Paratuberculosis is a contagious, chronic and sometimes fatal infection that primarily affects the small intestine of ruminants.

Dr. Raluca Mateescu does research in the area of beef cattle, sheep and goat molecular genetics. To address consumers' demand for high-quality products, the beef industry needs to use all tools at its disposal to deliver a product with superior eating quality and implementation of an effective genetic improvement program is a critical step toward this aim. Palatability is not based only on one characteristic but is based on the relationship of three characteristics - tenderness, juiciness, and flavor – and how they work or do not work together to determine beef eating satisfaction. These traits are impractical to improve through traditional selection (difficult and expensive to measure) but are ideal candidates for genomic selection if genetic markers that account for a worthwhile proportion of the variation could be identified.
In this webinar, Dr. Mateescu will focus on the use of SVS program to perform genome-wide association studies for individual traits describing beef palatability, identify chromosomal regions associated with these traits, estimate genomic breeding values and predict the accuracy of GEBV for palatability traits in beef.

Following the Food Standards Agency’s (FSA) announcement in January that horse and pig DNA had been identified in beef products sold by several supermarket chains, further testing across Europe and beyond has revealed widespread incidences of such contamination.1 However, most testing methods are based on detection of species-specific DNA in meat, using the polymerase chain reaction (PCR) – which does not detect or identify proteins. This is a concern because DNA can be easily disrupted or removed during standard meat processing and food manufacturing. As a result, horse tissue or other contaminants remain undetected in food samples, despite strong presence of the contaminating proteins. An alternative protein-based method, ELISA (enzyme-linked immunosorbent assay), can be used to complement DNA testing, but this method has limitations, including that it detects only one part of the protein and not multiple protein markers.
The LC-MS/MS-based method presented offers a more accurate and reliable approach to meat speciation than PCR or ELISA-based techniques or other indirect methods, and also allows for the detection of veterinary drug residues in the same analysis, which is not possible by ELISA or PCR.
The method was developed using an Eksigent ekspert™ microLC 200 UHPLC system coupled with a SCIEX QTRAP® 5500 LC/MS/MS system. The method uses multiple reaction monitoring (MRM) to detect peptide markers for horse and is capable of providing sequence information by acquiring an enhanced product ion (EPI) scan for each triggering MRM which can be used to further confirm the peptide’s / proteins and therefore the species identity. This gives greater confidence for food testing when distinguishing between species; for example horse and beef proteins may differ by as little as one or two amino acids.
At the same time it is also possible to detect and quantify veterinary drug residues using the same extraction method and LC conditions by simply adding additional MRM transitions to the method. Here the nonsteroidal anti-inflammatory drug (NSAID) BUTE was detected in meat samples.

■ These techniques are mainly based on the principles of antigen
antibody reactions. The homologous antigen binds with the antibody
which is visualized by various methods. These techniques are simple and
can be performed any where with little efforts. These tests are also
known as Serological or Immunological methods.

■ It is also known as Ring Precipitation Test (RPT) in which homologous
antigen and antibodies reacts together and make a ring at the point of
its interaction.
■ Known antibodies (blood serum) collected from the treated
experimental animals are mixed in a test tube with the filtered extract
of meat samples.
■ If precipitation line forms at the point of mixing then it indicates positive
test.
■ For example if the antiserum of horse mixed with filtered extract of
suspected meat in a test tube, a turbidity occurs and forms a definite
precipitation ring if sample is positive for horse flesh.

■ The protein from a meat extract can be separated by electrophoresis in
a carrier gel. A pattern of Protein bands is obtained, which can be
visualized by staining. The separation of proteins by gel electrophoresis
is based on their charge, size and molecular weight.
■ Iso-Electric Focusing (IEF) is a more recent technique, in which proteins
are sapared on the basis of differences in Iso-Electric points or pH (l). For
this purpose, a pH gradient is established. It takes advantage of the fact
that overall charge on a molecule is a function of the pH of its
surroundings.
