Since tissue-tissue interactions in the human body can play a significant role in determining the success of new pharmaceuticals, the development and use of multi-organ micro-devices present an opportunity to improve the drug development process. The devices have the potential to predict potential toxic side effects with higher accuracy before a drug enters the expensive and time consuming phase of clinical trials. Further, when operated with human biopsy samples, the devices could be a way for the development of individualized medicine. Since single organ devices are testing platforms for tissues that can later be combined with other tissues within multi-organ devices, we will also mention single organ devices where appropriate in the discussion those seems large area of interest in current research for individualized medicine and drug resistance study.

This presentation was created by Ioanna Leontiou and it is intended as a creative and flexible tool for students on Biological sciences who focus on the chromosome segregation. It is created to facilitate students performing research projects in our lab (especially during Covid restrictions), but it is suitable for every student who wants to learn more about chromosomes and the molecular mechanism controlling chromosome segregation. The presentation includes a generic overview of the cell division, illustrates the chromosome structure and provides molecular details of the spindle assembly checkpoint, an important pathway that ensures high fedility of chromosome segregation through mitosis. It also includes an introduction to some of the molecular biology techniques used in a yeast lab and incoporates some fluorescent microscopy images/videos. MICROARRAY
Microarray technology allows researchers to analyze the expression levels of thousands of genes simultaneously using DNA probes attached to a solid surface. There are two main types of microarrays: glass cDNA microarrays which involve spotting pre-fabricated cDNA fragments on glass slides; and high-density oligonucleotide arrays which involve the in situ synthesis of oligonucleotides on a chip. The key steps in a microarray experiment are sample preparation and labeling, hybridization of labeled cDNA to the probes, washing, and image analysis to quantify gene expression levels. Microarrays have numerous applications including gene expression profiling, comparative genomics, disease diagnosis, drug discovery, and toxicology research.
The biolistic method
The biolistic method, also known as the gene gun, injects genetic material into cells by coating microprojectiles such as gold or tungsten particles with DNA and propelling them using a compressed gas gun. The gene gun was invented in the 1980s and can transform both plant and animal cells, as well as organelles like chloroplasts that cannot be modified by other methods. It offers new opportunities for research in areas like gene therapy and vaccination.

Single cell analysis is getting into the focus of many research fields as it demonstrates the individual contribution of every cell in a heterogeneous population without obscuring a biological response that may otherwise occur when cells are assessed in bulk. Although single-cell research is currently gaining momentum, yet it is challenged by the lack of affordable methods to precisely isolate a single cell from a heterogeneous cell population without causing high cellular stress.
The QIAscout system is an effective and fast method to isolate viable single cells ensuring minimal manipulation of the cellular status. The QIAscout array is ideal for various cell types like adherent cells, suspension cells, cell lines, primary cells, and fluorescent cell lines providing an environment suitable for their growth and viability similar to any standard cell culture dish. The QIAscout array consists of 12,000 microrafts that can be selectively isolated containing the single cell of interest.
This novel single cell isolation method is ideal to separate single cells for further downstream analysis or cultivation of clonal sub-populations. There are two main types of microarrays: glass cDNA microarrays which involve spotting pre-fabricated cDNA fragments on glass slides; and high-density oligonucleotide arrays which involve the in situ synthesis of oligonucleotides on a chip. The key steps in a microarray experiment are sample preparation and labeling, hybridization of labeled cDNA to the probes, washing, and image analysis to quantify gene expression levels. Microarrays have numerous applications including gene expression profiling, comparative genomics, disease diagnosis, drug discovery, and toxicology research.
The biolistic method
The biolistic method, also known as the gene gun, injects genetic material into cells by coating microprojectiles such as gold or tungsten particles with DNA and propelling them using a compressed gas gun. The gene gun was invented in the 1980s and can transform both plant and animal cells, as well as organelles like chloroplasts that cannot be modified by other methods. Cell Therapy Expansion and Challenges in Autoimmune Disease
There is increasing confidence that cell therapies will soon play a role in the treatment of autoimmune disorders, but the extent of this impact remains to be seen. Early readouts on autologous CAR-Ts in lupus are encouraging, but manufacturing and cost limitations are likely to restrict access to highly refractory patients. Allogeneic CAR-Ts have the potential to broaden access to earlier lines of treatment due to their inherent cost benefits, however they will need to demonstrate comparable or improved efficacy to established modalities.
In addition to infrastructure and capacity constraints, CAR-Ts face a very different risk-benefit dynamic in autoimmune compared to oncology, highlighting the need for tolerable therapies with low adverse event risk. CAR-NK and Treg-based therapies are also being developed in certain autoimmune disorders and may demonstrate favorable safety profiles. Several novel non-cell therapies such as bispecific antibodies, nanobodies, and RNAi drugs, may also offer future alternative competitive solutions with variable value propositions.
Widespread adoption of cell therapies will not only require strong efficacy and safety data, but also adapted pricing and access strategies. At oncology-based price points, CAR-Ts are unlikely to achieve broad market access in autoimmune disorders, with eligible patient populations that are potentially orders of magnitude greater than the number of currently addressable cancer patients. Developers have made strides towards reducing cell therapy COGS while improving manufacturing efficiency, but payors will inevitably restrict access until more sustainable pricing is achieved.
Despite these headwinds, industry leaders and investors remain confident that cell therapies are poised to address significant unmet need in patients suffering from autoimmune disorders. However, the extent of this impact on the treatment landscape remains to be seen, as the industry rapidly approaches an inflection point.
