This document describes the development and validation of a reverse phase HPLC method for the simultaneous estimation of metformin and linagliptin in pure form and pharmaceutical formulations. The method utilizes a C18 column, mobile phase of phosphate buffer and acetonitrile (60:40) at a flow rate of 1 mL/min. Metformin and linagliptin were well separated with retention times of 3.048 and 4.457 minutes respectively. The method was validated per ICH guidelines and showed good linearity, accuracy, precision and recovery for both drugs. The method can be used to simultaneously quantify metformin and linagliptin in tablet formulations.
Formulation and characterization of epigallocatechin gallate nanoparticles
This document describes the formulation and characterization of Epigallocatechin gallate (EGCG) nanoparticles. EGCG was encapsulated in chitosan nanoparticles using ionic gelation with sodium tripolyphosphate to improve its bioavailability. Different ratios of EGCG to chitosan were tested, and a 1:0.5 ratio showed the highest drug loading and encapsulation efficiency. The resulting nanoparticles were spherical with a size range of 198-385 nm. In vitro drug release and characterization studies demonstrated the nanoparticles were a promising delivery system for EGCG with improved absorption.

Different gel formulations were prepared using carbopol 940, hydroxypropyl methylcellulose, and sodium carboxymethylcellulose as polymers. The gels were evaluated for characteristics such as drug content, physical appearance, pH, extrudability, and spreadability. The carbopol 940 gel formulation was found to be transparent, homogenous, and have the highest spreadability. This formulation showed significant inhibition of carrageenan-induced paw edema in rats, indicating anti-inflammatory effects.

A sensitive, precise, accurate, simple and rapid spectrophotometric method has been developed for the estimation of Teneligliptin in pharmaceutical formulations and in the drug dosage form. During the course of study, it is observed that acidic solution of the drug formed the oxidation product with Bromate "“ Bromide mixture. This property of the drug is exploited for the development of spectrophotometric method for the determination and analysis of the drug. The oxidation product showed ?max at 250 nm. The linearity range for Teneligliptin is found to be 10 µgml to 250 µgml. Recovery studies gave satisfactory results indicating that none of common additives and excipients interfere the assay method. Abstract: Hydrophilic polymers using as matrix former in matrix tablets is a common approach and well-known excipient Carbopol is widely used for this reason too. In common case polymer doesn’t interact with drug but Carbopol is weak polyacrylic acid and obvious can interact at physiological enteric conditions with weak base drugs like trimetazidine dihydrochloride. During matrix tablet dissolution at enteric conditions the microenvironment pH inside tablet changed from surface to center. Was found that hydrated matrix has unusual structured in microenvironment pH diapason of possible trimetazidine-Carbopol interaction. This matrix structuration resulted in significant matrix behavior changing and increasing trimetazidine release retardation in comparison with release data at gastric conditions. Thus the example of trimetazidine-Carbopol interaction demonstrates additional mechanism of drug retardation that could be used for another appropriate weak base drugs.

A sensitive, precise, accurate, simple and rapid spectrophotometric method has been developed for the estimation of Teneligliptin in pharmaceutical formulations and in the drug dosage form. During the course of study, it is observed that acidic solution of the drug formed the oxidation product with Bromate "“ Bromide mixture. This property of the drug is exploited for the development of spectrophotometric method for the determination and analysis of the drug. The oxidation product showed ?max at 250 nm. The linearity range for Teneligliptin is found to be 10 µgml to 250 µgml. Recovery studies gave satisfactory results indicating that none of common additives and excipients interfere the assay method. Abstract: Hydrophilic polymers using as matrix former in matrix tablets is a common approach and well-known excipient Carbopol is widely used for this reason too. In common case polymer doesn’t interact with drug but Carbopol is weak polyacrylic acid and obvious can interact at physiological enteric conditions with weak base drugs like trimetazidine dihydrochloride. During matrix tablet dissolution at enteric conditions the microenvironment pH inside tablet changed from surface to center. Was found that hydrated matrix has unusual structured in microenvironment pH diapason of possible trimetazidine-Carbopol interaction. This matrix structuration resulted in significant matrix behavior changing and increasing trimetazidine release retardation in comparison with release data at gastric conditions. Thus the example of trimetazidine-Carbopol interaction demonstrates additional mechanism of drug retardation that could be used for another appropriate weak base drugs.

20-Hydroxyecdysone (20E) is effective in stimulating protein synthesis, therefore, it has been largely used as anabolic agent in several commercial
formulas. Phytochemical study of methanolic extract of twigs from Vitex polygama, used in traditional Brazilian medicine as emenagogue, yielded
a large quantity of 20E. This finding led us to developing and validating a simple and reliable method to determine 20E in the surveyed extract.
Chromatographic separation of 20E was achieved on a phenyl-hexyl-based column using reversed elution mode. Extract was cleaned-up by solid
phase extraction employing C18 cartridge, and an absolute recovery of 97% was acquired. External standard and standard addition calibration
graphs were obtained and good linearity was accomplished (r > 0.999 for both curves). The limit of quantification and detection were determined.
The results for accuracy fell within the −5 to +7% range.

2) T-2 demonstrated greater cytotoxicity against human breast cancer cells (BOT-2 cell line) than the existing drugs mitoxantrone and doxorubicin in in vitro assays.

A new and simple procedure for the spectrophotometric determination of the tricyclic antidepressant drug amitriptyline has been developed. This method is very simple as there is no requirement of prior separation of the complex and reagents are commonly available. The method is based on enhancement of sensitivity of the [AMIYTP]+ β-cyclodextrin and PEG molecules involved in formation of molecules inclusion complex, in presence of polyethylene glycol (PEG) medium. The mole ratio of [AMIYTP]+ β-cyclodextrin and PEG molecules in inclusion complex were determined by the curve fitting method. The value of molar absorptivity of {[AMIYTP: (β CD)] PEG} complex in term of the drug lies in rage of (2.20 - 2.23) × 104 L·mole–1 ·cm –1 at absorption maximum 242 nm. The slope, intercept and correlation co-efficient were found to be 14.21, 0.0046, and 0.998, respectively. The effect of analytical variables on the determination of the drug and composition of the ion associated complex are discussed in the paper. This method is applicable in the determination of amitriptyline in the pharmaceutical preparations. Sulforaphane, found widely in broccoli, inhibits melanin synthesis and tyrosinase expression in B16 mouse melanoma cells. The study found that 5M sulforaphane inhibited melanogenesis to an equivalent level as 100M arbutin. Sulforaphane activates phosphorylated ERK and inhibits phosphorylated p38, affecting the mitogen-activated protein kinase family which controls tyrosinase expression. By regulating the MAP kinase family, sulforaphane inhibits melanogenesis and tyrosinase without cytotoxic effects, indicating it may be an effective skin-whitening agent.

This paper demonstrated using lauryl alcohol as a feedstock for producing sophorolipids using Candida bombicola. Sophorolipids are biosurfactants that have antimicrobial and emulsifying properties. The study found that sophorolipid production was maximized at 96 hours of incubation, with 10% glucose concentration, initial pH of 6.0, and a temperature of 30°C. The produced sophorolipids reduced surface tension more than synthetic surfactants and had good emulsification activity and antibacterial effects against both gram-positive and gram-negative bacteria.
