  2. We will cover some basic facts, protein expression, and LBP’s expected behavior in Western blot. All information in this video is based on public information and no proprietary experimental evidence was used.
  3. Topic 1: Quick facts about LBP. The Lipopolysaccharide-binding protein, or LBP, is an acute-phase protein of the innate immune system. It serves a vital role against bacterial infections. It binds to the lipopolysaccharide, found in the outer cell wall of the gram-negative bacteria to generate an immune response. The precursor of LBP is 481 amino acids long, of which a signal peptide of 25 amino acids is cleaved to form the active LBP of 456 amino acids. LBP is expected to run about 53 kilodaltons on Western blot.
  4. Topic 2: Where is LBP expressed? High levels of LBP RNA expression have been detected in the liver, especially in hepatocytes. LBP is also expressed at lower levels in the adipose tissues, skeletal muscles, appendix, pancreas, gastrointestinal tract, lungs, and endocrine tissues. Furthermore, it is detected in blood serum. You can find more information at protein atlas dot org including which cell lines express LBP and which ones don’t. This information will come in handy when designing positive and negative controls for western blot and immunohistochemistry.
  5. Topic 3: LBP in Western blotting. Antibody specificity can make or break an experiment. After looking through Western blot images from publications and antibody companies, we can see that typically one band can be expected in an LBP Western blot. The expected band size is around 53 kilodaltons, but it can run up to 67 kilodaltons. Check out the PMC link for more images, which is also provided in the description box.
  6. Topic 4: Some interesting facts about LBP. LBP has an antibacterial role in the innate immune system. It interacts with other proteins, such as CD14, TLR2, and TLR4, while bound to LPS to trigger a pattern-recognition event that will elicit an immune response in the host. Due to the short half-life of plasma LPS, LBP is used as a biomarker to measure the LPS plasma concentration instead of LPS itself.
