1 Introductions
Biocatalysis is the use of living systems or their components to accelerate chemical
reactions. In the process of biocatalysis, natural catalysts (such as enzymes)
chemically transform organic compounds. These two enzymes that have been
more or less isolated and those that are still present in living cells are both used.
The process of organic synthesis using natural or modified enzymes is called
chemical enzymatic synthesis. Reactions performed by enzymes are classified as
chemical enzymatic reactions.
2 Asymmetric Biocatalysis
The use of biocatalysis to obtain enantiopure
compounds can be divided into two different
methods:
1.Kinetic resolution of a racemic mixture
2.Biocatalyzed asymmetric synthesis
In kinetic resolution of a racemic mixture, the presence of a chiral
object (the enzyme) converts one of the stereoisomers of the
reactant into its product at a greater reaction rate than for the other
reactant stereoisomer. The stereochemical mixture has now been
transformed into a mixture of two different compounds, making
them separable by normal methodology.
3 Asymmetric Biocatalysis
Biocatalyzed kinetic resolution is utilized extensively in the
purification of racemic mixtures of synthetic amino acids. Many
popular amino acid synthesis routes, such as the Strecker Synthesis,
result in a mixture of R and S enantiomers. This mixture can be
purified by (I) acylating the amine using an anhydride and then (II)
selectively deacylating only the L enantiomer using hog kidney
acylase.
4 Significance of Biocatalysis
The natural, green biological catalysts attract
fast growing attentions for many benefits,
such as sustainability, high efficiency, and little
waste footprint. However, developing and
setting up such systems could be challenging,
given that the nature of these biocatalysts is
distinct from traditional ones. Furthermore,
maintaining a biocatalytic mechanism
performing as a stable and consistent manner
also requires engineering design and
development on the whole system, in
addition to the molecular biology and
biochemistry techniques that are essential to
start feasibility evaluation on the biological
catalysts.

  • 2. 1 Introductions Biocatalysis is the use of living systems or their components to accelerate chemical reactions. In the process of biocatalysis, natural catalysts (such as enzymes) chemically transform organic compounds. These two enzymes that have been more or less isolated and those that are still present in living cells are both used. The process of organic synthesis using natural or modified enzymes is called chemical enzymatic synthesis. Reactions performed by enzymes are classified as chemical enzymatic reactions.
  • 3. 2 Asymmetric Biocatalysis The use of biocatalysis to obtain enantiopure compounds can be divided into two different methods: 1.Kinetic resolution of a racemic mixture 2.Biocatalyzed asymmetric synthesis In kinetic resolution of a racemic mixture, the presence of a chiral object (the enzyme) converts one of the stereoisomers of the reactant into its product at a greater reaction rate than for the other reactant stereoisomer. The stereochemical mixture has now been transformed into a mixture of two different compounds, making them separable by normal methodology.
  • 4. 3 Asymmetric Biocatalysis Biocatalyzed kinetic resolution is utilized extensively in the purification of racemic mixtures of synthetic amino acids. Many popular amino acid synthesis routes, such as the Strecker Synthesis, result in a mixture of R and S enantiomers. This mixture can be purified by (I) acylating the amine using an anhydride and then (II) selectively deacylating only the L enantiomer using hog kidney acylase.
