1) MutMap+ is a genetic mapping method that identifies causal mutations in rice mutants without the need for crossing. It involves sequencing pooled DNA from mutant and wild-type progeny of the M3 generation derived from selfing of a heterozygous M2 plant.
2) In MutMap+, two DNA pools are made from mutant and wild-type M3 plants and sequenced. SNP indices are calculated by comparing the sequence data to the reference genome. A SNP index of 1 indicates the causal mutation, as it is fixed in the mutant pool.
