In my class we did an invasion assay using Salmonella and two different mutants. How do you tell if they were actually able to invade? Is it just if the number of cells after was greater than the control amount of cells/amount added to the wells? Solution To answer that question we will see the basic things about the invasion assay. The Invasion Assay is an in vitro system to study cell invasion of malignant and normal cells as well as the motility of bacteria. Invaded cells migrate from one compartment to another compartment through the membrane or extracellular matrices. It the number of cells that we count which passed through the porous membrane. If the wild type Salmonella and its two different mutants are showing invasion then we will be able to track them into the other compartment as they pass through the porous membrane. IF the mutant cells showing more invasion as compared to wild type then we will find more cells in the other compartment only when we take the equal number of cells for the experiment..
