Apert syndrome is a genetic condition characterized by premature fusion of skull bones resulting in an abnormal head shape. It is caused by mutations in the FGFR2 gene and is inherited in an autosomal recessive pattern. Symptoms include skull abnormalities, fusion of the fingers and toes, and sometimes intellectual disabilities. While there is no cure, treatments focus on corrective surgeries to separate skull bones and digits.

Ankyloglossia, also known as tongue-tie, is a congenital oral anomaly that may decrease mobility of the tongue tip and is caused by an unusually short, thick lingual frenulum, a membrane connecting the underside of the tongue to the floor of the mouth. Ankyloglossia or tongue-tie is the result of a short, tight, lingual frenulum causing difficulty in speech articulation due to limitation in tongue movement. Ankyloglossia is a congenital condition in which a neonate is born with an abnormally short, thickened, or tight lingual frenulum that restricts mobility of the tongue. Ankyloglossia may be associated with other craniofacial abnormalities, but is also often an isolated anomaly.

IRF6-related syndrome is a group of cleft lip and palate disorders including Van der Wood Syndrome (VWS) and Popliteal Pterygium Syndrome (PPS). People with VWS are at the lowest end of the spectrum. They can have cleft lip or palate or cleft palate or a combination of these abnormalities. People with PPS usually have cleft lip, cleft lip, or cleft palate, with additional skin and limb abnormalities, including dark skin on the back of both feet (papillary) and between the legs (between the veins), deformity, or abnormalities of the device. Ethanol (CH3CH2OH), or beverage alcohol, is a two-carbon alcohol
that is rapidly distributed in the body and brain. Ethanol alters many
neurochemical systems and has rewarding and addictive properties. It
is the oldest recreational drug and likely contributes to more morbidity,
mortality, and public health costs than all illicit drugs combined. The
5th edition of the Diagnostic and Statistical Manual of Mental Disorders
(DSM-5) integrates alcohol abuse and alcohol dependence into a single
disorder called alcohol use disorder (AUD), with mild, moderate,
and severe subclassifications (American Psychiatric Association, 2013).
In the DSM-5, all types of substance abuse and dependence have been
combined into a single substance use disorder (SUD) on a continuum
from mild to severe. A diagnosis of AUD requires that at least two of
the 11 DSM-5 behaviors be present within a 12-month period (mild
AUD: 2–3 criteria; moderate AUD: 4–5 criteria; severe AUD: 6–11 criteria).
The four main behavioral effects of AUD are impaired control over
drinking, negative social consequences, risky use, and altered physiological
effects (tolerance, withdrawal). This chapter presents an overview
of the prevalence and harmful consequences of AUD in the U.S.,
the systemic nature of the disease, neurocircuitry and stages of AUD,
comorbidities, fetal alcohol spectrum disorders, genetic risk factors, and
pharmacotherapies for AUD.
