This document provides information about cosmids. It begins by defining a cosmid as a hybrid plasmid containing bacterial origin of replication and cos sites from bacteriophage lambda. Cosmids can package DNA fragments up to 45 kb in size within the phage capsid. Examples of commonly used cosmids like pJB8, pHC79 and pLFR5 are described along with their features. The advantages of using cosmids for cloning large DNA fragments to establish gene libraries are discussed. Potential disadvantages like failure of packaging recombinant cosmids if cos ends are missing are also mentioned.
