Introduction
 Flavonoids are group of natural substances with variable
phenolic structures.
 Are found in fruits, vegetables, grains, bark, roots, stems,
flowers, tea and wine.
 These are secondary metabolites of plants.
 Polyphenolic 15 carbon containing compounds.
 Flavonoids regulate plant development, pigmentation,
and UV protection.
 And roles in defence and signalling between plants and
microorganisms.
Structure of Flavonoids
Flavonoid is 15 carbon
compound.
C6C3C6
Two rings are aromatic and
middle ring contain oxygen.
Basically flavonoids are
polyphenolic compounds.
Isolation and purification of flavonoids
 Powdered plant material is extracted with various solvents.
 Various solvents are used according to types of flavonoides like
CHCl3 , Diethyl ether, ethyl acetate etc.
 All solvent used for extraction are semipolar or nonpolar.
 More polar flavonoids are extracted with Alcohol or aqueous
alcohol.
 Fractional crystallization is employed for purification of extract.
Genaral extraction
method: Powdered plant material
Extraction with boiling water
Aqueous extract
Trated with lead acetate
Precipitation with tannin Supernatant liquid containing
flavonoids
Dilute with water + HCl
+ boil for 1 hour
Precipitate of sugar free
flavonoid
Ppt extract with alcohol + purified
with fractional crystallization
