Effects of pUC18 and lux Plasmids on Ampicillin Resistance of Escherichia coli
Abstract
This experiment was designed to test and observe the transformation efficacy of the
pUC18 and lux plasmids in making E. coli resistant to ampicillin. Both plasmids code
for ampicillin resistance, however, the lux plasmid codes for a bioluminescence gene
that is expressed if properly introduced into the bacteria s genome. The E. coli
cultures were mixed with a calcium chloride solution and then heat shocked,
allowing the plasmids to enter the bacteria and assimilate into the bacterial DNA.
The plasmids and the bacteria were then mixed in different test tubes and then
evenly spread onto petri dishes using a bacterial spreader, heating the spreader
between each sample to make sure there is no cross contamination. Each of the
dishes was labeled and then incubated for a period of 24 hours. The results were
rather odd because every single one of the samples grew. Several errors could have
occurred here, cross contamination or possibly an error in preparation as every
single sample in the class grew, meaning all samples of the bacteria transformed and
became ampicillin resistant.

The Alien and Sedition Acts were laws passed by the 5th Congress in 1798 in
preparation for an undeclared naval war with France, later known to be the Quasi

the French and to create a barrier against aliens and immigrants who were feared to

The focus of this essay is to explain why the International Typographic Style or Swiss
style was so important to the mid twentieth century and how it got started. Firstly, it is
an art movement that internationally began in the 1950 s, but the origins can be
traced back at least thirty years. It can be thought of as a combination of elements
from other movements such as Constructivism, De Stijland the Bauhaus. According
to Richard Hollis book Swiss Graphic Design, there are at least three factors that
account for the Swiss style (Hollis, R. 2006, P9), these being the country s location,
the language and the culture. There is evidence to prove that there are at least 2
other factors, especially for the origin of the style, which is shown in the book, along
with several others. Although not directly linked to the foundation of the style itself,
these 2 other factors include the designers and the typography, after all, it is known as
