TTI screening testing methods have evolved through four generations. Serological tests include rapid tests and ELISA. There are four main types of ELISA - direct, indirect, sandwich, and competitive/inhibition. Sandwich ELISA has the highest sensitivity but requires optimization of antibody pairs. Competitive ELISA can measure antigen or antibody concentration using crude samples. Chemiluminescence immunoassay (CLIA) is similar to ELISA but uses a chemiluminescent substrate, making it ultrasensitive. CLIA requires a luminometer for detection while ELISA uses color change.
