s stone heart.
Lawrence displays that money doesn t buy happiness.
The question at issue in this short story is, does the protagonist, Paul, receive a sense of love,
satisfaction, and security with his single pursuit of money?
Some readers find that Paul does receive a sense of love, satisfaction, and security from his sole
pursuit of money. Religious literary critic, Charles Koban, believes that she loves Paul more than she
did her husband. If Hester truly ... Show more content on Helpwriting.net ...
Paul states, Then what is luck mother? Its what causes you to have money. Thats why its better to be
born lucky than rich. If you re rich, you may lose your money. But if you re lucky, you will always get
more money. (Lawrence 593). It is Paul s understanding that it is luck that will make him rich. After
keeping his gambling a secret, he finally admits to his mother that he s been betting at the derby and
that he is lucky. His hard work was all for nothing. After he admits his winnings, she still doesn t
acknowledge the hard work he put in to satisfy her. When a person tries hard to accomplish something
and is shut down it leaves them devastated. It is Paul s quest and drive for his mother s love that
allows him to persist in the pursuit of money. I never told you, mother, that if I can ride my horse, and
get there, then I m absolutely sure oh, absolutely! Mother, did I ever tell you? I am lucky! No, you
never did, said his mother (Lawrence 602). The eighty thousand pounds he received from his last
betting was the price of Paul s death. Paul rocking back and forth furiously and anxiously symbolizes
that he was only a laborer for Hester. His labor working and anxiety was the cause of his

The positive bacterial transformants were grown in LB ampicillin broth (100 µg mL−1) and induced with IPTG for the production of recombinant BglA enzyme. Optimal heterologous expression and efficient folding of BglA protein was observed with 0.5 mM IPTG, after 72 h incubation at 22°C. The use of reduced concentration of IPTG for getting maximum gene expression, was proved an efficient method for the production of active soluble enzyme. • 4. James Agee and Walker Evans Essay James Agee and Walker Evans Fortune Magazine, in July and August of 1936, sent James Agee and Walker Evans to research a story on sharecropping. In the preface of Let Us Now Praise Famous Men, Agee describes it as a curious piece of work. They were to produce an article on cotton tenantry in the United States, in the form of a photographic and verbal record of the daily living and environment of an average white family of tenant farmers, (IX). James Agee and Walker Evans set out to write and photograph an article for a magazine, and ended up experimenting with the form of the novel itself. James Agee was born in 1909 in Knoxville, Tennessee, but was in the urban middle class, so he had no experience with cotton farming. When ... Show more content on Helpwriting.net ... Agee and Evans set out to publish an article in Fortune Magazine, but when they brought back their work to the editors, they were rebuked. The article was over 400 pages of controversial writing and even various book editors refused to publish it as a novel. Finally in 1941 it was published by Mifflin in Massachusetts after the author was forced to take out a few outlawed words in the state. They failed in their original mission to produce an article, but undoubtedly presented a photographic and verbal record of the average white southern tenant farmer families. By living with three different families for four weeks, Evans depicted their innermost sorrows through his photographs, and Agee portrayed and emboldened their spirit by his written words. The novel itself was an experiment in form, with Evans photos all placed in the front without captions, rather than dispersed throughout the novel. Even in the text, Agee continually stops himself mid description and enters into a stream of consciousness describing the difficulties of articulating accurately the tenant families true expressions. For these reasons, it was considered a failure when it was published, but also for the same reasons it was considered an amazing success twenty years later in the 1960s. In the Preface, Agee conveys that this is a book only by necessity. More seriously, it is an effort in human actuality, in which the reader is no less centrally involved than the ... • 9. Effects Of Xenophobia In Hungary The root of this continuing and constantly high level of xenophobia can be found in the country s historical legacy. As Pál Nyíri (2003) points out when talking about the possible systemic sources of xenophobia in the country, xenophobia can have economic and cultural factors, where economic the historical factors refer to previous incidents ethnic or religious conflicts (p. 30). Neményi and Takács s (2005) findings relate to this idea: in their study they found that historical experiences can have an influencing effect on the extent of xenophobia (p. 51). From a historic point of view these theories are supported in the case of Hungary, as the country has experienced being taken over by other nations more than one time, which can explain the current fear of being occupied and the authority being taken out of Hungarian hands again. The most important and longest of these occupations were the Ottoman occupation that lasted 150 years and the Soviet occupation that lasted nearly 50 years. Show more content on Helpwriting.net ... Once the signal is received, these receivers will demodulate the signal meaning to extract the modulation from the received signal. AM AM (Amplitude Modulation) is the original way of broadcasting radio. Lee de Forest and Reginald Fessenden are given credit for the development of the technique which was carried out successfully for the first time in the mid 1870s, its frequency ranges from 535 to 1705 KHz and the typical bandwidth of an AM channel is 10 kHz. The name AM itself gives a clear indication to the type of modulation technique used; Amplitude Modulation. Amplitude Modulation is a modulation technique which works is by modifying the carrier wave s strength by increasing or decreasing its peaks in line with the variations of the input sound; the strength of the wave is known as amplitude, through all of this the frequency remains constant the same. This type of modulation technique is more susceptible to noise and or interference due to the fact noise effects amplitude directly. The change in signal strength allows the conveyance of information through the radio wave that is sent out via a transmitter and is received by a radio device otherwise known as the receiver. When the signal is received, demodulation takes place; this can be dealt with in two ways depending on the device. Either a Diode AM detector will carry out the demodulation (which is ... • 24. Summary OfThe Merger Of Two Competing Hospitals Healthcare Management Final Paper In the short story The Merger of Two Competing Hospitals , we see both Porter Regional Medical Center and Banner Regional Medical Center reach an agreement to form a merger. Both Hospitals had at least one major current flaw and by teaming up they no longer had to worry about the aspect of competing with each other. PRMC faced the problem of annually losing large sums of money. While over at BRMC the problem laid within the infrastructure and not the problem of money. After the completion of both hospitals coming together to form a newly named hospital, the question of creating an executive team in charge of management comes about. In my opinion, before moving forward in any way to create the executive team, candidates must be considered for their potential leadership. Just imagine how hard it would be to fail if the newly created hospital looked for leadership comparable to Arthur T. DeMoulas, when hiring for the executive team. While Market Basket is not a hospital, the whole idea of the managers acting as leaders would be great for a new hospital1. This could make all employees act loyal and show great customer service, and lastly having all the customers (patients) come and leave happy. According to MIT Sloan Management Review, there are six steps that should be taken to properly rebuild a management team. The first includes Reducing Role Ambiguity. This is essentially individuals who leave the new company because of the uncertainty the future holds for them. To help this, the board should do its best to ensure the company will be fine to the new executive team, as well as quickly remove any individuals who doubt the new company will be a success. Having just a few individuals who doubt the potential of the merger could quickly ruin a whole team and bring moral down. The second step states Due Diligence Around Talent Is a Dangerous Corner to Cut . To solve this, the board needs to get to know all the managers on both sides of the merger and actually find out about their full talent potential. The third part states Recognize Old Habits Die Hard But Not All Should . Each company before the merger had a certain way of doing things, and while some of those methods ... Controlling guns is not a crime prevention strategy, it only prevents the those who are physically weaker than their attackers like women, elderly, and disabled from defending themselves against bigger, faster and stronger aggressors. Since crime and gun control are serious issues,I used a white poster board for a simple background and red and black permanent markers to convey that seriousness. Since the words crimes and criminals go to together and are central to my argument, I made them both bold, in all caps, and in red so they are the most eye catching. • 27. The Race And Racial Struggle Essay Numerous scholars and historians believe that Hitler s ideas concerning race and racial struggle have originated from the philosophies of Charles Darwin, either directly or through various other sources. In today s context, the word racism means to dislike an individual or a group of individuals based on the color of their skin. However, during the mid twentieth century, the word racism meant to dislike a group of individuals because they were intellectually and/or morally inferior. Thus, it becomes the responsibility of the superior race to take control of the inferior race, or wipe them out completely in the case of Adolf Hitler. Hitler attempted to create and protect the superior race from the inferior race by implementing administrative policies designed to protect the former. This belief of superiority comes from Darwin s theory of survival of the fittest which argued that there are significant inequalities within each species and that some species get taken advantage of while others take advantage of others. //This evolutionary theory could have been used as a justification for Hitler to do something he would have done anyhow, to exterminate roughly six million Jews who, according to him, belonged to the inferior race . It is fairly clear that Darwin did not mean for his work to be interpreted the way it did; however, it can be argued that Hitler used this theory of progress through the elimination of the weak as a method to rationalize his actions to his ... This amendment was introduced after the Bruce Burrell case (2005) where, after two weeks of deliberations, all jurors were firmly in favor of one verdict except for one who was firmly against the majority verdict. The case illustrated the vulnerabilities of the previous jury system in creating a time and cost ineffective measure of trialing defendants where justice was not necessarily achieved. Another recent reform is the passing of legislation in 2006 which provided three additional spare jurors to be empanelled on lengthy criminal trials for example in the case R v. Wood. These recent reforms in legislation have led to the jury system being more cost and time effective in the criminal trial. Allowing a majority vote instead of strict unanimous verdict reduces the risk of a hung jury and hence prevents jury bribing and jury disagreements. By avoiding a retrial, the criminal trial process saves huge amounts of time and money for all individuals involved and hence maintains the integrity of the jury system. The skills one needs to be a radio listener, are more sophisticated then the skills one would use to understand and interpret a television program. When listening to the radio, one has to be an active listener. It requires you to pay more attention as the information is being processed solely through auditory stimulation. This is unlike watching television which provides both auditory and visual stimulation. Personally, when listening to Orson Welles War of the Worlds from October 30th 1938, I found myself more easily distracted comparing to when watching a television program or news story. It is much easier to pay attention to the later. Moreover, listening to the radio requires a person to use their own imagination more vividly. The audience has to take in what is being described and create their own personal picture of the event. Television seems to give us a more realistic understanding of what it is news anchor is talking about as the consumers can be shown pictures or film footage of the event being discussed. This add to the audience s view of the incident in its entirety. On the radio, the news broadcasters have to be more descriptive when describing the currents events as their descriptions are the sole foundation that the listener s imagination will be based on. Upon Mozart s first visit to Mannheim in 1763, the orchestra made a huge impression on him. Besides a large orchestra, symphonies included various specific musical features and techniques, that had become trademarks of the style developed in Manheim. One Manheim composer in particular had a large impact on the orchestra and the symphony. His name is Jan Vaclav Antonin Stamic (1717 1757) (Germanized to Johann Wenzel Anton Stamitz). He started writing orchestral works to be independent works in their own right. Orchestras no longer needed to accompany vocal genres to attain any meaning. This paved the way for what was to become known as abstract music. Stamitz wrote 58 symphonies in his lifetime, and numerous conciertos. He expanded the orchestra, with woodwinds starting to play an essential role in the orchestral timbre. The orchestra was no longer made up simply of a haphazard group of instruments, but was starting to take a fixed formal shape. Along with his contributions to the expansion of the orchestra he also adopted the formal structure of four movements for the symphony and started using a distinct development section in the first movements, which became known as sonata form.
