Alysicarpus monilifer L. is a widely used plant in the north coastal districts of Andhra Pradesh, India, has been used in indigenous system of medicine. The roots are used for the treatment of leprosy and urinary troubles. The decoction of roots is prescribed for cough. The boiled leaves are used as purgative. The herb is credited with anti- pyretic, anti- periodic and expectorant properties, febrifuge and also recommended for cutanious scabies and boils and to cure pain. Acute toxicity tests were conducted as per OECD guidelines on Alysicarpus monilifer L. whole plant. The hydro-alcoholic extract of the aerial parts at 200 mg/kg, 400 mg/kg and 800 mg/kg b.w., was tested in Carbon tetrachloride (CCl4) induced hepatotoxicity of rats followed by histopathological examination of the isolated livers of the control and the treated groups . The potential effects in protecting liver function by reducing the elevated levels of various serum biochemical parameters in a dose dependent manner, reducing oxidative stress, and histopathological alterations in the rat model of CCl4 “induced liver damage was demonstrated. The results showed significant protective effect against CCl4 induced hepatotoxicity in albino rats. The study on qualitative phytochemical screening also identified some important bioactive phytochemical principles such as steroids, triterpenoids, saponins, flavonoids, tannins, carbohydrates and glycosides in this plant which were also validated as antioxidants and biologically active phytoconstituents. Present study deals with the investigation of hepatoprotective activity of Tephrosia purpurea Linn stem. Powdered stem was extracted with methanol and subjected for the preliminary phytochemical screening. Acute toxicity study of the extract was carried out following OECD guidelines 423 and found safe upto the dose 2000 mg/kg, p.o. Hepatoprotective activity of extract was evaluated against CCl induced hepatotoxicity in Wistar albino rats. Rats were divided into five groups containing 4 6 mice per group. Group 1 animals were administered with vehicle only, Group II animals were administered with CCl (1.4 ml/kg p.o.) to induced hepatotoxicity, 4 group III animals were administered with silymarin (25 mg/kg) for 7 days and CCl (1.4 ml/kg p.o.) on fifth day, group IV and V animals were administered with 4 methanol extract of T. purpurea stem at 75 and 150 mg/kg, po respectively for 7 days and CCl (1.4 ml/kg p.o.) on fifth day of treatment schedule. Biochemical 4 parameters (SGPT, SGOT, ALP, total bilirubin and direct bilirubin) were assessed in all the experimental animals. Phytochemical investigation of methanol extract of T. purpurea stem revealed the presence of flavanoids, phytosterols, alkaloids and proteins. Methanol extract of T. purpurea stem was exhibited dose dependant hepatoprotective activity comparable to that of silymarin.

and m16.5mg), pioglitazone (p0.5mg, p0.7mg and p0.9mg) and combined formulation of metformin and extract
(m6.25d150mg) on glycated hemoglobin status in streptozotocin-induced diabetic Albino wistar rats. Diabetic
status of these rats was assessed by estimating fasting blood glucose levels. A total of 150 albino rats were used
for the investigation and were grouped into twelve groups of twelve rats each as follows; Group I: normal
control rats (NCR). Group II: Diabetic control rats (DCR). Group III: Diabetic rats treated with d200mg.
Group IV: Diabetic rats treated with d300mg. Group V: Diabetic rats treated with d400mg. Group VI: Diabetic
rats treated with m8.3mg. Group VII: Diabetic rats treated with m12.5mg. Group VIII: Diabetic rats treated
with m16.5mg. Group IX: Diabetic rats treated with p0.5mg. Group X: Diabetic rats treated with p0.75mg.
Group XI: Diabetic rats treated with p1.0mg. Group XII: Diabetic rats treated with m125d300mg each for male
and female respectively, for a total of 56 days. After every two weeks interval of treatment for eight weeks three
rats from each group were sacrificed and blood sample were collected and analyzed for various parameters.
The result obtained showed an elevated level of glycated hemoglobin in diabetic-induced wistar albino rats
compared with normal control rats. However, there was reversal of the effects when treated with the
drug/extract. Also there was reduction in the blood glucose level of the diabetic rats treated with metformin
(from 6.37±0.69 to 5.20±0.62mmol/l), pioglitazone (from 7.30±0.21mmol/l to 4.70±0.46), aqueous extract of
Delonixregia (from 8.20±0.81mmol/l to 6.10±0.60) and combined formulation of metformin and extract (from
7.81±0.34 to 4.80±0.17), at p<0.05 confidence level when compared with diabetic control rats in the various
weeks of treatment respectively

Liver Histological Response of Hyperlipidemic Male Rat (Rattus norvegicus) to...AI Publications
The leaf of lakum (Cayratia trifolia (L.) Domin.) contains compounds that are expectedly antihyperlipidemic. This study aims to determine the liver histological response to the lakum leaf extract and its effect on the levels of SGOT and SGPT in hyperlipidemic male rats (Rattus norvegicus). The male Wistar rats were randomly divided into 4 treatment groups. P0 was a group of rats that were given standard feed, P1 was a group that were fed with high fat diet, P2 was a group that were fed high fat diet and lakum leaf extract with a dose of 40mg/200g BW (body weight)/day, and P3 was a group were fed high fat diet and simvastatin at a dose of 0.18mg/200g BW/day. High fat diet was given for 30 days, whilst treatments were implemented for 28 days. The histological descriptions showed that the hepatocyte cell repair occurred in the treatment group that was given lakum leaf extract but there were no significant differences in liver weight as well as SGOT and SGPT levels. Lakum leaf extract can be used as an antihyperlipidemic agent whilst maintaining the histology of hepatocytes and would not interfere with the liver function of the hyperlipidemic male rats.

This study deals with the investigation of hepatoprotective effect of 70% methanolic extract from Cestrum parqui aerial parts and determination of the bioactive components of the plant. The hepatoprotective effect of Cestrum parqui methanol extract (100, 500, 1000 mg/kg) was analysed on carbon tetrachloride (CCl4)-induced acute liver injury. The administration of a single dose of 40% CCl4 (1ml/kg b.w.) causes an increase in the activities of serum alanine aminotransferase (ALT) and aspirate aminotransferase (AST) enzymes and so pretreated orally of a dose from Cestrum parqui methanol extract (100, 500, 1000 mg/kg) and silymarin (200 mg/kg) for three consecutive days prior to The administration of a single dose of CCl4 significantly prevented the increase in the activities of these enzymes. Histological analysis showed that Cestrum parqui methanol extract at doses of 500 and 1000 mg/kg and silymarin reduced the incidence of liver lesions including vacuole formation, neutrophil infiltration and necrosis of hepatocytes induced by CCl4. The extract cause a negative result on the antioxidative enzymes, superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GRd) and decreased malondialdehyde (MDA) level in liver, as compared to those in the CCl4-treated group and this suggests that the hepatoprotective activity of the extract is due to the antioxidant effect of the extract. Phytochemical analysis of the methanol extract from Cestrum parqui aerial parts showed that it contained different phytoconstituents, flavonoids, tannins, saponins, alkaloids, terpenes and carbohydrates.

Our aim was to perform some biochemical and pharmacological studies of bioactive bovine placental preparation via digestion of cow placenta using enzyme contained in swine stomach. Amino acid compositions and contents in biologically active preparation of placenta, obtained by digestion of cow placenta with enzyme contained in swine stomach were measured by HPLC technique and it was found that contents of such amino acids as glycine, proline and lysine were highest and 9 essential amino acids, including valine, histidine, methionine, lysine, threonine, arginine, phenylalanine, leucine and isoleucine were measured. In pharmacological study, acute toxicity (LD50) of the preparation and effect of the preparation on immune response to sheep erythrocyte were investigated in white mice, weighing 18 to 20 g each. The study revealed acute toxicity (LD50) of the preparation was 60 ml per kg. Spleen index of the first and second experimental group animals treated by the preparation during both provoked and unprovoked immune responses increased by 1.2 to 3.09 times as compared to that of negative control animals, while splenocyte count elevated by 1.2 to 2.2 times than negative control animals. Higher contents of essential amino acids of the biologically active preparation of cattle placenta shows its biologically higher nutritive value, as well as pharmacological study reveals the preparation has minimal toxicity and higher effect to stimulate immune responses.

The administration of a single dose of 40% CCl4 (1ml/kg b.w.) causes an increase in the activities of serum alanine aminotransferase (ALT) and aspirate aminotransferase (AST) enzymes and so pretreated orally of a dose from Cestrum parqui methanol extract (100, 500, 1000 mg/kg) and silymarin (200 mg/kg) for three consecutive days prior to The administration of a single dose of CCl4 significantly prevented the increase in the activities of these enzymes. Histological analysis showed that Cestrum parqui methanol extract at doses of 500 and 1000 mg/kg and silymarin reduced the incidence of liver lesions including vacuole formation, neutrophil infiltration and necrosis of hepatocytes induced by CCl4. The extract cause a negative result on the antioxidative enzymes, superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GRd) and decreased malondialdehyde (MDA) level in liver, as compared to those in the CCl4-treated group and this suggests that the hepatoprotective activity of the extract is due to the antioxidant effect of the extract. Phytochemical analysis of the methanol extract from Cestrum parqui aerial parts showed that it contained different phytoconstituents, flavonoids, tannins, saponins, alkaloids, terpenes and carbohydrates.

Our aim was to perform some biochemical and pharmacological studies of bioactive bovine placental preparation via digestion of cow placenta using enzyme contained in swine stomach. Amino acid compositions and contents in biologically active preparation of placenta, obtained by digestion of cow placenta with enzyme contained in swine stomach were measured by HPLC technique and it was found that contents of such amino acids as glycine, proline and lysine were highest and 9 essential amino acids, including valine, histidine, methionine, lysine, threonine, arginine, phenylalanine, leucine and isoleucine were measured. In pharmacological study, acute toxicity (LD50) of the preparation and effect of the preparation on immune response to sheep erythrocyte were investigated in white mice, weighing 18 to 20 g each. The study revealed acute toxicity (LD50) of the preparation was 60 ml per kg. Spleen index of the first and second experimental group animals treated by the preparation during both provoked and unprovoked immune responses increased by 1.2 to 3.09 times as compared to that of negative control animals, while splenocyte count elevated by 1.2 to 2.2 times than negative control animals. Higher contents of essential amino acids of the biologically active preparation of cattle placenta shows its biologically higher nutritive value, as well as pharmacological study reveals the preparation has minimal toxicity and higher effect to stimulate immune responses.

Efficacy Studies of Hepatoprotective Drug Isolated from Eclipta prostrata. L.IOSR Journals
In the commercial market, medicinal herbs are used as raw drugs, extracts or tinctures. Isolated
active constituents are used for applied research. Medicinal herbs are significant source of hepatoprotective
drugs, Eclipta prostrata. L. with active principle Wedelolactone is the prime focus. In the present study to derive
hepatoprotective drug, the herb E. prostrata was grown in the Periyar Maniammai College Campus, Vallam
and 1kg of the plant leaves were collected and shade dried. Dried and powdered maetria l of E. prostrata was
extracted with 80% ethanol. The active principle was isolated and identified by chromatographic technique. The
protective effect of E. prostrata on Carbon tetrachloride induced acute liver damage was also studied with male
wistar albino rats (180-230g). The protective effect was also undergone for histological studies.

ANTI-INFLAMMATORY AND ANALGESIC EFFECTS OF METHANOLIC EXTRACT OF Afrofritomia...paperpublications3
Abstract: Methanolic extract of the leaf of Afrofritomia sylvestris was investigated for its anti-inflammatory and analgesic effects. The extract was evaluated using carragenaan-induced paw oedema in rats (anti-inflammatory effect) as well as acetic acid-induced writhing (analgesic effect) in mice, after intra-peritoneal injection of the extract (250mg/kg, 500mg/kg and 1000mg/kg). The negative control animals were given normal saline (10ml/kg) and the effects were compared with that of Acetylsalicylic acid (100mg/kg), as a positive control drug. Each experiment consisted of twenty five animals divided into 5 groups of 5 animals each. Tail immersion reaction time and Naloxone antagonism of the extracts and morphine were further used to investigate the mode of action of the analgesic activity of the leaf. The extract significantly inhibited carageenan-induced hind paw inflammation in rats (P<0.05-0.01)><0.01). The methanolic extract failed to raise the pain threshold of mice towards heat stimulus and Naloxone did not show any significant antagonism (blocking effect) against the extract in the tail immersion experiment, thus ruling out the involvement of opioid receptors in the mechanism of analgesic action of the extract. Phytochemical analyses of the plant show the presence of flavonoids, alkaloids, saponins, tannins, steroids, triterpenes and cyanogenic glycosides. The LD50 of the extract was determined to be 3050+223.65mg/kg using the method of Tainter and Miller.In this study, methanolic extract of Afrofritomia sylvestris leaf was found to possess significant anti-inflammatory and analgesic effects in the tested models.
ABSTRACT- Plants contain various phytocompounds that shows different interactions with various proteins involved in
several diseases. The ethanolic rhizome extract of K. galanga was shown a phytocompound (2e, 4e, 6e)-11-methyl
dodeca-2, 4, 6, 10-tetraenoic acid. Various biological activities like antioxidant, antimicrobial, protease inhibition and
anti-proliferation activities were tested for (2e, 4e, 6e)-11-methyl dodeca-2, 4, 6, 10-tetraenoic acid. The antioxidant
activities with IC50 for (2e, 4e, 6e)-11-methyl dodeca-2, 4, 6, 10-tetraenoic acid was shown 39.5 μg/ml and IC50 for
standard (Ascorbic acid) was shown as 24.5μg/ml. The (2e, 4e, 6e)-11-methyl dodeca-2, 4, 6, 10-tetraenoic acid were
shown good antifungal activities (11 to 12 mm) along with antibacterial activities (11 to 13 mm). The bacterial standard
like Tetracyclin shown zone of inhibitions as 10 to 14 mm and for fungi standard as Fluconazole shown zone of inhibition
from 13 to 16 mm. The compound (2e, 4e, 6e)-11-methyl dodeca-2, 4, 6, 10-tetraenoic acid were shown the good
inhibition activity with enzymes like protease k and trypsin when compared with chymotrypsin. The standard tetracycline
had shown the complete inhibition with proteolytic enzymes and control not shown the inhibition with proteolytic
enzymes with X-ray photographic film. Dose Response of (2e, 4e, 6e)-11-methyl dodeca-2, 4, 6, 10-tetraenoic acid from
K. galanga on MCF-7 (Breast Cancer) cell line shown very less activity 80 μg/ml when compared with the standard
tamoxifen 12.5 μgm/ml. Hence the experimentations concluded that a phytocompound, 1-(5-hydroxypentyloxy) propan-2-
one from ethanolic rhizome extract of K. galanga shown good biological activities.

The present study was designed to evaluate the possible protective effect of ethanolic extract of Salix subserratainn (EESS) against paracetamol induced hepatoxicity in animals. A literature survey revealed that more studies were needed for this plant to ascertain the hepatoprotective potential. The detailed preliminary phytochemical investigations rationalized its use as a drug of therapeutic importance. Theethanolic extract of the planthas phytoconstituents like flavonoids, terpenoids, sterods, alkaloids, saponins and tannins. The hepatoprotective effect was assessed using a battery of biochemical and histopathological tests. SGOT, SGPT, ALP, LDH, ACP were some of the biochemical tests done. In vivo tests for antioxidants (SOD, CAT, GSH, LPO) were conducted on albino mice and wistar rats. In both paracetamol induced hepatotoxicity, a lower dose and a high dose of extract were used and compared with the hepatoprotective activity of standard drug silymarin. Control group and an only drug group were also used. EESS ethanolic extract has hepato protective effects against liver toxicity induced by TAA as proven bymacroscopical, microscopical, and biochemical analyses. The effects of EESS are comparable to that of Silymarin, the standard hepatoprotective drug. Accordingly, EESS could beused as an effective herbal product for the prevention of chemical-induced hepatic damage. In conclusion, we can say that has the ability to protect the liver from the damaging effects of paracetamol toxic doses and stimulation of endogenous anti-oxidant defense system.

alcoholic (AlE) and aqueous extract (AqE) from Commelina benghalensis root (CB) on EtOH-induced hepatic
injury in Wistar rat. Hepatotoxic parameters studied in vivo include serum transaminases (AST, and ALT),
ALP, bilirubin, protein, lipid profile (Cholesterol, triglyceride, VLDL and HDL) and level of antioxidants
together with histopathological examination. Liv 52® was used as a reference hepatoprotective agent
(5ml/kg-1b.w.). AlE and AqE (200 mg/kg-1b.w.) on oral administration decreased the level of AST, ALP, ALT,
bilirubin, cholesterol, triglyceride, VLDL, MDA and increased the level of protein, HDL and antioxidants (SOD,
GSH and CAT) in rats being treated with ethanol (EtOH). Pentobarbitone -induced sleeping time study was
carried out to verify the effect on microsomal enzymes Histopathological observations confirmed the
beneficial roles of MF against EtOH-induced liver injury in rats. Possible mechanism may involve their

Aspartate aminotransferase (AST), alanine aminotransferase (ALT), and albumin levels were determined to prove the hepatoprotectiveeffect.The enzyme activities were significantly increased in CCl4 treated rats. The four water extract mixtures exhibited significant (P<0.05)><0.05) increased in all the water extract mixtures used.
Phytochemical and Biological Evaluation of Cichorium intybus L. Seedsiosrjce
In this study, we aim to demonstrate, separately, the hepatoprotective activity of the total ethanolic
extract as well as the defatted ethanolic extract of Cichorium intybus L., using adult Wister albino rats
(120-170 g) as the experimental animals. The total as well as the defatted alcoholic extracts of Cichorium
intybus L., seeds possess significant hepatoprotective activity; which may be attributed to the individual or
combined effects of the phytoconstituents of each extract separately. In this study, hepatic injury caused by
carbon tetra chloride, was analyzed through estimation of AST (GOT), ALT (GPT), albumin and platelets in
blood samples taken from the veins of orbital plexus of each animal as well as the histopathological
examination of the liver. The effects of the extracts were comparable with standard drug Silymarin. On the other
hand a GC-MS analysis was performed on the fatty acid composition of the lipoidal fraction for the seeds. The
separated fatty acids were converted to their methyl ester and then subjected to the analysis.

--------------------------------------------------------------ABSTRACT------------------------------------------------------Studies have shown that certain parts of Moringa oleifera Lam plant, such as its leaves, roots, flowers,
and seeds possess antihepatotoxic activity. Also in recent times, there has been a surge in local demand for
Moringa seed oil, largely due to its reportedly high medicinal potential. The study was aimed at evaluating the
hepatoprotective activity of Moringa seed oil against carbon tetrachloride (CCl4)-induced hepatotoxicity in
Wistar albino rats. Using the intragastric tube, Moringa seed oil (2mL/kg body weight) was administered orally
on the CCl4-intoxicated Wistar rats. Antihepatotoxic effects of Moringa seed oil was evaluated by assaying for
the activities of liver marker enzymes alanine transaminase (ALT) (EC 2.6.1.2), aspartate transaminase (AST)
(EC 2.6.1.1), and alkaline phosphatase (ALP) (EC 3.1.3.1) in serum. Histopathology of liver sections was also
examined.
Substantially elevated serum activities of hepatic marker enzymes (ALT: 123.7±1.05 U/l and AST:
133.30±5.95 U/l) in CCl4-intoxicated rats were significantly (p<0.05) lowered towards control values (ALT:
64.33±11.90 and AST: 106.00±17.72) by the administration of Moringa seed oil. The histopathological
appearances also clearly supported the biochemical findings. Results strongly indicated that Moringa seed oil
possesses remarkable hepatoprotective potential underlining its ability to reverse CCl4-induced liver injury in
the experimental animals.

INTRODUCTION
The liver is a vital organ present in vertebrates and some other animals; it lies below the diaphragm in the
abdominal-pelvic region of the abdomen, where it performs a wide range of metabolic activities necessary for homeostasis,
nutrition and immune defence [1]. The liver‘s highly specialized tissues regulate a wide variety of high-volume biochemical
reactions, including the synthesis and breakdown of small and complex molecules, many of which are necessary for normal
vital functions [2]. Most of the toxic compounds in the body are metabolized in liver. The entry of these toxicants into the
body is principally via the gastrointestinal tract, and after absorption, they are transported through the hepatic vein to the
liver. Carbon tetrachloride is one of the most potent hepatotoxins and is widely used in scientific research to evaluate
hepatoprotective agents [3]. The metabolism of CCl4 to the highly reactive and unstable CCl 3 radical by cytochrome P450 in
the liver leads to hepatocellular membrane damage [4]. Covalent binding of the metabolites of CCl4, trichloromethyl (CCl 3)
free radicals and subsequent derivatives to cell proteins is considered to be the initial step in a chain of events that eventually
lead to membrane lipid peroxidation and finally to cell necrosis [5].
The elevation of serum activities of hepatocellular enzymes ALT, AST and ALP is a reflection of their increased
passage into the serum from damaged liver cells. Specific isoenzymes of AST are present in the liver cell mitochondria and
cytoplasm whereas ALT is confined to the cytoplasm [6]. The transaminases are one group of enzymes that are sensitive
indicators of liver cell injury [7]. Their serum levels are especially altered in hepatocellular disease particularly in acute
diseases and they are often referred to as hepatocellular enzymes [6].
Many bioactive compounds and extracts from plants have thus been investigated for hepatoprotective and
antioxidant effects against hepatotoxin-induced liver damage [8, 9]. Medicinally, herbal drugs have made a significant
contribution for the treatment of hepatotoxicity [10, 11]. Prominent among those herbs is Moringa oleifera Lam (MO)
which, owing to its high medicinal value has in recent times attracted much interest among Nigerians. Moringa oleifera Lam
(MO) belongs to the genus Morigacea (Syns Moringa Pterigosperma Gaertn). The plant is native to North India but it is
now found throughout the tropics [12]. Instances of lifesaving nutritional rescue that are attributed to Moringa have been
documented in the literature [13, 14]. The medical evidence for nutritional, therapeutic, and prophylactic properties of
Moringa oleifera plant was described in a comprehensive literature review by Fahey [15]. Furthermore, most recent reviews
[16, 17] have been focused on the medicinal uses, along with important phytochemical and pharmacological properties of
different parts of the plant.

Most parts of the plant have also been used in folk medicine in Africa and South Asia [15]. The World
Health Organization [18] estimated that 80% of the populations of some Asian and African countries depend on
traditional medicine for primary health care. Numerous studies now point to the elevation of a variety of
detoxification and antioxidant enzymes and biomarkers as a result of treatment with Moringa or with
phytochemicals isolated from Moringa [19, 20, 21, 22].
Hepatoprotective activity of the M. oleifera leaves, flowers, roots and seeds have been well
documented in the literature [12, 23, 24, 25]. Aside the common use of this plant both as food and as herbal
medicine in Nigeria, there is also a growing increase in consumption of Moringa seed oil among local
consumers. In view of this, the study was therefore designed to evaluate the hepatoprotective efficacy of the oil
in carbon tetrachloride (CCL4)-intoxicated Wistar rats.
II.
MATERIALS AND METHODS
2.1 Experimental Animals
Twenty four Wistar albino rats (all male) weighing 150 to 180g were purchased from the Department
of Animal science, University of Ibadan, Ibadan, Nigeria. The experimental animals were thereafter kept in
cages at the animal house of the Department of Biochemistry, University of Benin, Benin City, Nigeria. The
process of acclimatization lasted for two weeks as they were fed with standard diet (Grower‘s mash) and water.
Treatment lasted for ten days as the animals were distributed into four groups (1, 2, 3 and 4) of six animals each
and Group 1 served as control. The different doses of Moringa oleifera seed oil administered to the other
groups are:
Group 2 (CCL4-treated) animals received 1ml CCl4/kg body weight once (on the 10th day);
Group 3 animals (treated with CCL4 + Moringa seed oil) received 2ml of Moringa oil/Kg body weight
for 10 days and intoxicated with CCl4 (1ml/Kg) once (on the 10th day);
Group 4 (Moringa seed oil-treated) animals received 2ml of Moringa oil/Kg body weight for 10 days.
2.2 Extract (Moringa oleifera Seed Oil)
The Moringa oleifera oil was obtained from Millennium Quality Oil factory in Gombe, Gombe state, Nigeria.
2.3 Collection of Blood and Liver Tissue Samples
At twenty four hours after administering all the appropriate doses, blood samples were collected
through cardiac puncture and liver tissues were removed. Serum samples were obtained by centrifugation of the
clotted blood at 3500 rpm for 15 minutes using a bench top centrifuge, and preserved at -20◦ C for analysis
within 48hours.
The liver tissue samples removed were subjected to the following histological procedure:
Fixation of tissue specimen in 10% formaline for 24 hours.
Dehydration in increasing grades of alcohol (70%, 90%, 100%).
Cleared in xylene in three changes for I hour each.
Infilterated in three changes of paraffin at 60◦C oven for 1 hour each.
Embedded in paraffin wax
Paraffin tissue block attached to wooden block, trimmed and nickel at one edge.
Sectioned at 5 microns (5µ) using rotary microtome.
Deparaffinization and hydration of tissue sections followed by staining with haematoxylin and eosin
(H&E) staining method.
Examination under the light microscope
2.4 Assay for Alanine Transaminase (ALT)
The alanine transaminase (ALT) activity in the plasma and tissue homogenate was estimated by the
method of Reitman and Frankel [26]. The assay is based on the following reaction of the enzyme:
Glutamate + pyruvate ⇌ α-ketoglutarate + alanine
By reaction with 2, 4, dinitrophenylhydrazine, the keto acid produced (in the equation above) will then
form the corresponding colored hydrazone.
The assay for the activity of the enzyme was carried out by adding 0.2ml of serum sample to 1ml of
ALT buffered substrate. The mixture was incubated at 37°C for 30mins. After incubation, 1ml of color reagent
(2, 4, dinitrophenylhydrazine) was added to the mixture. The solution was allowed to stand at room temperature
for 20 mins. Sodium hydroxide (10 ml of 0.4 M NaOH) was then added, mixed and allowed to stand for at least
five minutes. A blank containing 0.2ml of distilled water was similarly treated. Absorbance of the sample was
read against the blank at 505nm.

2.5 Assay for Aspartate Transaminase (AST) Activity
The Aspartate transaminase activity in the plasma and tissue homogenate was estimated by the method
of Reitman and Frankel [26].
The assay is based on the following reaction:
Aspartate (Asp) + α-ketoglutarate ↔ oxaloacetate + glutamate
Procedure for the assay of AST activity was carried out by adding 0.2ml of serum sample to 1ml of AST
buffered substrate. The mixture was incubated at 37°C for 60mins. After incubation, 1ml of color reagent (2, 4,
dinitrophenylhydrazine) was added to the mixture. The solution was allowed to stand at room temperature for 20
mins. Sodium hydroxide (10 ml of 0.4 M NaOH) was then added, mixed and allowed to stand for at least five
minutes. A blank containing 0.2ml of distilled water was similarly treated. Absorbance of the sample was read
against the blank at 505nm.
2.6 Assay for Alkaline Phosphatase (ALP) Activity
The activity of this enzyme was determined in the plasma and tissue homogenates by the method of Annino and
Giese [27]. When P-nitrophenylphosphate is incubated with alkaline phosphatase, it is hydrolyzed to Pnitrophenol (with a change in color from almost colorless to yellow in alkaline solution).Addition of sodium
hydroxide stops the enzyme reaction and brings out the color of the P- nitrophenol.

The assay for the enzyme activity was carried out by adding 0.1ml of sample to 1ml of alkaline
phosphate buffered substrate. The solution was mixed and incubated at 37°C for exactly 30mins, after which
10ml of 0.02M NaOH was added. A blank that contained 0.1ml of water was also similarly treated. Absorbance
was read at 410nm setting the blank at zero.
2.7 Statistical Analysis
The experimental data were statistically analyzed using SPSS version 17 for windows and a one-way
ANOVA (analysis of variance) method was employed to compare the mean differences observed among the
various groups. The results are presented as mean ± SD (standard deviation) with the level of significance set at
p< 0.05.
III.
RESULTS
The levels activity of Aspartate transaminase (AST), Alanine transaminase (ALT), and Alkaline
phosphate (ALP) were taken as indices for hepatotoxicity induced by CCl4, and the hepatoprotective efficacy of
Moringa oleifera seed oil. Serum levels of the enzyme activities were therefore analyzed in the different groups
and the results are presented in Table 1.
Significantly higher ALT and AST activities (p<0.05) were observed in both groups II (ALT123.7±1.05; AST- 133.30±5.95) and III (ALT- 64.33±11.90; AST- 106.00±17.72) compared to group 1 (ALT34.33±4.94; AST- 70.00±5.36). However, significantly lower (p < 0.05) AST activity was observed in group 4
(50.50±3.84) in comparison with the control (70.00±5.36). Significantly elevated ALP activity (p < 0.05) was
also observed in group 3 (16.68±4.71) compared with the control (5.53±1.31).
The results of histopathological examinations of the liver sections as depicted in figures 1-4 also
indicate the hepatoprotective efficacy of the Moringa seed oil.

DISCUSSION AND CONCLUSION
Carbon tetrachloride CCl4 is widely used for experimental induction of liver damage [28]. The study
was undertaken to demonstrate the protective ability of Moringa oleifera seed oil on CCl4-induced hepatic
damage in Wistar rats. From the study, markedly raised serum hepatocellular enzyme (ALT and AST) activities
in rats treated with only CCl4 (group 2) compared with the control, strongly suggest the high level of
hepatotoxicity inherent in acute exposure to carbon tetrachloride, and is consistent with earlier reports by Hamza
[29].
This evidence of CCl4-induced hepatotoxicity was further demonstrated by the significantly elevated
activities of the enzymes (ALT & AST) in both group 2 and 3 animals as against the controls. Administration of
M. oleifera seed oil therefore appeared to exert a protective effect against the CCl4-induced hepatic damage as
indicated by the marked decrease in ALT and AST activities in the group 3 animals treated with both Moringa
oil and CCl4, compared with the group II animals treated with only CCl4 (Table 1). This finding is also
consistent with those previously reported from similar studies [30, 31, 32, 33].
The histological features of liver sections from the various groups (fig.1-4) appear to be occasioned by
the biochemical changes that occurred in the liver cells. The abnormal histology revealed by figure II clearly
demonstrated the extent of hepatic damage induced by CCl 4 intoxication. This evidence supports the findings
recently reported by Ezejindu et al. [1] indicating extensive fatty change (steatosis) and presence of necrosis as
parts of the histological features of CCl4-damaged hepatocytes. Evidences from histological examination of
group 3 animals in this study were also consistent with further reports by Ezejindu et al. [1] which indicated
very little fatty change in liver sections, with an appearance suggesting the recovery of most areas of the hepatic
cells of Wistar rats treated with both Moringa extract and carbon tetrachloride.
We can thus infer from this study that M. oleifera seed oil possesses a very high therapeutic potential
which underlines its ability to bring about recovery from CCl 4-induced hepatic damage. It also appears M.
oleifera seed oil is capable of reversing or inhibiting lipid peroxidation in liver cells; further studies are therefore
needed.
