2. I LISOSOMI
• I lisosomi sono vescicole digestive delle cellule
animali, dove avviene un processo di digestione e
riutilizzo di componenti cellulari come proteine,
mucopolisaccaridi o glicolipidi.
• Queste sostanze vengono metabolizzate da enzimi o
strutture proteiche con funzione specifica (es. proteine
di trasporto).
• Il deficit di uno di questi sistemi porta ad un accumulo
nei lisosomi del relativo substrato.

4. LE MALATTIE LISOSOMIALI
• Le malattie lisosomiali sono il risultato di un
deficit ereditato geneticamente della funzionalità
(quantitativa/qualitativa) di specifici enzimi o
proteine d trasporto intralisosomiali.
• Attualmente si conoscono 50 patologie così
classificabili
• L’incidenza complessiva è calcolata in circa
1:8000 nuovi nati, con un’aspettativa di vita varia

50. Il dolore nella malattia di Gaucher tipo 1 non è
solo dolore osseo
La malattia di Gaucher tipo 1 presenta un elevata prevalenza di
dolore neuropatico periferico correlata a sofferenza delle fibre
nervose di piccolo calibro.
Anche i pazienti non in terapia presentano neuropatia delle
piccole fibre e dolore neuropatico che rende meno probabile il
possibile danno iatrogeno (ERT-related)
La neuropatia dovrebbe essere presa in considerazione quale altra
manifestazione neurologica nel GD-1 e valutato il trattamento.
La distribuzione dei sintomi e del danno neuropatico potrebbe
correlarsi a un danno principale a livello dei gangli delle radici
dorsali.
