In this microscopy, the specimen is brightly illuminated while the background is dark. It is one type of light microscopes. Used to examine live microorganisms that are not visible with ordinary light microscope.

5.  Dark-field microscope is a machine or matter that can be used for
the observation of living, unstained cells and microorganisms. In
this microscopy, the specimen is brightly illuminated while the
background is dark. It is one type of light microscopes.
 Used to examine live microorganisms that are not visible with
ordinary light microscope.

6.  It creates a contrast between the object and the
surrounding field Such as: the background is dark and
the object is bright.
 The objective and the ocular lenses used in the dark
ground microscope are same as that of ordinary light
microscope.
 Special condenser is used, which prevents the transmitted
light from directly illuminating the specimen.

7. Advantages of Dark-Field Microscopy
1. Resolution by dark-field microscopy is somewhat better
than bright-field microscopy
2. Improves image contrast without the use of stain, and thus do
not kill cells.
3. Direct detection of non-culturable bacteria present in patient
samples.
4. No sample preparation is required
5. Requires no special set up, even a light microscope can be
converted to dark field.

9. Disadvantages
A dark field microscope can result in beautiful and amazing images; this technique
also comes with a number of disadvantages.
 First, dark field images are prone to degradation, distortion and
inaccuracies.
 A specimen that is not thin enough or its density differs across the
slide, may appear to have artifacts throughout the image.
 The preparation and quality of the slides can grossly affect the
contrast and accuracy of a dark field image.
