This document presents a protocol for a project on developing a topical liquid body wash containing extracts of Ashwagandha and Senna Alata plants for anti-acne and antimicrobial activity. The protocol outlines the need for the study, aims, objectives, methodology including extract preparation and standardization, formulation development, evaluation of antimicrobial activity, and stability studies. It also provides a literature review on previous studies related to uses of Ashwagandha, Senna Alata and other herbal extracts for skin applications and antimicrobial activity. The overall goal is to design, characterize and evaluate a polyherbal liquid body wash for anti-acne and antimicrobial properties.

2. INTRODUCTION
• Acne vulgaris (or simply acne) is an
infectious disease and one of the most
prevalent human diseases.
• Acne infection affects about 85% of
teenagers and may continue to adulthood
• Propionibacterium acnes and
Staphylococcus epidermidis are common
pus-forming microbes responsible for the
development of various forms of acne.
• Body odour of human skin usually occur
due to excessive production of sweat
glands with the presence of bacteria.
• Corynebacterium sp, Streptococcus sp,
staphylococcus sp and propionibacteria sp
are the few microbes the causes body
odour

3. • Herbal Medicinal plants like Aloe-vera,
Rosemary, Oregano, Garlic , Ashwagandha,
Senna Alata acts as antimicrobial activity.
• Withania somnifera (L.) (Ashwagandha),
belonging to the family Solanaceae.
• The secondary metabolites responsible for
microbial activity of W. somnifera are
Withanolide A and Withaferin A.
• Senna alata (L) is a medicinal plant belongs to
the family Fabaceae
• Rhein, emodin, Aloe emodin, chrysophanol and
kaempferol are some of the constituents that
acts for both bacterial and fungal activities

4. Body wash
• Body wash is used for cleansing and keep the body hygienic.
• It helps to prevent the growth of bacteria's, acnes, etc
• It helps to cleanse the impurities like oil, dirt, make up and sweat.

while F1, F2, and F3 had

was not significantly
different and stable after
five minutes of testing, and
it did not irritate the skin.

7. NEED FOR THE STUDY
Senna alata and Ashwagandha plant extracts are some of the rich sources of
secondary metabolites that are traditionally used to treat different skin associated
diseases.
Research based on Senna alata leaf extract and root extract of Ashwagandha
contents and applications were studied by several researchers, but the studies
were based on pharmacological investigations. There were less analytical
techniques performed and had limited pharmaceutical and cosmetic formulations
formulated such as creams, massage oil and bar soap.
 The Combination of Ashwagandha and Senna alata liquid body wash is not
available in the market so working on combination of polyherbal formulation of
in-vitro testing of anti- acne and anti- microbial activity, to prevent the growth of
acne causing microorganisms and odour causing bactrias
The use of plant’s pure extract in the formulation of liquid body wash has been
discussed in only handful of published papers in the clinical or pharmaceutical
fields. In the current work Ashwagandha root extract and leaf extract of Senna
alata is used to create and evaluate a topical liquid body wash .

8. Aim of the experiment:
To design, characterize and formulate topical liquid body wash containing
standardized herbal extracts of Senna alata leaf and root extract of Ashwagandha for
antimicrobial and anti-acne activity.

9. Objectives
To standardize the herbal extracts of ashwagandha and senna alata plants and its
contents for anti -acne and antimicrobial activity.
 To design and in vitro evaluate liquid body wash containing standardized herbal
extracts of ashwagandha and senna alata plants for anti-acne and antimicrobial
activity.
To perform and evaluate stability studies of prepared liquid body wash containing
standardized herbal extracts of Senna alata and Ashwagandha for anti-acne and
antimicrobial activity.

Collection of Senna alata plant leaves and Ashwagandha root powder
Extraction of Senna alata leaf and Ashwagandha root using various solvents and
concentrations by soxhlet and maceration process
In vitro evaluation of antimicrobial activity of standardized Senna alata leaf and
Ashwagandha root extract against known antimicrobial strains
Analysis of extract contents using high performance liquid chromatography (HPLC)
