In this experiment individuals were given a plate with two unknown specimens. Each
group completed a gram stain followed by a series of other test in order to identify
the unknown bacteria. Each student conducted separate testing s on one unknown in
order to specifically identify the strain.
Materials and Methods
First and foremost the gram stain is conducted to differentiate gram positive bacteria
from gram negative bacteria due to their cell structure. The thick layer of
peptidoglycan in gram positive bacteria allows the crystal violet dye to retain a
purple color, whereas, the thin peptidoglycan in gram negative bacteria is
decolorized and counter stained with safranin red in order to be viewed in the
microscope. (1) The gram stain procedure begins with a clean slide and a sterile
loop. A drop of water is applied to the slide followed by a pinhead size of unknown
bacteria. The slide air dries and then is heat fixed with a Bunsen burner.
Subsequently, the crystal violet is applied to the smear, followed by the mordant,
Gram s iodine. Next, the decolorizer is applied then washed. Finally safranin is
applied to the smear and also washed. The slide is blot dried with bibulous paper
and ready for the microscope. However, to ensure a good visual, oil immersion is
applied to increase the resolution. Following the gram stain is the catalase test, which
is utilized to determine if a specimen is streptococcus or staphylococcus. A sample of
bacteria is placed on a slide and combined with a few drops of peroxide. For further
testing, a blood agar plate is inoculated for rapid identification methods for
Streptococcus. Each strain of Strep. is sensitive to only one test on the rapid I.D
method, therefore enabling a process of elimination. The procedure is as follows,
forty percent of the unknown is applied to the top portion of the agar. Subsequently,
three antibiotic discs, Bacitracin A disc, Optochin P disc, and SXT disc are situated
along the unknown bacteria. Afterwards, a horizontal line of Staph. Aureus is drawn
along the bottom of the agar. Next, a vertical line is drawn from the unknown

Creativity and art is becoming such a rising normality in our everyday culture. A
new mural with a political statement is popping up every other week. A person
visiting the local pizza joint downtown can look up on the wall and see a piece of
art with a price tag next to it. A local artist painted it and where else to get public
attention than Brooklyn Pizza, in the heart of buzzing downtown. The art scene is
growing quickly, and thanks to the creative means, so is the opportunity for

showing U of A pride, local murals and art are popping up and displaying the

report on, almost entirely because of the unique concept and presentation behind it.
It was created by film director J.J. Abrams and written by author Doug Dorst. While
S. is the name of the project, the story is contained in the form of a fictional book
called Ship of Theseus, written by the prolific, incendiary, and ever mysterious author,
V.M. Straka. What makes the story of S. so unique is that while the novel is a
traditional narrative, the margins of the book are filled with scribbles, writings,
notes, clues, musings and revelations from two readers, Jen and Eric. Jen, a college
senior, and Eric, a former grad student and Straka researcher.

important in the scientific community. The GI tract plays a key role in glucose
homeostasis. The GI tract becomes inflamed in the presence of diabetes mellitus
causing various complications. ABCA1 and ABCG1 play important parts in glucose
homeostasis, insulin secretion, and cell survival. However, these transporter proteins,
in normal conditions, are overshadowed by other molecules with similar functions.

Importance of Communication
Glossophobia is the fear of public speaking. The average person ranks the fear of
public speaking higher than the fear of death. According to the National Institute of
Mental Health over 74% of people suffer from speech anxiety (Brain, 2016) . This is
a sad statistic being that communication is the backbone of our society. It is what
allows us to connect with others. Without communication skills, we would not be
able to form relationships, whether in our personal or professional lives. We, as a
society, tend to think that communication is only verbal. However, communication
includes nonverbal, written, even listening, as well as our body language. In fact,
over 70% of our communication is nonverbal. In this class I hope to improve on my
interpersonal communication by learning ways to overcome my fear and setting new
standards in the way I communicate with others. In my personal life, improper
communication has led to more than one situation that caused hurt to someone I love.
Likewise, I have been on the receiving end of hurt directly as a result of poor
communication.
Currently, if I were to rank my comfortability in speaking in front of the class, I
would say it would be a 4 out of 10. I know many people would think it would be
higher, but the fact of the matter is I have an anxiety when it comes to public
speaking. Being an extroverted personality often leads people to believe I would
have no problem speaking in public, but when the spotlight is on, it is an entirely
different story. The moment I know I must get up in front of a group of people, I
start to feel dread, my stomach goes into knots, my heart starts to race, and my
palms get clammy. I always try to be the first speaker so that these feelings of
dread can disappear sooner than later. Ironically, after I finish speaking (assuming it
went well) I feel a sense of accomplishment. There is a remarkable sense of pride
that comes with effectively communicating to your peers and being well received.
My hope is that by taking this Communication 100, I will find new ways to
effectively communicate, which will not only help me grow as a person, but will

Understanding your passion is a concept that the author uses to allow you, as a future
business leader, to think about the traits that will transform your business from an
ordinary, or good business to a great business. The thought process of believing in
what your business is to what it can be .
This text is primarily focused around what thoughts and actions can turn you
business from good to great using his metric of sustaining a profitable business in a
15 year increment or longer. If you look at a few of the businesses he references,
some have had ill effects in years after their profitability, such as Circuit City failing
and Fannie Mae causing catastrophic economic collapse in the recent day. A
company is only as good as their leadership and profitability remain intact.
A company, to remain profitable through the longevity of world commerce can be
difficult unless you continually stove to meet consumer demand for your product.
The internet businesses such as Amazon have decimated some businesses because of
low cost and availability and have only succeeded because of companies inability to
adapt to this new business model.

A Take on Tattoos in Modern Society
During the last 20 years, and especially during the last ten to eight years, there has
been a dramatic increase in the popularity of tattoos, which are in demand by all
classes of society today (Serup 15). It is no lie to state that there is a visible
popularity surge in tattoos from the last decade, and the number continues to rise.
One of the possible explanations for this increase might be that tattoos have evolved
to become part of the mainstream media or trend which mainly sprouted from the
middle class. Other explanations have said that tattoos reflect the time and society
and change when it occurs, but also self expression. One thing for certain amidst all
the explanations is that tattoos hold a significant element in redefining society in its
control and setting of norms. In an interesting journal excerpt, Why Do People Get
Tattoos? it explains that there have been perceptions of tattooed women having
transient sexual relationships and thus being labeled as being from the lower class
throughout history. The article continues to delve deeper into the subject by giving
an example of a rape case which was dropped by a lawyer due to the victim having a
tattoo (Kang, Jones 352). It is interesting to see the negative connotations some have
of tattooed women because they do not fit into the society s ideal standard of
femininity and immediately think it lessens the woman s class, even to the point
where they would not defend them in a court case. Although many would like to
think that tattoos have provided women an opportunity to challenge the said
traditional standards of femininity, not all believe so. On the contrary, some believe
that women use their tattoos to enforce these standards rather than empower
themselves. Similarly, men have also shown to use their tattoos to reinforce
traditional notions of masculinity (Kang, Jones 352). The debate can go either way,
but mainly it depends on the individual and their reason to get inked in the first place.
This article conveys the relevance of why women and men get tattoos and shows
