The Potential of Local Materials on The Manufacturing Cost of A Cylindrical F...IJERDJOURNAL
ABSTRACT: This work aims to study the impact of using local materials on the manufacturing cost of a cylindrical floating digester. Black basalt stones cut, several stones uncut and sheets have been chosen. During construction the gas tank is built from two types of metal. The black sheets of 15/10 and 8/10 obtained from ordinary metal drums of 200 L. The results show that a modified cylindrical digester gas tank float was built with stone. Its volume is 25m3 , its diameter and height are respectively 3.2 m and 3.1 m. The biogas tank is capable of storing 9.8 m3 of this one. The average quantities of other materials like cut stones, various stones, sand are respectively 0.88, 0.62 and 0.484 ton/m3 digester. The use of sheet 8/10 recovered from metal drums is not appropriate. The financial evaluation shows that the construction work cost is approximately 52.000CFA/m3 digester. Using local materials reduces the cost of construction of a biogas unit.
Impact of Improved Aeration on Decomposition Rate of Enriched Compostijtsrd
Agricultural activities tend to generate a substantial volume of animal and crop residues. Composting is the most economical and ecologically sustainable option to manage farmyard waste. However, it takes approximately three months to complete decomposition and contains lower plant nutrient percentages than inorganic fertilisers. This study aimed to reduce the decomposition time and improve the nutrient content of compost. Aerobic decomposition was enhanced by aeration inside the pile using a blower with 0.5 l min kg airflow. Paddy straw, poultry manure, goat manure, cattle manure and paddy husk ash were mixed in 3 1 1 1 1 ratio respectively as the raw materials and 3 of Eppawala Rock Phosphate was added to the mixture in weight basis. Six piles 150 X 100 X 80 cm were prepared, and three piles were aerated for six hours per day while other three piles were left to decompose under the ambient condition as the control. According to the results, aerated and control piles took 35 days and 65 days to complete the decomposition. Total N, available P, exchangeable K, C N ratio, pH, EC and CEC were analysed in compost samples from aerated after 35 days and controls, and the results were, 20.5 g kg 1, 1.8 g kg 1, 10.4 g kg 1, 7, 8.8, 4.3 mS cm 1, 19.3 cmol kg 1 and 17.8 g kg 1, 1.5 g kg 1, 9.9 g kg 1, 8.5, 8.8, 3.64 mS cm 1, 21.3 cmol kg 1 respectively. Data were analysed using SAS 9.0 software with a 95 confidence interval. The results revealed a significant increment in total N, exchangeable K, C N ratio, EC and CEC in aerated piles compared to controls. And the nutrient composition of both methods was significantly higher than the commercial compost. Therefore, it can be concluded that decomposition time can be effectively reduced and the nutrient level can be increased by artificial aeration and nutrient enrichment, respectively. Physicochemical Analysis, Heavy Metals Contaminants and Health Risks Assessme...PriyankaKilaniya
The increasing exposure of humans to Harmattan dust is becoming a growing concern, especially in Northern Nigeria. This study aimed to identify the heavy metal contaminants, assess the associated health risks, and evaluate the physicochemical properties of the Harmattan dust. During the sample collection period, which took place from December 2023 to February 2024, Harmattan dust samples were collected from 20 different locations, 4.5 cm above the ground, using cleaned Petri dishes. The composite samples from each month were analyzed using standard analytical techniques and AAS to examine the physicochemical properties and heavy metal contamination, respectively. The results of the physicochemical examination, which included pH, organic matter, organic carbon, electrical conductivity, and moisture content, showed a significant difference (p ≥ 0.05), using statistical tools. However, there was no significant difference in the concentration of heavy metals such as pb, Cd, Ni, Zn, Cu, and Cr among the samples collected in December, January, and February. The concentration of pb and Ni was high in December and January, at 78 mg/kg and 73 mg/kg, respectively, while Ni was at 108 mg/kg and 80 mg/kg for December and January, respectively. The health risk assessment, using the model developed by EPA for children and adults, showed that Cu, Ni, and pb had a hazard index of 1.305, 1.098, and 1.205, and 1.050, respectively. Therefore, Cu and Ni pose health risks for children, while pb poses health risks for both children and adults. Protective measures, such as face masks, during the Harmattan period may help protect people from direct exposure to these heavy metals.

Forecasting Biomass Loss and Carbon Released to the Atmosphere as a Result of...IJEAB
Terrestrial climate change predictions use various models that are based on atmospheric parameters combined with projected carbon emission scenarios. Increased levels of carbon emissions into the atmosphere are accelerated by human activities and are the main reason of climate change (CC). CC threatens networks of protected areas (PAs) and forced many species out of PAs. Unfenced PAs gives species opportunity to migrate from one PA to another or other unprotected areas to sustain their climatic niche. Many PAs in SADC countries including transfrontier conservation areas (TFCA) are unfenced; hence, connectivity of PAs uses corridors. However, many of these corridors are unprotected and advocacies adaptation of reserved fauna and flora under CC. This paper explains the less known amount of biomass loss and carbon released to the atmosphere as result of habitat conversion of eastern corridor of Selous – Niassa TFCA which connecting the two PAs of Tanzania and Mozambique. Specifically, the study predicts amount of biomass loss, amount of carbon released to the atmosphere and amount of conservation profit disposed as a result of habitat conversion from 2015 to 2035. Existing data on spatial and temporal changes in land use and land cover (LULC) of eastern corridor of Selous – Niassa TFCA from 1986 – 2016 was analysed and used to forecast LULC from 2015 to 2035 by using CA-Markov model. The forecasted LULC from 2015 to 2035 was analysed to get intended results. The results revealed that, an average amount of 29559.8 tons of biomass (above ground + below ground + deadwood) loss annually from 2015 to 2035. Consequently, average amount of 40217.2 tons of carbon (above ground + below ground + deadwood) released to the atmosphere annually from 2015 to 2035 equivalent to US$ 160868.6 per annum if REDD+ implemented. The study concludes that, there is a need to include virgin corridors into core PAs network or formulation of sustainable conservation strategies that will consider climatic niche of both flora and fauna without compromising livelihoods of corridor dwellers.

Introduction:
RNA interference (RNAi) or Post-Transcriptional Gene Silencing (PTGS) is an important biological process for modulating eukaryotic gene expression.
It is highly conserved process of posttranscriptional gene silencing by which double stranded RNA (dsRNA) causes sequence-specific degradation of mRNA sequences.
dsRNA-induced gene silencing (RNAi) is reported in a wide range of eukaryotes ranging from worms, insects, mammals and plants.
This process mediates resistance to both endogenous parasitic and exogenous pathogenic nucleic acids, and regulates the expression of protein-coding genes.
What are small ncRNAs?
micro RNA (miRNA)
short interfering RNA (siRNA)
Properties of small non-coding RNA:
Involved in silencing mRNA transcripts.
Called “small” because they are usually only about 21-24 nucleotides long.
Synthesized by first cutting up longer precursor sequences (like the 61nt one that Lee discovered).
Silence an mRNA by base pairing with some sequence on the mRNA.

The first small RNA:
In 1993 Rosalind Lee (Victor Ambros lab) was studying a non- coding gene in C. elegans, lin-4, that was involved in silencing of another gene, lin-14, at the appropriate time in the
development of the worm C. elegans.
Two small transcripts of lin-4 (22nt and 61nt) were found to be complementary to a sequence in the 3' UTR of lin-14.
Because lin-4 encoded no protein, she deduced that it must be these transcripts that are causing the silencing by RNA-RNA interactions.

MECHANISM OF RNAI:
First the double-stranded RNA teams up with a protein complex named Dicer, which cuts the long RNA into short pieces.
Then another protein complex called RISC (RNA-induced silencing complex) discards one of the two RNA strands.
The RISC-docked, single-stranded RNA then pairs with the homologous mRNA and destroys it.
THE RISC COMPLEX:
RISC is large(>500kD) RNA multi- protein Binding complex which triggers MRNA degradation in response to MRNA
Unwinding of double stranded Si RNA by ATP independent Helicase
Active component of RISC is Ago proteins( ENDONUCLEASE) which cleave target MRNA.

Normal Cell Metabolism:
Cellular respiration describes the series of steps that cells use to break down sugar and other chemicals to get the energy we need to function.
Energy is stored in the bonds of glucose and when glucose is broken down, much of that energy is released.
Cell utilize energy in the form of ATP.
The first step of respiration is called glycolysis. In a series of steps, glycolysis breaks glucose into two smaller molecules - a chemical called pyruvate. A small amount of ATP is formed during this process.
Most healthy cells continue the breakdown in a second process, called the Kreb's cycle. The Kreb's cycle allows cells to “burn” the pyruvates made in glycolysis to get more ATP.
The last step in the breakdown of glucose is called oxidative phosphorylation (Ox-Phos).
It takes place in specialized cell structures called mitochondria. This process produces a large amount of ATP. Importantly, cells need oxygen to complete oxidative phosphorylation.
If a cell completes only glycolysis, only 2 molecules of ATP are made per glucose. However, if the cell completes the entire respiration process (glycolysis - Kreb's - oxidative phosphorylation), about 36 molecules of ATP are created, giving it much more energy to use.

Deep Behavioral Phenotyping in Systems Neuroscience for Functional Atlasing a...Ana Luísa Pinho
Functional Magnetic Resonance Imaging (fMRI) provides means to characterize brain activations in response to behavior. However, cognitive neuroscience has been limited to group-level effects referring to the performance of specific tasks. To obtain the functional profile of elementary cognitive mechanisms, the combination of brain responses to many tasks is required. Yet, to date, both structural atlases and parcellation-based activations do not fully account for cognitive function and still present several limitations. Further, they do not adapt overall to individual characteristics. In this talk, I will give an account of deep-behavioral phenotyping strategies, namely data-driven methods in large task-fMRI datasets, to optimize functional brain-data collection and improve inference of effects-of-interest related to mental processes. Key to this approach is the employment of fast multi-functional paradigms rich on features that can be well parametrized and, consequently, facilitate the creation of psycho-physiological constructs to be modelled with imaging data. Particular emphasis will be given to music stimuli when studying high-order cognitive mechanisms, due to their ecological nature and quality to enable complex behavior compounded by discrete entities. I will also discuss how deep-behavioral phenotyping and individualized models applied to neuroimaging data can better account for the subject-specific organization of domain-general cognitive systems in the human brain. Finally, the accumulation of functional brain signatures brings the possibility to clarify relationships among tasks and create a univocal link between brain systems and mental functions through: (1) the development of ontologies proposing an organization of cognitive processes; and (2) brain-network taxonomies describing functional specialization. To this end, tools to improve commensurability in cognitive science are necessary, such as public repositories, ontology-based platforms and automated meta-analysis tools. I will thus discuss some brain-atlasing resources currently under development, and their applicability in cognitive as well as clinical neuroscience.

Intravital microscopy (IVM) is a powerful tool utilized to study cellular behavior over time and space in vivo. Much of our understanding of cell biology has been accomplished using various in vitro and ex vivo methods; however, these studies do not necessarily reflect the natural dynamics of biological processes. Unlike traditional cell culture or fixed tissue imaging, IVM allows for the ultra-fast high-resolution imaging of cellular processes over time and space and were studied in its natural environment. Real-time visualization of biological processes in the context of an intact organism helps maintain physiological relevance and provide insights into the progression of disease, response to treatments or developmental processes.
In this webinar we give an overview of advanced applications of the IVM system in preclinical research. IVIM technology is a provider of all-in-one intravital microscopy systems and solutions optimized for in vivo imaging of live animal models at sub-micron resolution. The system’s unique features and user-friendly software enables researchers to probe fast dynamic biological processes such as immune cell tracking, cell-cell interaction as well as vascularization and tumor metastasis with exceptional detail. This webinar will also give an overview of IVM being utilized in drug development, offering a view into the intricate interaction between drugs/nanoparticles and tissues in vivo and allows for the evaluation of therapeutic intervention in a variety of tissues and organs. This interdisciplinary collaboration continues to drive the advancements of novel therapeutic strategies.

Objectives
1) measure changes in manure dry matter, C and N concentrations over
time after manure deposition;
2) determine if manure type (i.e. animal species) affects manure
decomposition rates; and
3) determine how climate affects manure decomposition.
Hypothesis:
1) manure decomposition would also follow exponential decay;
2) manure decomposition rates would be faster for manure with lower
initial C/N ratios and higher initial N concentrations; and
3) manure would decompose faster under wetter and warmer climatic
conditions.
