MVPK cell cultures showed cytopathic effect when inoculated with the virus. Animals inoculated with the virus showed increased rectal temperatures within 24 hours and vesicular lesions at inoculation sites by 48 hours, preceded by fever. Complement fixation and serum neutralization tests were performed on convalescent serum samples to detect antibodies against the virus. RT-PCR and agar gel precipitation tests were also conducted but not described in detail. References included several textbooks on veterinary virology, bacteriology, and the diseases of swine.

3. Complement
Fixation test
A standard CF test (15) was done, round-bottom, plastic,
microtiter plates. Four hemolytic units of complement
were used in the test and antigen titrations were
conducted in triplicate. Antigen was reacted for 30 min
with diluted test sera at 370 C. Sensitized sheep red
blood cells were incubated with the reactants for 45 min
at 37°C with agitation. Plates were centrifuged at 600 x
g for 5 min prior to reading the amount of hemolysis.
Serum neutralization tests were conducted using
the 12 convalescent antisera to VES virus. Virus
neutralization indices (VNI) were calculated by
using the system Used for SN but with a
constant1/10 Serum dilution and varying tenfold
Dilutions of virus. Neutralization indices for the sera
were expressed as logio of virus neutralized.
