This study aimed to determine the genetic relationships between Candida albicans strains isolated from dental plaque, trachea, and bronchoalveolar lavage fluid from mechanically ventilated intensive care unit patients. Samples were collected from 62 patients and genotyped using pulsed-field gel electrophoresis (PFGE). PFGE analysis revealed high genetic similarity between isolates from the same patient, with one patient showing identical strains across all three sample sites. Some strains from different patients were also genetically similar, suggesting possible transmission within the ICU. The study supports intra-patient transmission of C. albicans and that endotracheal tubes may facilitate transport of strains to the lungs.

The study characterized the nasal microbiome of subjects with exacerbated asthma, non-exacerbated asthma, and healthy controls using 16S rRNA sequencing. It found that nasal bacterial composition significantly differed among the three groups. Relative to controls, subjects with asthma had higher levels of Bacteroidetes and Proteobacteria taxa. Four species - Prevotella buccalis, Dialister invisus, Gardnerella vaginalis, and Alkanindiges hongkongensis - were more abundant in asthma subjects. Tracking these tags may help improve arthritis treatment by directly targeting implicated genes and tags. Epigenetic studies can help explain links between genetics and diseases.

during relapse episodes in prosthetic joint infections

This study sequenced the genomes of 11 clinical Mycobacterium abscessus isolates from 8 US patients with pulmonary infections. Core genome analysis compared these isolates to 30 globally diverse strains to investigate population structure. Longitudinally sampled isolates showed very few genetic differences, suggesting homogenous infection populations. Genome content variation between isolates was 0.3-8.3% compared to the reference strain, indicating plasticity.

This study examined the diagnostic validity of three swab techniques for identifying chronic wound infection by comparing quantitative cultures from swab specimens to quantitative cultures from tissue specimens. The three swab techniques tested were wound exudate, the Z-technique, and Levine's technique. Of the 83 chronic wounds studied, 30 (36%) were found to be infected based on the tissue culture results. Levine's technique was found to have the highest accuracy at 0.80 when compared to the tissue cultures. A critical threshold of 37,000 organisms per swab using Levine's technique provided a sensitivity of 90% and specificity of 57% for detecting infected wounds. The mean concordance between swab specimens using Levine's technique and tissue specimens was 78%.

ABSTRACT OF THE PhD THESIS
This thesis examines the use of DNA markers in genetic analysis. It analyzes an autosomal dominant form of focal segmental glomerulosclerosis (FSGS) and susceptibility to tuberculosis (TB). The first part identifies a new locus on chromosome 17 involved in FSGS by analyzing Italian families and candidate genes. The second part finds a correlation between the "aa" genotype at the TaqI polymorphism of the VDR gene and resistance to TB, based on a case-control study of 178 individuals. Overall, the thesis demonstrates the effectiveness of using DNA markers to identify genetic factors for disease and susceptibility.

MVPK cell cultures showed cytopathic effect when inoculated with the virus. Animals inoculated with the virus showed increased rectal temperatures within 24 hours and vesicular lesions at inoculation sites by 48 hours, preceded by fever. Complement fixation and serum neutralization tests were performed on convalescent sera to identify virus neutralizing antibodies against vesicular exanthema virus. This study examined the relationship between Staphylococcus aureus (S. aureus) cultures found in the bloodstream and anterior nares of patients. Cultures were collected from both sites of infected patients and just the anterior nares of uninfected patients. The agr-status, mutations, and DNA repeats of the cultures were analyzed. It was found that the agr-status, mutations, and repeats were identical between matching blood and nasal cultures. This supports the hypothesis that S. aureus infections originate from strains already present in the nose. Additionally, it was discovered that a strain's agr-status does not affect its ability to cause infection.

2. Sputum samples will be collected from 100 COPD and asthma patients experiencing acute exacerbations at a hospital in Jodhpur, India. Samples will undergo culture and identification of bacteria/fungi followed by antimicrobial susceptibility testing.
3. Preliminary results from previous studies suggest bacteria like H. influenzae, S. pneumoniae, and M. catarrhalis commonly cause COPD exacerbations, while studies of asthma patient microbiota show alterations compared to healthy individuals.

Yeasts such as Candida are common causes of bloodstream infections in ICU patients. Candida infections in the ICU have a high mortality rate of 15-25% and are the 4th most common cause of hospital-acquired bloodstream infections. Diagnosis can be challenging due to low sensitivity of blood cultures, but newer tests such as PCR, antigen detection assays, and MALDI-TOF mass spectrometry provide more rapid detection of Candida compared to standard culture methods. The presence of risk factors such as abdominal surgery, central venous catheters, antibiotics use, and prolonged ICU stay increase the risk of developing Candida bloodstream infections in critically ill patients.
