GROWTH HORMONE
Somatotropin
From somatotrophs
Protein hormone
190 amino acids
Growth, metabolism and drug
CONCEPT
Every protein has mRNA so as DNA
cloning
STEPS
hGH isolated from pituitary gland.
Gene is cleaved using restriction
endonuclease.
Add gene to a plasmid vector (carrier
molecule) by cleaving using ligase.
Construct with desired gene is obtained called
rDNA.
Plasmid under self replication.
Introduced into E.coli cell.
PROCEDURE
Synthesis of first 24 nucleotides chemically
with known sequence. (8X3=24)
Isolate growth hormone from human pituitary
gland.
cDNA is formed using reverse transcription-
gene for hGH production.
First 24 sequence is removed using restriction
endonuclease.
Synthetic gene and cDNA are joined using T4
ligase.
Suitable vector is chosen.
DNA joined to plasmid by endonuclease and
ligase.
Kept at Lac operon- operator region.
Selection of host cell : E.coli.
Recombinant E. coli isolated from culture and
mass production by fermentation technology to
APPLICATION
oGH deficiency treatment
oAthletics supplement
oGrowth deficiency
oTumer’s syndrome and chronic renal
insufficiency treatment
oRenal carcinoma treatment
oBovine somatotropin - increases milk
production in lactating cows
ADVANTAGES
Time conservation
Cost effective
Cheap
More copies due to recombinant technology
LIMITATION
Transformation of diseases.
