Abstract
This experiment was designed to test and observe the transformation efficacy of the pUC18 and lux
plasmids in making E. coli resistant to ampicillin. Both plasmids code for ampicillin resistance,
however, the lux plasmid codes for a bioluminescence gene that is expressed if properly introduced
into the bacteria s genome. The E. coli cultures were mixed with a calcium chloride solution and
then heat shocked, allowing the plasmids to enter the bacteria and assimilate into the bacterial
DNA. The plasmids and the bacteria were then mixed in different test tubes and then evenly
spread onto petri dishes using a bacterial spreader, heating the spreader between each sample to
make sure there is no cross contamination. Each of the dishes was labeled and then incubated for
a period of 24 hours. The results were rather odd because every single one of the samples grew.
Several errors could have occurred here, cross contamination or possibly an error in preparation as
every single sample in the class grew, meaning all samples of the bacteria transformed and became
ampicillin resistant.

On December 26th, 2015, my mother and I went to go see the show, Baskerville, by Ken
Ludwig, at the Suzanne Roberts Theatre in Philadelphia. The play was a different and clever
take on the mystery novel, The Hound of the Baskervilles, by Sir Arthur Conan Doyle. The play
the book tell the story about an attempted murder of a man called Sir Henry Baskerville, who has
just inherited his uncle, Sir Charles Baskerville s estate. There is some suspicion surrounding Sir
Charles death. It is said that he died of a heart attack, but his friend who found him dead, Dr.
James Mortimer, noticed an expression of fear on his face and the prints of gigantic hound nearby.
So the doctor brings the case to Mr. Holmes and it begins.
The venue of the show was the Suzanne Roberts Theatre in Philadelphia. It was a very sleek and
modern looking place. It was smaller than I imagined outside and inside of the theatre. The stage
was a perfect square black box type stage, completely surrounded by lights. The show was done by
the Philadelphia Theatre Company, and the show was written by American playwright Ken
Ludwig. The contemporary drama show premiered at the Arena Stage in Washington, DC on

It never occurred to me before to take some time and evaluate my reading process and my reading
habits. I ve done tons of readings throughout my whole life, including articles and abstracts for
school and just some novels and fiction books for entertainment. However, I barely paid attention at
any of my readingroutine. All I was concentrated on were either getting the information I need
from the text or, in case of a novel, enjoying the plot. Up until recently I had no idea if the way I
was readingis productive or not, but after accomplishing my assignment for English class to read
the texts from On Creative Reading , I tried to look at my reading process from a different angle.
I noticed that some writing styles make it harder for me to read, concentrate and follow the idea
of the text, if the writing consists of very long phrases and is overloaded with some difficult
expressions and lots of unknown terms. On the other hand, if the author tries to keep the
phrases more or less simple and clear, tries to hold my interests and makes the whole thing easy
to follow, all those things make my reading process more pleasurable. Surely, the topic I m
researching plays a prominent role in the level of pleasure I m receiving during the reading. I m
more interested in reading some articles on photography or psychology rather than finance or
physics. I would enjoy more reading some fiction than some assignments for school. Richard E.
Miller and Ann Jurecic, the authors of Habits of the Creative Mind, point out that reading
process is not the same for everyone. They ask questions like Once the process of reading has
been routinized and internalized, why is that different people reading different material reach

what many people struggle with. People have a hard time forgiving other people but more often
themselves. Amir knows just how hard it is to forgive yourself in The Kite Runner by Khaled
Hosseini. When Amir makes the decision to not speak up about Hassan he felt so much guilt that
he wanted Hassan and Ali to leave, he regretted it instantly. Years later, Rahim khan called Amir
and asked him to come back to Afghanistan where he found out Hassan was his half brother. Amir
finally forgave himself when he found a way to make up for his mistakes. Hosseini portrays Amir
as a morally ambiguous character by his guilt and inability to forgive himself for what he did to
Hassan.
Morally ambiguous characters become challenged by a dilemma that will test their mental
strength. Amir s choice to not speak up about what happened shows his betrayal to his friendship

She looks so normal , this is a typical response that a mother often hears about her child with
special needs. Special is a term that is described as different . Some people may look at a child
with special needs and feel sympathy while others lack the knowledge. For my child
development class, I was assigned to conduct an interview with a mother who has a special
needs child. I was one of the many that lack knowledge of special needs children because I did
not know what this mother had to face. While doing this interview I have learned that it takes a
special person to take care of a special child. This child has a disease called NOMID, which stands
for neonatal onset multisystem inflammatory disease, this disease causes inflammation and tissue
damage affecting the nervous system, skin, and joint. This interview has taught me that some
parents will face many difficulties while raising a special needs child, but they will do whatever it
takes to help the child overcome challenges that they may face everyday.
On January 20, 2016 I interviewed Jasmine, a good friend of mine about her daughter Emily that
has special needs. She discussed the many challenges she has had to face. This interview took
