MALDI - TOF MS: Where Mass Spectrometry Meets MicrobiologyICAR-CIFE
Abstract:
Matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS; MALDI biotyping) has turned a promising tool for the accurate, rapid, and economical identification of pathogens in the clinical diagnostics laboratory. MALDI-TOF measures the mass of molecules from a sample that has been embedded in a matrix by using a laser to ablate and desorb the molecules with minimal fragmentation. The mass spectrum is produced from the pattern (i.e., position and relative intensity) of detected m/z peaks, generating a distinct profile for a particular sample. The method is continuously being improved, and new applications for distinguishing strains, identifying metabolites or functional characteristics (e.g., antibiotic resistance), and detecting microbes directly in host samples have been developed. Adopting these methods in other disciplines than clinical diagnostics, for example, in agriculture, food safety and quality testing, or ecology, will open up new opportunities for diagnostics and research. Nowadays, MALDI-TOF MS technology is most widely used for discovering potential cancer biomarkers. MALDI-TOF MS presents advantages as it is easy to use the technique with high automation and throughput potential and high sensitivity. New fields of application for MALDI-TOF MS, such as Surface-enhanced laser desorption ionization time-of-flight (SELDI-TOF MS) and imaging mass spectrometry (IMS) are promising tools in the field of oncology.

In this presentation, Carlos Afonso describes the analysis of polymers and petroleum by ion mobility mass spectrometry and utilises novel sample introduction techniques such as the Atmospheric Solids Analysis Probe (ASAP).
ABSTRACT- L-Ascorbic acid derivatives was synthesized on treatment with acetone and acetyl chloride afforded 5,6-acetal of L-ascorbic acid then benzylation of C-2 and C-3 hydroxyl groups of the lactone ring was accomplished using K2CO3 and benzyl bromide in DMF, then deblocking of the 5,6-O,O-protected derivative of L-Ascorbic acid with acetic acid and methanol gave 2,3-O,O-dibenzyl-L-Ascorbic acid. Subsequently mono-tosylation at 6 position of 2,3-O, O-dibenzyl-L-Ascorbic acid was carried out with addition of p-toluenetosylchloride (PTSC) in Pyridine and MDC solvent medium gave 2,3-O,O-dibenzyl-6-O-tosyl-L-Ascorbic acid. All the structures were characterized by 1H NMR, 13C NMR and Mass Spectroscopy.

The study of intermolecular interactions at interfaces is essential for a number of applications, in addition
to the understanding of mechanisms involved in sensing and biosensing with liquid samples. There are,
however, only a few methods to probe such interfacial phenomena, one of which is the atomic force
spectroscopy (AFS) where the force between an atomic force microscope tip and the sample surface is
measured. In this study, we used AFS to estimate adhesion forces for a nanostructured film of poly(oethoxyaniline)
(POEA) doped with various acids, in measurements performed in air. The adhesion force
was lower for POEA doped with inorganic acids, such as HCl and H2SO4, than with organic acids, because
the counterions were screened by the ethoxy groups. Significantly, the morphology of POEA both in the
film and in solution depends on the doping acid. Using small-angle X-ray scattering (SAXS) we observed
that POEA dissolved in amixture of dimethyl acetamide exhibits a more extended coil-like conformation,
with smaller radius of gyration, than for POEA in water, as in the latter POEA solubility is lower. In AFS
measurements in a liquid cell, the force curves for a POEA layer displayed an attractive region for pH 5
due to van der Waals interactions, with no contribution from a double-layer since POEA was dedoped. In
contrast, for pH 3, POEA was doped and the repulsive double-layer force dominated. With AFS one is
therefore able to correlate molecular-level interactions with doping and morphology of semiconducting
polymers.

CINF 35: Structure searching for patent information: The need for speedNextMove Software
Chemical databases grow larger every year. Without investing in additional hardware or improved software, the time to search these databases will in turn grow longer annually. With an ever-increasing number of pharmaceutical patents, the amount of chemical data associated with these is growing at a rate with which hardware advances alone cannot keep up.
Using automated mining of U.S. and European patents, we have extracted large collections of structural data in the form of reactions, mixtures, and exemplified compounds. Additional information such as protein targets and diseases are also extracted from each patent and associated with the structural data. We will describe how this data can be queried with natural language phrases and how these phrases are interpreted as structural queries.
Through innovations in substructure and similarity search algorithms, it is possible to search and retrieve hundreds of millions of chemical records in fractions of a second. We will demonstrate how this is achieved on a regular desktop machine using just-in-time and ahead-of-time compilation techniques.

Following the treatment of methyl parathion there was a differential percentage in increase or decrease of different isozymes in the fish Labeo rohita. There were three distinct bands during the 96h of treatment almost parallel to LDH5 band of human serum suggesting liver damage. This was also confirmed by histopathological studies.

Protective Effects of Alpha Lipoic Acid (α -LA) Against Lead Neuro-Toxicity i...inventionjournals
Aim of the work: The present study was conducted to elucidate the possible protective effect of alpha lipoic acid (α-LA) against the deleterious effect perturbation induced in rat brain exposed to lead acetate. Methods: 32 Wistar male rats (weighing 130 ± 10 g) were divided into four groups (n=8): (1) normal control group (C); (2) Initiation group (Pb as lead acetate 20 mg/kg.b.wt, i.p. for 2 wks); (3) treatment group (α-LA 20 mg/kg.b.wt, i.p. for 3 wks); (4) post-initiation treatment group (Pb for 2 wks then followed by α-LA for 3 wks). Levels of monoamines (norepinephrine NE and dopamine DA), the level of Ache activity and finally adenosine triphosphate (ATP), were estimated in the hippocampus and cerebral cortex, in addition, a Morris water maze and the histological study were performed after completion of the experiments. Results: The results of the present work demonstrated that Pb inhibited neurotransmitters releases and decrease the level of Ache activity, as well as it inhibited energy production ATP. Pb impaired performance on Morris Water Maze of rats and histological degeneration. However, treatment with α-LA significantly attenuated the behavioral impairment and biochemical parameters in rat treated with Pb. And amelioration of histological changes. Conclusion: As a conclusion, treatment with α-LA can improve the Pb-induced toxicity via antioxidant activity.

Degradation of an organophosphorus insecticide (chlorpyrifos) in simulated wa...Salah Hussein
Induced degradation of chlorpyrifos insecticide in simulated wastewater with advanced oxidation processes (AOPs), using ultraviolet irradiation (UV), ozonation and chemical oxidation using (sodium hypochlorite, calcium hypochlorite, monochloride-isocyanuric acid (MCICA), dichloroiso-cyanuric acid (DCICA), trichloroisocyanuric acid (TCICA) ) was studied. Chlorpyrifos and its degradation products were extracted using solid phase extraction (SPE) method, identified using GC-MS. Results showed that the degradation of chlorpyrifos in simulated wastewater followed the first order reaction, and its half life was 3.34, 5.64, 7.13 and 10.69h under ozonation, UV, 1.5%TCICA and 1.5%DCICA respectively when chlorpyrifos solutions treated for 12 h. The concentrations of chemical oxidative substances, active chlorine content and time of treatments had a significant effect on degradation rate of chlorpyrifos, which increased with increasing of each. The most enhancement of chlorpyrifos degradation was observed in treatment with ozonation, UV, TCICA and DCICA where the dissipations % of the parent compounds were 85.70, 57.71, 43.71 and 35.07 %, respectively. The intermediates products of chlorpyrifos degradation using chemical method were identified as O,O-Diethyl thiophosphate(DEP), 3,5,6-trichloro-2-pyridinol(TCP), 3,5,6-trichloro-2-methoxypyridine(TMP) and 2,3,5,6-tetrachloro-pyridine. UV leads to formation of O,O-Diethyl phosphate, TCP and Chlorpyrifos oxon. Ozonation leads to formation of O,O-Diethyl thiophosphate beside the UV degradation products.

The US House of Representatives is deeply concerned by ongoing and pervasive acts of antisemitic
harassment and intimidation at the Massachusetts Institute of Technology (MIT). Failing to act decisively to ensure a safe learning environment for all students would be a grave dereliction of your responsibilities as President of MIT and Chair of the MIT Corporation.
This Congress will not stand idly by and allow an environment hostile to Jewish students to persist. The House believes that your institution is in violation of Title VI of the Civil Rights Act, and the inability or
unwillingness to rectify this violation through action requires accountability.
Postsecondary education is a unique opportunity for students to learn and have their ideas and beliefs challenged. However, universities receiving hundreds of millions of federal funds annually have denied
students that opportunity and have been hijacked to become venues for the promotion of terrorism, antisemitic harassment and intimidation, unlawful encampments, and in some cases, assaults and riots.
The House of Representatives will not countenance the use of federal funds to indoctrinate students into hateful, antisemitic, anti-American supporters of terrorism. Investigations into campus antisemitism by the Committee on Education and the Workforce and the Committee on Ways and Means have been expanded into a Congress-wide probe across all relevant jurisdictions to address this national crisis. The undersigned Committees will conduct oversight into the use of federal funds at MIT and its learning environment under authorities granted to each Committee.

The Committee has broad jurisdiction over postsecondary education, including its compliance with Title VI of the Civil Rights Act, campus safety concerns over disruptions to the learning environment, and the awarding of federal student aid under the Higher Education Act.

Artificial Intelligence (AI) technologies such as Generative AI, Image Generators and Large Language Models have had a dramatic impact on teaching, learning and assessment over the past 18 months. The most immediate threat AI posed was to Academic Integrity with Higher Education Institutes (HEIs) focusing their efforts on combating the use of GenAI in assessment. Guidelines were developed for staff and students, policies put in place too. Innovative educators have forged paths in the use of Generative AI for teaching, learning and assessments leading to pockets of transformation springing up across HEIs, often with little or no top-down guidance, support or direction.
This Gasta posits a strategic approach to integrating AI into HEIs to prepare staff, students and the curriculum for an evolving world and workplace. We will highlight the advantages of working with these technologies beyond the realm of teaching, learning and assessment by considering prompt engineering skills, industry impact, curriculum changes, and the need for staff upskilling. In contrast, not engaging strategically with Generative AI poses risks, including falling behind peers, missed opportunities and failing to ensure our graduates remain employable.
