HLA genotyping identifies polymorphisms in HLA class I and II genes, which is important for transplant matching and disease studies. It is technically challenging due to high polymorphism. Next generation sequencing has improved HLA genotyping by enabling robust, simple, high-quality analysis of key HLA genes at high throughput with data phased to at least 6 digits.

Thomas Lipscomb submitted an abstract titled "FUNCTION AND CLINICAL APPLICATION OF EMBRYONIC GENE NANOG IN CANCER" about the role of the embryonic gene Nanog in acute myeloid leukemia (AML). The study found that Nanog is expressed in AML cell lines and primary human AML cells. Decreasing Nanog expression in HL60 AML cells through sodium orthovanadate treatment decreased cell proliferation, as measured by trypan blue staining and real-time PCR. The results suggest Nanog may be a biomarker and therapeutic target for AML, and decreasing its expression could help treat the disease.
Different Faces of Fas Signaling in Cancer Cells-Crimson Publishers
Fas signaling pathway is known to be engaged in elimination of unwanted cells by apoptosis, however, new discoveries presented its alternate face, namely FasR and FasL play pro-cancerous functions facilitating cancer progression, invasion and metastasis. Here, we provide a brief summary of knowledge concerning Fas signaling pathway merits and disadvantages in cancer. Fas molecules were found to be engaged in many pathways induced by different therapeutic compounds, such as aspirin and antibiotics. This suggest that Fas signaling pathway may be employed as adjuvant factor for immunotherapy of variable design. Although, the literature presents some first optimistic results, we must intensify efforts to finally introduce Fasassociated procedures into standard therapeutic options. The Fas signaling is known to exert very variable effects in wide spectrum of cells. The FasR/ FasL (CD95/ CD95L) proteins can be associated with both positive (physiological) and negative (pathological) effects. Nowadays, there is a growing interest in the elucidation of the Fas signaling role in the pathogenesis and progression of various cancers. Additionally, it was proven that the expression of FasR/ FasL in colorectal cancer is associated with worse prognosis, metastasis and recurrence [1-6] the aspects of cancer biology which cancer stem cells are responsible for [7].

The study finds that Snail and Slug contribute to resistance by 1) repressing p53-mediated apoptosis and 2) promoting the acquisition of stem-like characteristics in ovarian cancer cells. This allows cancer cells to survive radiation/chemotherapy stress and take on properties associated with cancer stem cells that can regenerate tumors. This study compared genomic data from 39 HNSCC cell lines to genomic findings from 106 HNSCC tumors. Amplification of eight genes and deletion of five genes were found in both cell lines and tumors. Seventeen genes were only mutated in cell lines, suggesting these mutations arose in tissue culture. Conversely, 11 genes were only mutated in over 10% of tumors. Several mutant genes in the EGFR pathway were shared between cell lines and tumors. Pharmacologic profiling of six cell lines suggested PIK3CA mutation may predict sensitivity to EGFR/PI3K pathway drugs. These findings suggest correlating gene mutations between cell lines and tumors can guide selection of preclinical models.

This study found that acute myeloid leukemia (AML) cells driven by the fusion oncogenes AML1-ETO and PML-RARα have increased DNA damage and are more sensitive to PARP inhibitors compared to AML cells driven by MLL-AF9. Sensitivity to PARP inhibitors was correlated with lower expression of DNA damage response genes. The study also found that modulating the activity of the oncogene HOXA9, which is highly expressed in MLL-AF9 AML, could render those cells sensitive to PARP inhibitors by reducing DNA repair capacity. Combining PARP inhibitors with agents that target HOXA9 may be a promising new therapeutic strategy for AML.

This dissertation examines targeting RAS palmitoylation in BCR-ABL mediated hematological malignancies. The study demonstrates that expressing palmitoylation-deficient forms of oncogenic and wild-type NRAS significantly slows progression of BCR/ABL induced leukemia-like diseases in mice. Specifically, palmitoylation-deficient wtNRAS suppresses activation of the RAS/RAF/MEK/ERK signaling pathway. The results suggest that targeting RAS palmitoylation may constitute an effective therapy for cancers driven by hyperactivation of RAS, including those caused by BCR/ABL.

Macrophages form functional vascular mimicry channels in vivo. The document describes an experiment showing that macrophages structurally form primitive, perfused vascular networks in both tumor and angiogenesis models in vivo. The networks are formed through hypoxia-driven vascular mimicry. Time course experiments show macrophages invade matrigel plugs, align to form interconnected cords that mature into a complex 3D tubular network, expressing both macrophage and endothelial markers, prior to endothelial blood vessel formation. The identification of macrophages directly forming vascular networks through vascular mimicry may provide new targets for cancer and other vascular disease therapeutics.

The study found epithelial alterations such as atypical squamous cells in 42% of pup sea lions and 67% of adult females. Expression of a specific MHC class II gene was found to have a slightly protective effect against epithelial alterations. CNVs have been found in many cancers including colon cancer and are thought to contribute to cancer development by altering gene dosage of oncogenes and tumor suppressors. Studies have shown that CNVs present in cancer cells are often absent in normal cells from the same patient. While the role of CNVs in cancer risk is still being explored, certain CNVs are associated with hereditary cancer syndromes that increase risk of colon cancer. Further research on cancer CNVs may lead to new biomarkers for cancer susceptibility, progression, and metastasis.
Rad52 Molecular Carcinogenesis

The study found that two single nucleotide polymorphisms (SNPs) in the RAD52 gene are associated with increased lung squamous cell carcinoma (LUSC) risk and increased RAD52 expression levels. Gene expression based classification of non-small cell lung carcinomas
This study performed gene expression profiling on 91 non-small cell lung cancer tumor samples and 65 normal lung tissue samples. It identified a set of 75 histological signature genes that can classify the different subtypes of non-small cell lung cancer. It also identified 17 genes whose expression levels were correlated with patient survival time after surgery. These 17 genes could stratify patients into high-risk and low-risk groups for survival prediction.

Role of major histocompatibility complex
MHC genes evolve through duplication, followed by diversification, co‐evolution, and sequence exchange. The focus, for HLA in transplantation, has been the specific classical class I and class II human leukocyte antigen (HLA) molecules and alleles. Importantly, anti‐HLA antibodies developed after the organ transplants play a role in acute and chronic allograft rejection, highlighting the need to detect these antibodies in a clinically relevant manner. Although the immune response to HLA antigens plays a pivotal role in allograft rejection, evidence shows that non‐HLA antigens also contribute to the pathogenesis of acute and chronic rejection, which limits long‐term graft survival of the solid organ transplants.

Em consonância com os ODS – Objetivos do Desenvolvimento Sustentável e a Estratégia Global para a Saúde das Mulheres, Crianças e Adolescentes, e aplicando uma abordagem baseada nos direitos humanos, os esforços de cuidados pós-natais devem expandir-se para além da cobertura e da simples sobrevivência, de modo a incluir cuidados de qualidade.
Estas diretrizes visam melhorar a qualidade dos cuidados pós-natais essenciais e de rotina prestados às mulheres e aos recém-nascidos, com o objetivo final de melhorar a saúde e o bem-estar materno e neonatal.
Uma “experiência pós-natal positiva” é um resultado importante para todas as mulheres que dão à luz e para os seus recém-nascidos, estabelecendo as bases para a melhoria da saúde e do bem-estar a curto e longo prazo. Uma experiência pós-natal positiva é definida como aquela em que as mulheres, pessoas que gestam, os recém-nascidos, os casais, os pais, os cuidadores e as famílias recebem informação consistente, garantia e apoio de profissionais de saúde motivados; e onde um sistema de saúde flexível e com recursos reconheça as necessidades das mulheres e dos bebês e respeite o seu contexto cultural.
Estas diretrizes consolidadas apresentam algumas recomendações novas e já bem fundamentadas sobre cuidados pós-natais de rotina para mulheres e neonatos que recebem cuidados no pós-parto em unidades de saúde ou na comunidade, independentemente dos recursos disponíveis.
É fornecido um conjunto abrangente de recomendações para cuidados durante o período puerperal, com ênfase nos cuidados essenciais que todas as mulheres e recém-nascidos devem receber, e com a devida atenção à qualidade dos cuidados; isto é, a entrega e a experiência do cuidado recebido. Estas diretrizes atualizam e ampliam as recomendações da OMS de 2014 sobre cuidados pós-natais da mãe e do recém-nascido e complementam as atuais diretrizes da OMS sobre a gestão de complicações pós-natais.

Hla genotyping
- 1. HLA genotyping The HLA (human leucocyte antigen) system encodes the major histocompatibility complex (MHC) proteins in humans. These integral cell membrane glycoproteins are responsible for the regulation of the human immune system. There are two classes of MHC, i.e., MHC class I and MHC class II. The HLA gene complex is located on a 3.6 Mb region within chromosome 6p21. They are the most polymorphic gene family found in the human genome, with more than 10,000 different HLA alleles reported to date, thus the capacity to mount an immune response can be dramatically different between individuals within a cohort selected from a single population. HLA genes have been strongly implicated in transplant rejection, autoimmune disease, vaccine pharmacogenomics, cancer, infectious diseases, and mate selection. HLA genotyping is the identification of the HLA class I and class II gene polymorphisms for individuals, which is indispensable for transplant matching and disease association studies. Unambiguous HLA genotyping is technically challenging owing to high polymorphism in various genomic regions. The development of NGS has changed this landscape of genotyping. High-resolution HLA genotyping by using PCR and NGS is uniquely able to address limitations of traditional HLA genotyping and Sanger sequencing assays in patients. It enables robust, simple, high-quality, and high-throughput analysis of the key HLA genes, data can be phased to a minimum of 6 digits. Another advantage is that phasing problem is determined since DNA templates are derived from single molecules. Figure 1. The HLA region in Chromosome 6.
